Membrane fluidity of human cheek cells was determined using fluorescence recovery after photobleaching (FRAP) and steady-state fluorescence anisotropy. The FRAP data showed that the lateral diffusion coefficient (D) and mobile fraction (%R) of lipid in the plasma membrane of control cells were 2.01×10–9 cm2/ sec and 54.25%, respectively. Trypsin treatment increased D and %R to 6.4×10–9 cm2/sec and 72.15%. In contrast, the anisotropy (r) for control cells was 0.270 which remained unchanged by trypsin treatment. The results show that diffusion of lipids in the plane of the membrane is restricted by trypsin-sensitive barriers.  相似文献

Thrips are one of the insect pests of persimmon (Diospyros kaki Thunb.) in the major production areas of astringent persimmon in Korea. We surveyed astringent persimmon orchards in the Damyang, Sangju and Cheongdo regions of Korea to determine thrips species composition and abundance. Orchards sprayed with either organic or conventional pesticides were sampled over the course of one flowering season, using yellow sticky traps to determine if this is a suitable method for monitoring thrips populations, and to determine thrips species composition and abundance. Eight thrips species were captured on yellow sticky traps in both the tree canopy and ground cover: Ponticulothrips diospyrosi Haga et Okajima, Scirtothrips dorsalis (Hood), Frankliniella occidentalis (Pergande), F. intonsa (Trybom), Thrips tabaci (Lindeman), T. hawaiiensis (Morgan), T. coloratus (Schmutz) and T. palmi (Karny). In all regions, F. occidentalis and F. intonsa dominated in both organic and conventional orchards. S. dorsalis, F. occidentalis, F. intonsa and T. hawaiiensis were found in persimmon flowers, with S. dorsalis the dominant thrips. Significantly more S. dorsalis were captured from flowers in the lower and middle canopy than in flowers from the upper canopy. Fruit damage was also significantly higher in fruit from the lower canopy than in fruit from the middle and upper canopy.  相似文献

The survival of indigenous and introduced strains of Azorhizobium caulinodans in flooded soil and in the rice rhizosphere, where in situ Sesbania rostrata was incorporated before the rice crop, is reported. The azorhizobia studied were both root and stem nodulating. In a pot experiment, two crop cycles each of inoculated and noninoculated Sesbania-rice were compared with two crop cycles of flooded fallow-rice. In a field experiment, the effect of repeated incorporation of in situ S. rostrata in the Sesbania-rice sequence was studied. Soils in which inoculated S. rostrata was incorporated contained about 3,000 times more azorhizobia than did soils in the flooded fallow treatment and about 50 times more azorhizobia than did soils in the noninoculated Sesbania treatment. Azorhizobial numbers in the inoculated Sesbania treatment declined toward rice harvest but remained much higher than in the flooded fallow-rice treatment. Repeated incorporation of S. rostrata increased the population density of indigenous soil azorhizobia, whereas the population of inoculated strain ORS571 (Strr Spcr) declined to an undetectable level; this finding suggested low competitiveness by the introduced strain. In the incorporated Sesbania treatment, the rice rhizosphere harbored significantly more A. caulinodans and supported higher nitrogenase activity per plant than did the rhizosphere of the flooded fallow-rice treatment. Sterile rice seedlings inoculated with A. caulinodans showed nitrogenase activity comparable to that of seedlings inoculated with Azospirillum lipoferum 34H, a rice root isolate. Rhizobia from Sesbania aculeata, Sesbania sesban, a Trifolium sp., and Vigna unguiculata did not support appreciable nitrogenase activity.  相似文献

Summary A short-term laboratory acetylene reduction assay using cut plant-soil samples incubated in the dark was developed for measuring relative N2-fixing activities associated with field-grown rice plants. The assay sample consists of rhizosphere soil, root, and cut stem and leaf sheath. The cut plant-soil assay is relatively simple, rapid, and convenient; it reduces, if not eliminates, the problems encountered in whole-plant (field, pot, and water culture) and excised roots assays. Varietal differences in N2-fixing activity were detected with the new assay technique.  相似文献

Seedlings of rice (Oryza sativa L. var. IR42) were inoculated with nitrogen-fixingAzospirillum lipoferum (strain 34H) by immersing the roots in the inoculum for 6 h. The plants were grown in the prescence of NH4 +-N for 47 days in a hydroponic system under greenhouse conditions. Inoculation significantly enhanced PO4-ion uptake of the plants in 4 of the 7 samplings tested while the uptake of NH4-ion was significantly increased in two samplings and was decreased in one sampling. Inoculation reduced root length significantly and caused significant increases in shoot fresh and dry weights. Root surface area was not affected by inoculation. Bacterial population counts suggested thatA. lipoferum survived on the roots till the end of the experiment.  相似文献

The pituitary pars intermedia of Camelus dromedarius is well developed and completely surrounds the pars nervosa. Two major groups of cells are present: endocrine (ec) and glial-like cells (glc). The ec group is composed of three morphologically distinct cell types. Type I, or polyhedral light cells (LC-I) and type II, or polyhedral dark cells (DC-II), have secretory granules of heterogeneous electron density whose size ranges from 170 to 300 nm. Type III cells are elongated with homogeneous electron-dense secretory granules of 80–200 nm. The glc make up an organized network, form follicles in the centrolobular zones and are positive for vimentin and S-100β immunolabelling. The nerve fibres penetrating the lobe are numerous, and can be classified into two types according to the membrane bound vesicles found in their endings (ne). Ultrastructural quantitative analysis revealed significant variations in PI elements between winter and summer seasons (F = 8.014, p = 0.006). DC-II cells characterized by developed biosynthetic machinery and a large pool of secretory granules storage are increased with the ne in winter. However, LC-I cells showing frequent cytoplasmic degranulation are predominant with glc in summer. Thus, important cellular remodelling occurs in the dromedary PI that may depend upon, or perhaps anticipate, external living conditions.  相似文献

Anthrax, caused by the bacterium Bacillus anthracis, is a zoonotic disease that persists throughout much of the world in livestock, wildlife, and secondarily infects humans. This is true across much of Central Asia, and particularly the Steppe region, including Kazakhstan. This study employed the Genetic Algorithm for Rule-set Prediction (GARP) to model the current and future geographic distribution of Bacillus anthracis in Kazakhstan based on the A2 and B2 IPCC SRES climate change scenarios using a 5-variable data set at 55 km2 and 8 km2 and a 6-variable BioClim data set at 8 km2. Future models suggest large areas predicted under current conditions may be reduced by 2050 with the A2 model predicting ∼14–16% loss across the three spatial resolutions. There was greater variability in the B2 models across scenarios predicting ∼15% loss at 55 km2, ∼34% loss at 8 km2, and ∼30% loss with the BioClim variables. Only very small areas of habitat expansion into new areas were predicted by either A2 or B2 in any models. Greater areas of habitat loss are predicted in the southern regions of Kazakhstan by A2 and B2 models, while moderate habitat loss is also predicted in the northern regions by either B2 model at 8 km2. Anthrax disease control relies mainly on livestock vaccination and proper carcass disposal, both of which require adequate surveillance. In many situations, including that of Kazakhstan, vaccine resources are limited, and understanding the geographic distribution of the organism, in tandem with current data on livestock population dynamics, can aid in properly allocating doses. While speculative, contemplating future changes in livestock distributions and B. anthracis spore promoting environments can be useful for establishing future surveillance priorities. This study may also have broader applications to global public health surveillance relating to other diseases in addition to B. anthracis.  相似文献

Female ixodid ticks are amazing invertebrate animals which efficiently convert a large amount of nutrients derived from their ingested blood meals into eggs. Although oocyte development (vitellogenesis) in ticks is triggered by a blood meal and is assumed to be supported by nutrition derived from ovarian cells connecting oocytes, little is known about the ovarian molecules processing nutrient materials for the vitellogenesis. In this study, we have suggested a putative function of leucine aminopeptidase (HlLAP) in the ovary of parthenogenetic adult ixodid tick Haemaphysalis longicornis regarding a negative output of reproduction following disruption of HlLAP gene by RNA interference. Endogenous HlLAP was shown to be localized in the ovarian cells, including ovarian epithelial and pedicel cells which were assumed to provide nutrients for the developing oocytes. Histological studies demonstrated that a majority of immature oocytes in HlLAP gene knockdown ticks were transformed into abnormal morpho-histological oocytes with vacuolated cytoplasm and/or condensed nucleus. Taken together, a reduction of the numbers of laid eggs in the HlLAP gene knockdown ticks may be due to the degeneration of immature oocytes following deprivation of nutrients such as amino acids supplied not only by midgut HlLAP but also by the ovarian HlLAP. Regulation of the tick molecules involved in nutrient metabolism for the reproduction, including blood digestion and vitellogenesis, would help in controlling the tick population and tick-borne pathogens.  相似文献

Some properties of the K141E mutant of human HSP22 that is expressed in distal hereditary motor neuropathy were investigated. This mutation slightly decreased intrinsic fluorescence of HSP22 and induced changes in the far UV CD spectra that correlate with increase of disordered structure. Destabilized K141E mutant was more susceptible to trypsinolysis than the wild type protein. Mutation K141E did not significantly affect the hydrophobic properties measured by bis-ANS binding and did not affect the quaternary structure of HSP22. With insulin as a substrate the chaperone-like activity of K141E mutant and the wild type protein were similar. However with alcohol dehydrogenase and rhodanese the chaperone-like activity of K141E mutant was remarkably lower than the corresponding activity of the wild type protein. It is concluded that K141E mutation induces destabilization of HSP22 structure and probably by this means diminish the chaperone-like activity of HSP22 with certain protein substrates.  相似文献
