Background

Multi-walled carbon nanotubes (MWCNTs) represent a human health risk as mice exposed by inhalation display pulmonary fibrosis. Production of IL-1β via inflammasome activation is a mechanism of MWCNT-induced acute inflammation and has been implicated in chronic fibrogenesis. Mice sensitized to allergens have elevated T-helper 2 (Th2) cytokines, IL-4 and IL-13, and are susceptible to MWCNT-induced airway fibrosis. We postulated that Th2 cytokines would modulate MWCNT-induced inflammasome activation and IL-1β release in vitro and in vivo during allergic inflammation.

Methods

THP-1 macrophages were primed with LPS, exposed to MWCNTs and/or IL-4 or IL-13 for 24 hours, and analyzed for indicators of inflammasome activation. C57BL6 mice were sensitized to house dust mite (HDM) allergen and MWCNTs were delivered to the lungs by oropharyngeal aspiration. Mice were euthanized 1 or 21 days post-MWCNT exposure and evaluated for lung inflammasome components and allergic inflammatory responses.

Results

Priming of THP-1 macrophages with LPS increased pro-IL-1β and subsequent exposure to MWCNTs induced IL-1β secretion. IL-4 or IL-13 decreased MWCNT-induced IL-1β secretion by THP-1 cells and reduced pro-caspase-1 but not pro-IL-1β. Treatment of THP-1 cells with STAT6 inhibitors, either Leflunomide or JAK I inhibitor, blocked suppression of caspase activity by IL-4 and IL-13. In vivo, MWCNTs alone caused neutrophilic infiltration into the lungs of mice 1 day post-exposure and increased IL-1β in bronchoalveolar lavage fluid (BALF) and pro-caspase-1 immuno-staining in macrophages and airway epithelium. HDM sensitization alone caused eosinophilic inflammation with increased IL-13. MWCNT exposure after HDM sensitization increased total cell numbers in BALF, but decreased numbers of neutrophils and IL-1β in BALF as well as reduced pro-caspase-1 in lung tissue. Despite reduced IL-1β mice exposed to MWCNTs after HDM developed more severe airway fibrosis by 21 days and had increased pro-fibrogenic cytokine mRNAs.

Conclusions

These data indicate that Th2 cytokines suppress MWCNT-induced inflammasome activation via STAT6-dependent down-regulation of pro-caspase-1 and suggest that suppression of inflammasome activation and IL-1β by an allergic lung microenvironment is a mechanism through which MWCNTs exacerbate allergen-induced airway fibrosis.  相似文献

Drawing on ethnographic research among transnational Moldovan households in Moscow, this essay considers how ideals of belonging, assertions of historically inflected rights, and aspirations for mobility are all part of the everyday practice of citizenship. Mobile subjects encountering increasingly restrictive post-Soviet citizenship regimes often recall incorporation into a greater historical polity than their current passports would suggest. Three key areas are examined: the intersection of citizenship regimes and popular understandings of belonging; the sense of rights driven by cultural logics informed by previous history; and the way in which ideals and practices of citizenship are diverse among migrants from apparently homogeneous migration streams. The post-Soviet context where the Soviet promise of enfranchisement continues to inform how people on the margins view citizenship illustrates just how deeply citizenship regimes come to be incorporated into popular understandings of belonging even long after formal citizenship ceases to exist.  相似文献

Is it possible to manipulate root anchorage in young trees?   总被引:1,自引:1,他引:0  
The optimal root system architecture for increased tree anchorage has not yet been determined and in particular, the role of the tap root remains elusive. In Maritime pine (Pinus pinaster Ait.), tap roots may play an important role in anchoring young trees, but in adult trees, their growth is often impeded by the presence of a hard pan layer in the soil and the tap root becomes a minor component of tree anchorage. To understand better the role of the tap root in young trees, we grew cuttings (no tap root present) and seedlings where the tap root had (?) or had not (+) been pruned, in the field for 7 years. The force (F) necessary to deflect the stem sideways was then measured and divided by stem cross-sectional area (CSA), giving a parameter analogous to stress during bending. Root systems were extracted and root architecture and wood mechanical properties (density and longitudinal modulus of elasticity, E L ) determined. In seedlings (?) tap roots, new roots had regenerated where the tap root had been pruned, whereas in cuttings, one or two lateral roots had grown downwards and acted as tap roots. Cuttings had significantly less lateral roots than the other treatments, but those near the soil surface were 14% and 23% thicker than plants (+) and (?) tap roots, respectively. Cuttings were smaller than seedlings, but were not relatively less resistant to stem deflection, probably because the thicker lateral roots compensated for their lower number. Apart from stem volume which was greater in trees (+) tap roots, no significant differences with regard to size or any root system variable were found in plants (?) or (+) tap roots. In all treatments, lateral roots were structurally reinforced through extra growth along the direction of the prevailing wind, which also improved tap root anchorage. Predictors of log F/CSA differed depending on treatment: in trees (?) tap roots, a combination of the predictors stem taper and %volume allocated to deep roots was highly regressed with log F/CSA (R 2 = 0.83), unlike plants (+) tap roots where the combined predictors of lateral root number and root depth were best regressed with log F/CSA (R 2 = 0.80). In cuttings, no clear relationships between log F/CSA and any parameter could be found. Wood density and E L did not differ between roots, but did diminish with increasing distance from the stem in lateral roots. E L was significantly lower in lateral roots from cuttings. Results showed that nursery techniques influence plant development but that the architectural pattern of Maritime pine root systems is stable, developing a sinker root system even when grown from cuttings. Anchorage is affected but the consequences for the long-term are still not known. Numerical modelling may be the only viable method to investigate the function that each root plays in adult tree anchorage.  相似文献

Bamboo is considered useful for controlling landslides, but we observed numerous shallow-slope failures in forests of big node bamboo (Phyllostachys nidularia) in Sichuan, China. Therefore, we inventoried landslide occurrence and vegetation type along one valley. To quantify bamboo root anchorage, we performed uprooting tests and measured plant morphological characteristics. Landslide occurrence was greatest at sites with bamboo and young trees. Culm failure was common because of the high length to diameter ratio (242 ± 6). Uprooting tests showed that the maximal force to cause failure was small (1615 ± 195 N). Uprooting force was strongly and positively regressed with a combination of the predictors lateral root number and volume (R(2) = 0.92), and root systems were highly superficial (depth = 0.15 ± 0.12 m), contributing little to slope stability. In P. nidularia, which grows on landslide-prone slopes, surprisingly few resources have been allocated to anchorage. We suggest that this strategy puts this pioneer at an advantage on steep slopes, where it contributes little to slope stability and colonizes frequently formed gaps through vegetative regeneration. Fewer disturbances would result in subsequent secondary succession and dying back of this shade intolerant species.  相似文献

Background and Aims In mountain ecosystems, predicting root density in three dimensions (3-D) is highly challenging due to the spatial heterogeneity of forest communities. This study presents a simple and semi-mechanistic model, named ChaMRoots, that predicts root interception density (RID, number of roots m–2). ChaMRoots hypothesizes that RID at a given point is affected by the presence of roots from surrounding trees forming a polygon shape.Methods The model comprises three sub-models for predicting: (1) the spatial heterogeneity – RID of the finest roots in the top soil layer as a function of tree basal area at breast height, and the distance between the tree and a given point; (2) the diameter spectrum – the distribution of RID as a function of root diameter up to 50 mm thick; and (3) the vertical profile – the distribution of RID as a function of soil depth. The RID data used for fitting in the model were measured in two uneven-aged mountain forest ecosystems in the French Alps. These sites differ in tree density and species composition.Key Results In general, the validation of each sub-model indicated that all sub-models of ChaMRoots had good fits. The model achieved a highly satisfactory compromise between the number of aerial input parameters and the fit to the observed data.Conclusions The semi-mechanistic ChaMRoots model focuses on the spatial distribution of root density at the tree cluster scale, in contrast to the majority of published root models, which function at the level of the individual. Based on easy-to-measure characteristics, simple forest inventory protocols and three sub-models, it achieves a good compromise between the complexity of the case study area and that of the global model structure. ChaMRoots can be easily coupled with spatially explicit individual-based forest dynamics models and thus provides a highly transferable approach for modelling 3-D root spatial distribution in complex forest ecosystems.  相似文献

Autophagy is a key degradative pathway coordinated by external cues, including starvation, oxidative stress, or pathogen detection. Rare are the molecules known to contribute mechanistically to the regulation of autophagy and expressed specifically in particular environmental contexts or in distinct cell types. Here, we unravel the role of RUN and FYVE domain–containing protein 4 (RUFY4) as a positive molecular regulator of macroautophagy in primary dendritic cells (DCs). We show that exposure to interleukin-4 (IL-4) during DC differentiation enhances autophagy flux through mTORC1 regulation and RUFY4 induction, which in turn actively promote LC3 degradation, Syntaxin 17–positive autophagosome formation, and lysosome tethering. Enhanced autophagy boosts endogenous antigen presentation by MHC II and allows host control of Brucella abortus replication in IL-4–treated DCs and in RUFY4-expressing cells. RUFY4 is therefore the first molecule characterized to date that promotes autophagy and influences endosome dynamics in a subset of immune cells.  相似文献

The finite element (FE) method has been used in recent years to simulate overturning processes in trees and to better comprehend plant anchorage mechanics. We aimed at understanding the fundamental mechanisms of root-soil reinforcement by simulating direct shear of rooted and non-rooted soil. Two- (2D) and three-dimensional (3D) FE simulations of direct shear box tests were carried out using readily available software for routine strength assessment of the root-soil composite. Both rooted and non-rooted blocks of soil were modelled using a simplified model of root distribution and root material properties representative of real roots. Linear elastic behaviour was assumed for roots and the soil was modelled as an ideally plastic medium. FE analysis showed that direct shear tests were dependent on the material properties specified for both the soil and roots. 2D and 3D simulations of direct shear of non-rooted soil produced similar results and any differences between 2D and 3D simulations could be explained with regard to the spatial complexity of roots used in the root distribution model. The application of FE methods was verified through direct shear tests on soil with analogue roots and the results compared to in situ tests on rooted soil in field conditions.  相似文献
