The aims of this study were (i) to determine the genetic variability of the striped field mouse (Apodemus agrarius Pallas, 1771) population in the area of the Warsaw conurbation and the genetic differentiation among local populations of this species within the city, and (ii) to examine the influence of the degree of isolation on the appearance of such genetic structure. Comparison of the genetic structure of the urban population with that of populations inhabiting rural areas was intended to shed light on the depth of genetic changes in this species caused by life in an urban environment. Eight microsatellite loci were used for genotyping and these molecular analyses were successfully performed for 269 individuals. Genetic differentiation (FST) was much higher in the case of the urban population. STRUCTURE analysis indicated that this population was subdivided into several local populations, whereas for the rural locations, the highest likelihood was for a single genetic group (one genetically unstructured population). Moreover, in contrast to the rural population, we found no isolation by distance in the urban population. Despite clear subdivision of the urban population, the level of genetic variability was very similar in both analyzed groups. Our findings indicate the occurrence of an advanced process of differentiation of the urban population by microevolution, whereas populations from rural locations displayed very small and mainly insignificant genetic differentiation. Urban local populations, situated near the city borders or close to the natural ecological corridor (banks of the Vistula River), showed higher genetic variability and were less differentiated from populations inhabiting rural sites than from local populations in the city centre. These results provide support for the importance of ecological corridors in preserving the genetic variability of the urban striped field mouse population.  相似文献

The results of employing (Ti, Fe)‐alloyed Si thin‐film anode for Li‐ion batteries are reported. The material demonstrates an impressive cyclic stability with stable operation for more than 500 cycles at a capacity higher than 1400 mAh g?1. Materials characterization using scanning electron microscopy and transmission electron microscopy illuminates an intriguing materials process behind the performance: ripple‐like pattern formation via electrochemically driven segregation of the inactive elements (Ti and Fe). The ripple structure plays a buffer role by suppressing loss of the active material upon further cycling, allowing the anode to gradually transform into an array of microbumps. The morphological evolution helps the anode endure long cycles (even up to 1000 cycles) without catastrophic failure as the bumps shrank slowly and steadily, consistent with the electrochemical data.  相似文献

The effect of exogenously applied natural [indole-3-acetic acid (IAA), phenylacetic acid (PAA), indole-3-butyric acid (IBA)] and synthetic [1-naphthaleneacetic acid (NAA)] auxins on the growth and metabolism of green microalga Chlorella vulgaris was examined. Exogenous auxins acted in a concentration-dependent manner on algal growth. Phytohormones at concentration of 100 μM inhibited algal growth expressed as the number of cells. IAA and IBA displayed the highest biological activity at 0.1 μM, whereas PAA and NAA were characterized by the greatest stimulatory effect on the number of cells at 1 μM. Treatment with IAA and IBA at 0.1 μM or NAA and PAA at 1 μM increased the concentration of photosynthetic pigments, monosaccharides and soluble proteins in C. vulgaris. Moreover, all auxins stimulated enzymatic (ascorbate peroxidase, catalase, superoxide dismutase) and non-enzymatic antioxidant (ascorbate, glutathione) systems in C. vulgaris, and therefore, suppressed lipid peroxidation and hydrogen peroxide accumulation. The data supports the hypothesis that auxins play a central role in the regulation of C. vulgaris growth and metabolism and the components of cellular redox systems that are thought to have a prominent role in the regulation of auxin-dependent processes.  相似文献

We performed cytogenetic analysis on triploid Cobitis hybrid specimens from a natural diploid-polyploid population in the Bug River (Poland). Chromosomes were examined by fluorescence in situ hybridization method (FISH) with 28S rDNA as a probe. The rDNA sites were commonly identified in the terminal position of p arms of two small submetacentric chromosomes, in the centromere area of two acrocentric chromosomes and in the terminal position of q arm of one acrocentric chromosome. Some of the observed signals were stronger than others. In few cases, one small acrocentric chromosome showing rDNA sites located terminally on both p and a q arm was observed. This study brings useful information about polymorphism of 28S rDNA sites regarding their number, location and size in triploid specimens. Moreover, it gives preliminary data for comparison with similar researches on the parental species of Cobitis hybrid females, viz. C. taenia, C. taurica and C. tanaitica, and other related Cobitis taxa of different ploidy level. The present study increases our knowledge about ribosomal genes inheritance between Cobitis species and their polyploid hybrids.  相似文献

Modulation of protein–protein interactions involved in the immune system by using small molecular mimics of the contact interfaces may lead to the blockage of the autoimmune response and the development of drugs for immunotherapy. The nonpolymorphic β‐regions, exposed to the microenvironment, of the modeled HLA‐DQ7, which is genetically linked to autoimmune diseases, were determined. Peptides 132–141 and 58–67, located at the β1 and β2 domains of HLA‐DQ7, respectively, were tested for their involvement in the interactions with CD4+ T lymphocytes. Linear, cyclic, and dimeric analogs that mimic the exposed surfaces of HLA‐DQ7 were designed and synthesized. Their immunosuppressory activities, found in the secondary, humoral immune response to sheep erythrocytes (SRBC) in mice in vitro, ranged from 11% to 53%. The significance of the total charge of the peptides, the pattern of the hydrogen bonding, and the presence of secondary structure were investigated in relation to the immunomodulatory effect of the peptides. Two dimeric analogs of the HLA‐DQ7 58–67 fragment, consisting of the two monomers covalently linked by a polyethylene glycol (PEG) spacer, able to mimic the superdimers, were also synthesized and studied. As the 58–67 segment is located at the β1 region of HLA‐DQ7, close to the major histocompatibility complex (MHC) groove, one may assume that the 58–67 peptide could accommodate the association between T‐cell receptor (TCR) and human leukocyte antigen (HLA) by activating a co‐stimulatory molecule of the TCR/HLA interaction. This hypothesis is supported by the confocal laser image of the fluorescein‐labeled 58–67 peptide and by the fact that it is an immunostimulator at low concentration. The trace elements studied in this work (Se, Cu, Zn) are the essential constituents or cofactors required to activate numerous enzymes and proteins, playing crucial role in various physiological processes. The disturbed levels of abovementioned elements may adversely affect the endocrine system, resulting in various thyroid disorders among other upsets. The aim of this study was to investigate possible associations between them and parameters of redox balance, thyroid function indices as well as clinical records (duration of disease and therapy, lag time between thyroid surgery and this study examination, LT4 dosage) in patients with different thyroid disorders, including malignant diseases of the gland. In the group of patients with papillary carcinoma, we found a statistically significant higher Cu concentration compared with controls and patients with Hashimoto disease. In the same groups, the parameter of Zn/Cu ratio demonstrated reciprocally arranged statistically significant differences. For the group of papillary cancer patients, there was a negative correlation between lag time since thyroid operation and GPX3 activity. Our data support hypothesis of indirect involvement of Zn and Cu in thyroid regulation. For selenium, lack of simple correlation between its serum level and thyroid indices implies the need for further research on other selenium status parameters more adequately depicting changes in endocrine system.  相似文献

The degradation of diesel fuel, B20 blend and biodiesel in liquid cultures by a seven-member bacterial consortium was compared under conditions with full aeration or with limited aeration with nitrate added as main electron acceptor. Community dynamics was assessed employing real-time PCR and the ddCt method for relative quantification. Biodegradation rates increased with increasing biodiesel content, but were significantly reduced under conditions with nitrate. Despite large variations in biodegradation rates, magnitude changes in population numbers were typically observed only from zero to one order, regardless the type of fuel and electron acceptor. Only Comamonadaceae and Variovorax sp. distinctly preferred aerobic conditions, and during aerobic growth showed suppression as fuel contained more biodiesel. Thus, the consortium is relatively stable and most of the degraders can shift their metabolism from hydrocarbons to biodiesel. The stability of the consortium is of interest in the context of biodiesel-mediated biodegradation of petroleum hydrocarbons.  相似文献

The objective of this study was to examine relations between body iron (Fe) status and cadmium (Cd) accumulation in a small rodent, the bank vole, caught from the wild population in late autumn (November) and early spring (March). The concentrations of Fe in the liver, kidneys, and duodenum in the bank voles from the spring were only 30%, 60%, and 70%, respectively, of those found in the animals from the autumn. An analysis of hematocrit and hemoglobin content of blood showed no significant effect of the season, suggesting that the animals from the spring were not anemic. The exposure to dietary Cd (10 μg/g) for 7 days resulted in 70% higher accumulation of Cd in the liver and kidneys of the spring than autumn bank voles, and the concentration of Cd in the duodenum was 3.5 times higher in the spring animals, despite the fact that relative Cd intake was significantly higher in the autumn bank voles. The data indicate that seasonal changes of body Fe status occurring in the wild bank voles may influence tissue accumulation of Cd.  相似文献
