Gene expression profiles can be regarded as sums of simpler modes, analogous to the modes of a vibrating violin string. Decomposition of temporal gene expression profiles into modes by singular value decomposition (SVD) was reported before, but the question as to what degree the SVD modes can be interpreted in terms of biology remains open. We report and compare the results of SVD of published datasets from hippocampal development, neuronal differentiation in vitro, and a control time-series hippocampal dataset. We demonstrate that the first SVD mode reflects the magnitude of expression, interpretable on the Affymetrix platform. In the datasets from gene profiling of hippocampal development and neuronal differentiation, the second mode reflects a monotonous change in expression, either up- or down-regulation, in the time course of experiment. We demonstrate that the top two SVD modes are conserved between datasets and therefore, likely reflect properties of the underlying system (gene expression in hippocampus) rather than of a particular experiment or dataset. Our results also indicate that the magnitude of expression, and the direction of change in expression during hippocampal development, are uncorrelated, suggesting that they are regulated by largely independent mechanisms.  相似文献

This study presents the results of an analysis of combustion waste, namely fly ashes produced from the burning of solid fuels mixed with municipal waste. Heavy metal concentrations in the fly ashes examined fell within the following ranges (mg/kg): Cd 0.39–0.79, Cr 13.48–58.51, Ni 17.01–49.37 and Pb 14.95–59.74. As this type of waste does not exceed the limit values of pollutants for organic and organic-mineral fertilisers, it is often discarded on agricultural land or dumped in illegal landfills. Fine particles of fly ashes travel considerable distances and pose a risk to human health as they stick to people's hands during field works. The mean hazard quotient (HQ) values for non-carcinogenic forms of the elements analysed were 1.06E+00 for children and 1.47E-01 for adults. A HQ>1 indicates the probability of adverse health effects. The study has shown that the dominant exposure pathway was ingestion, which accounted for approx. 91% of total health risk, while the respective percentages for other pathways were less than 9% for dermal contact and 0.05% for inhalation. The highest health risk in the ingestion exposure pathway was observed for Cr (HQing = 4.50E-01 for children, HQing = 5.23E-02 for adults) and Pb (HQing = 4.31E-01 for children, HQing = 5.01E-02 for adults). Health risk associated with the presence of Ni and Cd in the material analysed was markedly lower. For carcinogenic forms, the Risk index was established at 6.13E-08 for children and 2.13E-07 for adults. As the exposure limit (10E-5) was not exceeded, the conditions in the area studied should be considered satisfactory.  相似文献

Sexual dimorphism in anatomical traits has been widely studied in animals. Although pelvis dimorphism was mostly studied in humans, it occurs also in many other mammalian species. Here, we investigated sexual dimorphism in the pelvis of the bank vole Myodes glareolus using individuals with known sex and reproductive status of females (parous vs nulliparous). The analyses revealed that the size and shape of pelvis differed significantly between sexes, as well as between nulliparous and parous females. In comparison with males, females had a significantly longer pelvis and pubis bones and a longer obturator foramen length, but a smaller pubis width. Interestingly, the difference between parous and nulliparous females resembles that between females and males: parous females had bigger pelvis, which probably resulted from changes during pregnancy and after birth. Left bones were on average larger than right ones, but the magnitude of directional asymmetry was not different between sex and reproduction group. Moreover, we noticed that fluctuating asymmetry of pelvis and pubis length was higher in females than in males and higher in parous than in nulliparous females, what is presumably associated with locomotor performance. A discriminant function analysis performed for the four bone size traits showed that the traits can be effectively used for a nearly perfect recognition of sexes and also a quite reliable recognition of the reproductive status of females.  相似文献

We have designed a useful method of assessing reactive oxygen species generation in biological fluids. The novel assay utilizes tyrosine phosphatase CD45 as a biosensor of oxidative stress. Applying this new method, we examined oxygen species generation in the following cell culture media: RPMI 1640, DMEM, DMEM enriched with pyruvate and MEM. We discovered that the media (especially RPMI 1640) significantly reduced the activity of protein tyrosine phosphatase. The media-caused inactivation of CD45 was reversible after treatment with dithiothreitol being a powerful reducing agent. Interestingly, the media supplemented with catalase did not exhibit any inhibitory effect on CD45 activity which suggests a hydrogen peroxide-mediated mechanism of the enzyme inactivation. In addition to that, we assessed the impact of oxidative stress level on the activity of CD45 as measured in Jurkat cells cultured in RPMI 1640 either exposed or not exposed to the light of laminar flow cabinet fluorescent lamp. We found that Jurkat cells that were exposed to light displayed ca. 20% lower activity of CD45 than the cells protected against the light. The obtained results indicate that production of hydrogen peroxide in the medium leading to inhibition of CD45 was light-dependent, and that careful protection of cell culture media from the light may help to prevent the artifact in cell studies. Hydrogen peroxide, responsible for CD45 inactivation, can be generated in cell culture media after exposition to light due to photoreactive amino acids present in the media.  相似文献

The aim of this study was to determine the influence of age and ingested food (colostrum and mature milk) on the concentrations of selected blood biochemical components connected with nitrogen and mineral metabolism in dairy calves during their first week of life. The experiment was carried out on 13 Polish Black and White breed dairy calves. The animals were fed colostrum within the first 3 days of postnatal life and thereafter the mature milk of their dams until the end ofthe experiment (7 days). The obtained results showed that intensive catabolic and anabolic changes in nitrogen occur in the first week of life. These changes were particularly intense during the first 24-48 hours of life and may reflect dynamic tissue remodelling. The results of this experiment also show that healthy calves efficiently regulate water and electrolyte homeostasis.  相似文献

Background

PCNA (proliferating cell nuclear antigen) has been found in the nuclei of yeast, plant and animal cells that undergo cell division, suggesting a function in cell cycle regulation and/or DNA replication. It subsequently became clear that PCNA also played a role in other processes involving the cell genome.

Scope

This review discusses eukaryotic PCNA, with an emphasis on plant PCNA, in terms of the protein structure and its biochemical properties as well as gene structure, organization, expression and function. PCNA exerts a tripartite function by operating as (1) a sliding clamp during DNA synthesis, (2) a polymerase switch factor and (3) a recruitment factor. Most of its functions are mediated by its interactions with various proteins involved in DNA synthesis, repair and recombination as well as in regulation of the cell cycle and chromatid cohesion. Moreover, post-translational modifications of PCNA play a key role in regulation of its functions. Finally, a phylogenetic comparison of PCNA genes suggests that the multi-functionality observed in most species is a product of evolution.

Conclusions

Most plant PCNAs exhibit features similar to those found for PCNAs of other eukaryotes. Similarities include: (1) a trimeric ring structure of the PCNA sliding clamp, (2) the involvement of PCNA in DNA replication and repair, (3) the ability to stimulate the activity of DNA polymerase δ and (4) the ability to interact with p21, a regulator of the cell cycle. However, many plant genomes seem to contain the second, probably functional, copy of the PCNA gene, in contrast to PCNA pseudogenes that are found in mammalian genomes.  相似文献

We report large induction (>65fold increases) of volatile organic compounds (VOCs) emitted from a single leaf of the invasive weed mossy sorrel, Rumex confertus Willd. (Polygonaceae), by herbivory of the dock leaf beetle, Gastrophysa polygoni L. (Coleoptera: Chrysomelidae). The R. confertus VOC blend induced by G. polygoni herbivory included two green leaf volatiles ((Z)-3-hexenal, (Z)-3-hexen-1-yl acetate) and three terpenes (linalool, ß-caryophyllene, (E)-ß-farnesene). Uninjured leaves produced small constitutive amounts of the GLVs and barely detectable amounts of the terpenes. A Y-tube olfactometer bioassay revealed that both sexes of adult G. polygoni were attracted to (Z)-3-hexenal and (Z)-3-hexen-1-yl acetate at a concentration of 300 ng h−1. No significant G. polygoni attraction or repellence was detected for any VOC at other concentrations (60 and 1500 ng h−1). Yet, G. polygoni males and females were significantly repelled by (or avoided) at the highest test concentration (7500 ng h−1) of both GLVs and (E)-ß-farnesene. Mated male and female G. polygoni might be attracted to injured R. confertus leaves, but might avoid R. confertus when VOC concentrations (especially the terpene (E)-ß-farnesene) suggest high overall plant injury from conspecifics, G. viridula, or high infestations of other herbivores that release (E)-ß-farnesene (e.g., aphids). Tests in the future will need to examine G. polygoni responses to VOCs emitted directly from uninjured (constitutive) and injured (induced) R. confertus, and examine whether R. confertus VOC induction concentrations increase with greater tissue removal on a single leaf and/or the number of leaves with feeding injury.  相似文献
