-globin gene haplotypes were determined for 196 normal (-A) and 419 thalassaemia (-Th) chromosomes of individuals from four different regions of the Indian subcontinent; North-west Pakistan, Gujarat, Punjab and Sindh. Analysis of -A and -Th haplotypes and haplotype-mutation associations in each regional group along with a consideration of Indian history provided information about the origin and spread of -thalassaemia mutations on the Indian subcontinent. The data are consistent with relatively recent and local origins for most -thalassaemia mutations. The frequencies of particular alleles differ markedly in various regions and these may be useful population markers. Of the high frequency alleles, intervening sequence 1 (IVS-1) nucleotide 5 (G-C) and codons 41/42 (-CTTT) appear to be older as suggested by multiple haplotype associations and a widespread geographical distribution. The microepidemiology of -thalassaemia in this region reflects considerable ethnic diversity, gene flow from population migration and natural selection by malaria infection. 相似文献

Comparative mapping studies in human and mouse have shown that, to date, human Chromosome (Chr) 20 is completely syntenic with distal mouse Chr 2. The structural locus for S-adenosyl-l-homocysteine hydrolase (EC 3.3.1.1) in human, AHCY, maps to 20 qterq13.1, and we report here that the homologous locus in the mouse, Ahcy, maps to distal mouse Chr 2 with gene order Pcna-Ahcy-Ada. Analysis of 123 progeny of an interspecific backcross between a laboratory stock, AN, and Mus spretus using a rat cDNA probe revealed the presence of at least two other Ahcy-related sequences segregating independently in the mouse genome. One, Ahcy-rs1, was mapped to Chr 8 in the BXH recombinant inbred strains, and the other, Ahcy-rs2, shows a pattern of inheritance consistent with X-linkage. 相似文献

Larvae of the large cabbage white butterfly, Pieris brassicae L. are important pests of cruciferous crops. Third and fifth instars were treated topically with sublethal doses of the formamidine pesticide chlordimeform. A number of excitable actions were induced but they did not persist beyond 24 hours. Following the initial period, however, subsequent development was delayed and mortality increased, particularly when third instars were treated. Dosing fifth-instar larvae caused few further deaths. A link is suggested between these results and the nutritional status of the insects, and they are discussed in terms of how they might contribute towards control.
Résumé Les chenilles de troisième et cinquième stades de P. brassicae L. ont reçu une application superficielle de chlordiméforme, insecticide à la formamidine. Les doses inférieures à 335 g/g ont été sublétales, provoquant plusieurs formes d'excitation. Bien que leur comportement soit redevenu normal en 24 heures, les troisièmes stades traités ont eu leur développement retardé et une mortalité accrue. Le traitement des chenilles plus âgées a eu moins d'effets. Une relation est suggérée entre ces observations et le régime alimentaire de cet insecte; une réduction de l'alimentation, plus importante chez les chenilles de troisième que de cinquième stade de P. brassicae, accompagne le début des altérations du comportement provoquées par le chlordiméforme. To allow a more valid comparison between our previous ultrastructural data and the immunolocalization of type IX and other minor collagen species in cryosectioned cartilage, we examined both normal and testicular hyaluronidase-digested canine tibial cartilage by electron microscopy. Removal of matrix proteoglycans caused the pericellular capsule to collapse against the cell surface, suggesting that its normal anatomical position is mediated by pericellular matrix hydration. Detailed examination of the pericellular capsule and pericellular channel revealed fine, faintly banded fibrils and an amorphous component somewhat similar in structure to basement membrane collagens. Matrix vesicles and the electron-dense material of the interterritorial matrix were only partially digested by hyaluronidase. We propose that the pericellular capsule is composed of a "felt-like" network of minor collagen species which act synergistically to maintain both the composition of the pericellular matrix and the integrity of the chondrocyte/pericellular matrix complex during compressive loading. 相似文献

Thyroid ultrastructure changes were studied during the course of a low iodine diet in rats. At day 20, follicles were normal, but a number of them contained cells of higher density and with greatly elongated microvilli. Endoplasmic reticulum cisternae were frequently dilated. From day 20 until day 80, the most characteristic changes in the thyroid cells were the progressive accumulation of subapical peroxidase-positive exocytotic vesicles. After 80 days of the low iodine treatment, Golgi apparatuses were very active. Cell division could be observed. At this stage, exocytotic vesicles were generally very abundant. These data suggest that the remarkable accumulation of subapical exocytotic vesicles between day 20 and day 120 might represent an adaptation to the moderate and gradual increase in TSH stimulation that occurs in the conditions of low iodine diet. 相似文献

Abstract Hydrogen fluoride treatment of [14 C-glycerol]lipoteichoic acid synthesized by growing Streptococcus faecium ATCC9790 in the presence of 1,3[14 C]glycerol produced five radioactive, water-soluble products which were identified by chromatographic and analytical techniques to be tetraglucosyl glycerol, triglucosyl glycerol, diglucosyl glycerol, monoglucosyl glycerol and unsubstituted glycerol. The percent composition of each varied modestly from culture to culture and ranged between 7 and 8% for the tetra-, 20.5 and 31.2% for the tri-, 11.3 to 23.5% for the di-, 20.9 to 26.8% for the mono-, and 23.1 to 34.8% for the unsubstituted glycerol. The same glucosylated glycerol compounds could be obtained in an in vitro reaction in which a 30 000 × g particulate enzyme catalyzed the incorporation of [3 H]glucose from UDP [3 H]glucose into lipoteichoic acid. 相似文献

Abstract Subinhibitory concentrations of trimethoprim-sulfamethoxazole increased the total yield of Shiga-like toxin (SLT), produced by Shigella dysenteria 1 and by enterophathogenic and enterohemorrhagic strains of Escherichia coli . Stimulation of SLT synthesis by trimethoprim-sulfamethoxazole was demonstrated by an increase in cytotoxic activity for HeLa cells and the diameter of the zone formed around bacterial colonies probed with monoclonal antibodies to SLT. Thus, supplementation of culture media with trimetroprimsulfamethoxazol will facilitate SLT purification and detection of SLT-producing bacteria. 相似文献

When adult rat hepatocytes were co-cultured with another liver epithelial cell type in a medium supplemented or not with fetal calf serum (FCS), it was found that 1. They survived for more than 2 months 2. Albumin secretion levels remained high over the whole culture period 3. Decreased secretion might be reversed 4. This protein secretion activity appeared to be dependent upon both the presence of cell-cell contacts and the production of an extracellular material. The results demonstrate for the first time long-term stabilization and reversibility of a specific function (albumin secretion) at high levels by adult hepatocytes cultured in serum-free medium and suggest that both the presence of other liver cell type(s) and the production of an extracellular matrix are needed for the maintenance of specific functions in cultured hepatocytes. 相似文献

The present-day practices of electrocardiography and vectorardiography are based upon the theory that the surface potential
differences can be assumed to be due to a single dipole inside the body. It is shown in this paper that a dipole cannot account
for all the surface potentials due to realistic current generators, and hence the determination of the current generator from
surface potential measurements based upon such a theory will lead to inconsistent representations of the heart for one and
the same subject.
To demonstrate this point two eccentric dipoles of different strengths and locations representing two muscle fibers are taken
to be the current generator in a homogeneous spherical conductor. The exact surface potentials are then expressed by means
of the “interior sphere theorem” of the authors. With these expressions the magnitude, direction, and location of the resultant
dipole are determined by the method of D. Gabor and C. V. Nelson (J. App. Physics,25, 413–16, 1954). The surface potentials due to this resultant dipole are again exactly expressed by means of the “interior
sphere theorem” and compared with those due to the eccentric dipoles assumed. It can be seen that the differences can be considerable.
It is suggested that the multipole model of the authors (Bull. Math. Biophysics,20, 203–16, 1958) be used as a more accurate and the only unique representation of the heart.
