Polarized cell growth requires the establishment of an axis of growth along which secretion can be targeted to a specific site on the cell cortex. How polarity establishment and secretion are choreographed is not fully understood, though Rho GTPase- and Rab GTPase-mediated signaling is required. Superimposed on this regulation are the functions of specific lipids and their cognate binding proteins. In a screen for Saccharomyces cerevisiae genes that interact with Rho family CDC42 to promote polarity establishment, we identified KES1/OSH4, which encodes a homologue of mammalian oxysterol-binding protein (OSBP). Other yeast OSH genes (OSBP homologues) had comparable genetic interactions with CDC42, implicating OSH genes in the regulation of CDC42-dependent polarity establishment. We found that the OSH gene family (OSH1-OSH7) promotes cell polarization by maintaining the proper localization of septins, the Rho GTPases Cdc42p and Rho1p, and the Rab GTPase Sec4p. Disruption of all OSH gene function caused specific defects in polarized exocytosis, indicating that the Osh proteins are collectively required for a secretory pathway implicated in the maintenance of polarized growth.  相似文献

Abstract:  Studies were conducted under laboratory conditions to document the potential of Beauveria bassiana conidia applications for controlling the white pine weevil, Pissodes strobi (Peck). A screening test including six B. bassiana isolates allowed us to demonstrate that CFL (Centre de Foresterie des Laurentides) and IP-CPB (Île Perrault-charançon du pin blanc) were the most virulent isolates among the ones tested, with percentages of mortality after 3 weeks of 73% and 85.5%, respectively. These two B. basiana isolates were applied either onto soil or branch sections to compare the effectiveness of these potential control strategies. Greater than 75% mortality was observed within 3 weeks for both modes of application using suspensions of B. bassiana at a concentration of 1.0 × 108 (soil application) and 1.0 × 109 conidia/ml (branch application). The results demonstrate for the first time that B. bassiana is an effective entomopathogen against P. strobi .  相似文献

Like many phenotypic traits, biomechanical systems are defined by both an underlying morphology and an emergent functional property. The relationship between these levels may have a profound impact on how selection for functional performance is translated into morphological evolution. In particular, complex mechanical systems are likely to be highly redundant, because many alternative morphologies yield equivalent functions. We suggest that this redundancy weakens the relationship between morphological and functional diversity, and we illustrate this effect using an evolutionary model of the four-bar lever system of labrid fishes. Our results demonstrate that, when traits are complex, the morphological diversity of a clade may only weakly predict its mechanical diversity. Furthermore, parallel or convergent selection on function does not necessarily produce convergence in morphology. Empirical observations suggest that this weak form-function relationship has contributed to the morphological diversity of labrid fishes, as functionally equivalent species may nevertheless possess morphologically distinct jaws. We suggest that partial decoupling of morphology and mechanics due to redundancy is a major factor in morphological diversification.  相似文献

A common problem in molecular phylogenetics is choosing a model of DNA substitution that does a good job of explaining the DNA sequence alignment without introducing superfluous parameters. A number of methods have been used to choose among a small set of candidate substitution models, such as the likelihood ratio test, the Akaike Information Criterion (AIC), the Bayesian Information Criterion (BIC), and Bayes factors. Current implementations of any of these criteria suffer from the limitation that only a small set of models are examined, or that the test does not allow easy comparison of non-nested models. In this article, we expand the pool of candidate substitution models to include all possible time-reversible models. This set includes seven models that have already been described. We show how Bayes factors can be calculated for these models using reversible jump Markov chain Monte Carlo, and apply the method to 16 DNA sequence alignments. For each data set, we compare the model with the best Bayes factor to the best models chosen using AIC and BIC. We find that the best model under any of these criteria is not necessarily the most complicated one; models with an intermediate number of substitution types typically do best. Moreover, almost all of the models that are chosen as best do not constrain a transition rate to be the same as a transversion rate, suggesting that it is the transition/transversion rate bias that plays the largest role in determining which models are selected. Importantly, the reversible jump Markov chain Monte Carlo algorithm described here allows estimation of phylogeny (and other phylogenetic model parameters) to be performed while accounting for uncertainty in the model of DNA substitution.  相似文献

The spatial and vertical structure of the water currents and its relationship with the tidal cycles were studied using current meters in the Gulf of Nicoya. In the upper gulf, the vertical marine current differences increase as the depth increases. The water column at the station near Chira Island (upper gulf) shows the smaller changes in currents and in temperature. The flow at the station between San Lucas Island and Puntarenas (middle gulf) is the most stratified in this region. Currents with magnitudes over 100 cm/seg were measured during spring tides. Changes in the lags of the surface and bottom tidal flows were measured on the order of 100 minutes. In general, in this upper region the flows are toward the head of the gulf when the tide is flooding and toward the mouth when the tide is ebbing. In the lower gulf the circulation is more complex. Along an axis between Tárcoles and Negritos Islands, changes of velocity vectors are identified between surface and bottom. The current rotates in a different way in the water column in this region and their patterns cannot be explained only by the tidal cycles predicted for Puntarenas. These results demonstrate that the spatial and vertical variation of the marine currents of the Gulf of Nicoya is not only related to the thermohaline structure, but also to the tidal cycles and tide ranges that take place in this estuary.  相似文献

Using an energy dispersive X-Ray fluorescence analysis, simultaneous evaluation of K, Ca, Ti, Cr, Mn, Ge, Ni, Cu, Zn, Br, Rb, Sr and Pb in 74 marine sediment samples from the Costa Rica intertidal zones was conducted. Samples were collected between June 1999 and December 2001, from Caribbean and Pacific beaches of Costa Rica. Calcium and iron showed the highest abundances and are indicators of the natural origin of the sediments. Calcium is associated with biogenic processes such as coral reefs near the sampling sites and iron indicates a terrigenous origin. In general, the beaches of the Caribbean and North Pacific regions showed the greatest concentration of calcium. This is indicative of the abundant reef structures near these beaches. The beaches of the Central and South Pacific show the greatest iron concentrations, indicating an important lithosphere contribution and/or little contribution of calcium carbonate due to the poor development of coralline structures near the sampling sites. Finally, the analyses did not show evidence of elements associated with anthropogenic pollution. Only a northern section of Puerto Viejo beach showed high concentrations of lead, zinc and titanium, perhaps associated with hydrothermal sources.  相似文献

Neural stem cells are defined as clonogenic cells with self-renewal capacity and the ability to generate all neural lineages (multipotentiality). Cells with these characteristics have been isolated from the embryonic and adult central nervous system. Under specific conditions, these cells can differentiate into neurons, glia, and non-neural cell types, or proliferate in long-term cultures as cell clusters termed "neurospheres". These cultures represent a useful model for neurodevelopmental studies and a potential cell source for cell replacement therapy. Because no specific markers are available to unequivocally identify neural stem cells, their functional characteristics (self-renewal and multipotentiality) provide the main features for their identification. Here, we review the experimental and ultrastructural studies aimed at identifying the morphological characteristics and the antigenic markers of neural stem cells for their in vitro and in vivo identification.  相似文献

The Labridae is one of the most structurally and functionally diversified fish families on coral and rocky reefs around the world, providing a compelling system for examination of evolutionary patterns of functional change. Labrid fishes have evolved a diverse array of skull forms for feeding on prey ranging from molluscs, crustaceans, plankton, detritus, algae, coral and other fishes. The species richness and diversity of feeding ecology in the Labridae make this group a marine analogue to the cichlid fishes. Despite the importance of labrids to coastal reef ecology, we lack evolutionary analysis of feeding biomechanics among labrids. Here, we combine a molecular phylogeny of the Labridae with the biomechanics of skull function to reveal a broad pattern of repeated convergence in labrid feeding systems. Mechanically fast jaw systems have evolved independently at least 14 times from ancestors with forceful jaws. A repeated phylogenetic pattern of functional divergence in local regions of the labrid tree produces an emergent family-wide pattern of global convergence in jaw function. Divergence of close relatives, convergence among higher clades and several unusual 'breakthroughs' in skull function characterize the evolution of functional complexity in one of the most diverse groups of reef fishes.  相似文献

A statistical analysis is presented of patterns of variation in some physical, chemical, and biological variables for a 6 year series of data from the tropical, high altitude Lake Titicaca (Peru-Bolivia). ANOVA techniques and autocorrelation analyses were used to partition the variance in Titicaca, and in some comparison tropical and temperate series, into components with repeatable annual cycles and components attributable to other kinds of patterns.In Titicaca, insolation and stratification are highly seasonal in pattern of variation, although the amount of variance relative to means is small compared to temperate lakes. However, the seasonal pattern of physical variation is only weakly imposed on chemical and biological processes, to judge from analyses of silicate, oxygen, and primary production series. Comparable temperate series of primary production and chlorophyll a are much more seasonal.  相似文献

Cell polarization is essential throughout development for proliferation, migration, and differentiation. However, it is not known how extracellular cues correctly orient cell polarity at distinct stages of development. Here, we show that the endocytic adaptor protein Numb, previously characterized for its role in cell proliferation, subsequently plays an important role in cell migration. In neural precursors stimulated with the chemotactic factor BDNF, Numb binds to activated TrkB, the BDNF receptor, and functions both as an endocytic regulator for TrkB and as a scaffold for aPKC (aPKC). Thus, Numb promotes BDNF-dependent aPKC activation. Interestingly, Numb is also a substrate of aPKC. When phosphorylated, Numb exhibits increased efficacy in binding TrkB and in promoting a chemotactic response to BDNF. Therefore, Numb functions in a feed-forward loop to promote chemotaxis of neural precursors, linking BDNF, an extracellular cue, to aPKC, a critical component of the intrinsic polarity machinery.  相似文献
