The first detailed morphological study of larvae, pupae and adults of a species of the hooded beetles (Coleoptera: Corylophidae) – Sericoderus lateralis – is presented. Histological sectioning, scanning and transmission electron microscopy, laser confocal microscopy and 3D-computer reconstruction were used. For the first time we report that according to the morphometric data of S. lateralis, at least some corylophid beetles have three larval stages. A phylogenetic position of Corylophidae within a cucujoid-cleroid clade is confirmed, and also the placement of Sericoderini within a corlyophid subgroup, which does not include Periptycinae and Foadiini. The larvae of Sericoderus are mainly characterized by plesiomorphic features compared to those of other corylophid tribes, notably Peltinodini and Rypobiini. Morphological and developmental consequences of miniaturization are discussed. Corylophid beetles display much less specific and far-reaching morphological consequences of miniaturization compared to Ptiliidae. We report the presence of unique modifications in the neural system not shared with any other insects, such as a distinctly asymmetric supraoesophageal ganglion in first instar larva, and a total displacement of the brain to the thorax in the adult stage. A highly unusual feature of the digestive tract is the sclerotised, V-shaped ventral wall of the pharynx. Developmental and size dependent changes in the relative volume of different organs are addressed. All organ systems change allometrically in the development of S. lateralis. Allometric trends in the volume of organs confirm that the factors limiting miniaturization are the size of the neural system, associated with the number and size of neurons (most critical for first instar larva), the mass of the skeleton, the egg size, and consequently the volume of the reproductive system (for free-living insects). 相似文献

Adequate support of energy for biological activities and during fluctuation of energetic demand is crucial for healthy aging; however, mechanisms for energy decline as well as compensatory mechanisms that counteract such decline remain unclear. We conducted a discovery proteomic study of skeletal muscle in 57 healthy adults (22 women and 35 men; aged 23–87 years) to identify proteins overrepresented and underrepresented with better muscle oxidative capacity, a robust measure of in vivo mitochondrial function, independent of age, sex, and physical activity. Muscle oxidative capacity was assessed by 31P magnetic resonance spectroscopy postexercise phosphocreatine (PCr) recovery time (τPCr) in the vastus lateralis muscle, with smaller τPCr values reflecting better oxidative capacity. Of the 4,300 proteins quantified by LC‐MS in muscle biopsies, 253 were significantly overrepresented with better muscle oxidative capacity. Enrichment analysis revealed three major protein clusters: (a) proteins involved in key energetic mitochondrial functions especially complex I of the electron transport chain, tricarboxylic acid (TCA) cycle, fatty acid oxidation, and mitochondrial ABC transporters; (b) spliceosome proteins that regulate mRNA alternative splicing machinery, and (c) proteins involved in translation within mitochondria. Our findings suggest that alternative splicing and mechanisms that modulate mitochondrial protein synthesis are central features of the molecular mechanisms aimed at maintaining mitochondrial function in the face of impairment. Whether these mechanisms are compensatory attempt to counteract the effect of aging on mitochondrial function should be further tested in longitudinal studies. 相似文献

We have developed a model for evaluating the integral intercellular interactions in the “endotheliocyte‐neutrophil” system and have shown the high variability of adhesion contacts in different donors associated with different expression profiles of neutrophils. Two methods (forсe spectroscopy‐spectroscopy and scanning ion‐conductance microscopy) showed a decrease in the rigidity of the membrane‐cytoskeletal complex of neutrophils under the influence of Staphylococcus aureus 2879 M. Adding this strain to the “endotheliocyte‐neutrophil” system caused a statistically significant decrease in the adhesion force and adhesion work, which indicates a change in the expression profile and physicochemical properties of membranes of both types of interacting cells (neutrophils and endotheliocytes). 相似文献

The reflection of a test electromagnetic pulse from the plasma formed as a result of tunnel ionization of atoms in the field of a circularly polarized high-power radiation pulse is analyzed using the kinetic approach to describe electron motion. It is shown that the reflected pulse is significantly amplified due to the development of Weibel instability. Alzheimer disease is associated with the accumulation of oligomeric amyloid β peptide (Aβ), accompanied by synaptic dysfunction and neuronal death. Polymeric form of prion protein (PrP), PrPSc, is implicated in transmissible spongiform encephalopathies (TSEs). Recently, it was shown that the monomeric cellular form of PrP (PrPC), located on the neuron surface, binds Aβ oligomers (and possibly other β-rich conformers) via the PrP23–27 and PrP90–110 segments, acting as Aβ receptor. On the other hand, PrPSc polymers efficiently bind to Aβ monomers and accelerate their oligomerization. To identify specific PrP sequences that are essential for the interaction between PrP polymers and Aβ peptide, we have co-expressed Aβ and PrP (or its shortened derivatives), fused to different fluorophores, in the yeast cell. Our data show that the 90–110 and 28–89 regions of PrP control the binding of proteinase-resistant PrP polymers to the Aβ peptide, whereas the 23–27 segment of PrP is dispensable for this interaction. This indicates that the set of PrP fragments involved in the interaction with Aβ depends on PrP conformational state. 相似文献

Opisthorchis felineus or Siberian liver fluke is a trematode parasite (Opisthorchiidae) that infects the hepato-biliary system of humans and other mammals. Despite its public health significance, this wide-spread Eurasian species is one of the most poorly studied human liver flukes and nothing is known about its population genetic structure and demographic history. In this paper, we attempt to fill this gap for the first time and to explore the genetic diversity in O. felineus populations from Eastern Europe (Ukraine, European part of Russia), Northern Asia (Siberia) and Central Asia (Northern Kazakhstan). Analysis of marker DNA fragments from O. felineus mitochondrial cytochrome c oxidase subunit 1 and 3 (cox1, cox3) and nuclear rDNA internal transcribed spacer 1 (ITS1) sequences revealed that genetic diversity is very low across the large geographic range of this species. Microevolutionary processes in populations of trematodes may well be influenced by their peculiar biology. Nevertheless, we suggest that lack of population genetics structure observed in O. felineus can be primarily explained by the Pleistocene glacial events and subsequent sudden population growth from a very limited group of founders. Rapid range expansion of O. felineus through Asian and European territories after severe bottleneck points to a high dispersal potential of this trematode species. 相似文献
