Molecularly imprinted nano-particles (MINPs) selective for olanzapine were prepared using methacrylic acid (MA) as monomer, ethylene glycol dimethacrylate (EGDMA) as a cross-linker, and 2,2-azobis (2-isobutyronitrile) (AIBN) as the initiator in 36 different ratios. The reaction runs with considerable fine powder formation were selected for further binding and selectivity studies. The MINP with the best selectivity (MINP-32) was chosen for further structural characterization by Fourier transform infrared spectroscopy (FT-IR), thermal gravimetric analysis (TGA), scanning electron microscopy (SEM), adsorption-desorption isotherm for specific surface area, volume and average pore diameter determination. All characterization methods confirmed the successful formation of MINP. The optimum conditions for maximum template loading on the MINP-32 were found by experimental design using response surface methodology (RSM) and choosing absorbent amount, pH, and time as the main factors. MINPs with maximum template loading also indicated significant selectivity between template and its analog (clozapine). The release profile demonstrated a maximum release of about 95% after 288 h for MINP-32 in comparison with about 94% after 120 h for non-MINP-32. The same slow release of drug from MINP-32 was also observed during animal study of the plasma level of template, 20–28 μg/ml versus 5–10 μg/ml. The MINP-32 of this study represents a desirable ability to keep the memory of the template with significant selectivity and good capability to control the release of template in vitro and in vivo and hence could be a promising drug delivery system.  相似文献

This study was carried out in southwestern Caspian Sea coastal area to elucidate demersal bony fishes distribution and abundance. Twenty two species were found in shore line, which belong to 6 families. In deeper waters down to 7 meters only 5 species of Gobiidae and 1 species of Syngnathidae were identified. Two species Neogobius caspius and N. pallasi were the main components of demersal fishes; however in some regions Rutilus caspicus, Rutilus kutum and Liza spp. were dominated. Most places were dominantly occupied by Atherina boyeri, which had the highest abundance among the pelagic fishes. Conversely, the Ponticola gorlap, N. melanostomus and Proterorhinus nasalis showed the lowest abundances (less than 1 ind./100 m2). Benthophilus stellatus and P. nasali had the lowest niche overlaps with other species. Both of these two species and P. gorlap are suggested to be classified as endangered or vulnerable species based on their low abundance. The distribution of Syngnathus abaster depends on algae, while algae grow up into the stony construction of the coastal line. A negative insignificant correlation was observed between pelagic and demersal fish abundances. The commercial fisheries data of 76 beach seines in our study area during 2002–2013 was compared with our results on fish abundance and species composition. More than 93% of total fish catch constituted Liza spp. and Rutilus kutum. The relationship between the abundance and distribution of the studied species, and the restocking activities performed by Iranian fisheries organization are discussed. The high abundance and distribution of small size fish in tidal zone could be related to the distribution of crustacean, which provide a good source of food for these fish species.  相似文献

A probiotic is a living micro-organism administered to promote the health of the host by treating or preventing infections owing to strains of pathogens. Saccharomyces boulardii is a nonpathogen yeast that has a direct inhibitory effect on the growth of many pathogens, an anti-secretory effect and a trophic effect on enterocytes. The aim of this study was to determine the effect of S. boulardii on diarrhea in children. The children from 6 months to 6 years of age with acute watery diarrhea admitted in pediatric clinic in Kashan in 2012 were included in this trial. Exclusion criteria were high fever (T > 38.5 °C), severe dehydration, bloody diarrhea, severe malnutrition, using of antibiotics, anti-diarrheal or antifungal drugs and children with more than one complain. Two hundred patients were assigned into two groups: A total of 100 patients were treated with S. boulardii in addition to ORS (case group) and 100 patients were given placebo in addition to ORS (control group). The duration of diarrhea and frequency of stools were recorded by asking the mothers of the children every day. The results showed that the defecation frequency after second day of treatment in the case group was significantly less than the control group (P = 0.001) and the mean numbers of days of diarrhea was significantly lower in the case group (P = 0.001). The result of this study confirms that S. boulardii reduces the frequency of stool and duration of illness in children.  相似文献

BackgroundBurkholderia pseudomallei (B. pseudomallei), the causative agent of melioidosis, is a deadly pathogen endemic across parts of tropical South East Asia and Northern Australia. B. pseudomallei can remain latent within the intracellular compartment of the host cell over prolonged periods of time, and cause persistent disease leading to treatment difficulties. Understanding the immunological mechanisms behind persistent infection can result in improved treatment strategies in clinical melioidosis.MethodsTen-day LD50 was determined for the small-colony variant (SCV) and its parental wild-type (WT) via intranasal route in experimental BALB/c mice. Persistent B. pseudomallei infection was generated by administrating sub-lethal dose of the two strains based on previously determined LD50. After two months, peripheral blood mononuclear cells (PBMCs) and plasma were obtained to investigate host immune responses against persistent B. pseudomallei infection. Lungs, livers, and spleens were harvested and bacterial loads in these organs were determined.ResultsBased on the ten-day LD50, the SCV was ~20-fold less virulent than the WT. The SCV caused higher bacterial loads in spleens compared to its WT counterparts with persistent B. pseudomallei infection. We found that the CD4+ T-cell frequencies were decreased, and the expressions of PD-1, but not CTLA-4 were significantly increased on the CD4+ and CD8+ T cells of these mice. Notably, persistent infection with the SCV led to significantly higher levels of PD-1 than the WT B. pseudomallei. Plasma IFN-γ, IL-6, and IL-17A levels were elevated only in SCV-infected mice. In addition, skewed plasma Th1 and Th17 responses were observed in SCV-infected mice relative to WT-infected and uninfected mice.ConclusionB. pseudomallei appears to upregulate the expression of PD-1 on T cells to evade host immune responses, which likely facilitates bacterial persistence in the host. SCVs cause distinct pathology and immune responses in the host as compared to WT B. pseudomallei.  相似文献

Expansion of seeded human Wharton's jelly mesenchymal stem cells (hWJ-MSCs) on 2D culture plates and 3D nano-hydroxyapatite/chitosan/gelatin scaffolds, from morphology and osteoactivity points of view, were investigated. Cell attachment and spreading, temporal expression profiles of selected osteogenic gene and protein markers, intracellular alkaline phosphatase enzyme activity (ALP activity), and matrix mineralization were assayed over the course of the experiments. Morphological results demonstrated hWJ-MSCs had greater affinity to adhere onto the 3D scaffold surface, as the number and thickness of the filopodia were higher in the 3D compared with 2D culture system. Functionally, the intracellular ALP activity and extracellular mineralization in 3D scaffolds were significantly greater, in parallel with elevation of osteogenic markers at the mRNA and protein levels at all-time point. It is concluded that 3D scaffolds, more so than 2D culture plate, promote morphology and osteogenic behavior of WJ-MSCs in vitro, a promising system for MSCs expansion without compromising their stemness before clinical transplantation.  相似文献
