An analytical model, which describes the stationary transformation of light energy to the energy of pigment electronic excitation, has been constructed. A proton pump of the thylakoid membrane has been considered as a two-level conformon. The difference between the energies of the excited and ground states of both the pigment and the protein complex is assumed to be the energy of an absorbed photon. It has been found how the concentration of ions in a lumen and the potential across the thylakoid membrane depend on the concentration of ions in the stroma and the brightness temperature of absorbed radiation. Conditions for the maximum efficiency of the photosynthesis process have been analyzed. This model has been used to determine the electric potential (φ≈6.7 mV) at the chloroplast thylakoid membrane. The calculated value of the electric potential is in good agreement with the experimental data. A limitation on the stoichiometric coefficient of the proton transport through ATP-synthase, m>3, has been found theoretically.  相似文献

Preparation of pure ribosomal subunits carrying lethal mutations is necessary for studying every essential functional region of ribosomal RNA. Affinity purification via a tag, inserted into rRNA proved to be procedure of choice for purification of such ribosomal subunits. Here we describe fast and simple purification method for the 30S ribosomal subunits using affinity chromatography. Streptavidin-binding tag was inserted into functionally neutral helix 33a of the 16 S rRNA from Escherichia coli. Tagged ribosomal subunits were shown to be expressed in E. coli and could be purified. Purified subunits with affinity tag behave similarly to the wild type subunits in association with the 50S subunits, toe-printing and tRNA binding assays. Tagged 30S subunits could support cell growth in the strain lacking wild type 30S subunits and only marginally change the growth rate of bacteria. The presented purification method is thus suitable for further use in purification of 30S subunits carrying any lethal mutations.  相似文献

Monoclonal antibodies (Mabs) are a favorite drug platform of the biopharmaceutical industry. Currently, over 20 Mabs have been approved and several hundred others are in clinical trials. The anti‐LINGO‐1 Mab Li33 was selected from a large panel of antibodies by Fab phage display technology based on its extraordinary biological activity in promoting oligodendrocyte differentiation and myelination in vitro and in animal models of remyelination. However, the Li33 Fab had poor solubility when converted into a full antibody in an immunoglobulin G1 framework. A detailed analysis of the biochemical and structural features of the antibody revealed several possible reasons for its propensity to aggregate. Here, we successfully applied three molecular approaches (isotype switching, targeted mutagenesis of complementarity determining region residues, and glycosylation site insertion mutagenesis) to address the solubility problem. Through these efforts we were able to improve the solubility of the Li33 Mab from 0.3 mg/mL to >50 mg/mL and reduce aggregation to an acceptable level. These strategies can be readily applied to other proteins with solubility issues.  相似文献

We employ complementary field and laboratory‐based incubation techniques to explore the geochemical environment where siderite concretions are actively forming and growing, including solid‐phase analysis of the sediment, concretion, and associated pore fluid chemistry. These recently formed siderite concretions allow us to explore the geochemical processes that lead to the formation of this less common carbonate mineral. We conclude that there are two phases of siderite concretion growth within the sediment, as there are distinct changes in the carbon isotopic composition and mineralogy across the concretions. Incubated sediment samples allow us to explore the stability of siderite over a range of geochemical conditions. Our incubation results suggest that the formation of siderite can be very rapid (about two weeks or within 400 hr) when there is a substantial source of iron, either from microbial iron reduction or from steel material; however, a source of dissolved iron is not enough to induce siderite precipitation. We suggest that sufficient alkalinity is the limiting factor for siderite precipitation during microbial iron reduction while the lack of dissolved iron is the limiting factor for siderite formation if microbial sulfate reduction is the dominant microbial metabolism. We show that siderite can form via heated transformation (at temperature 100°C for 48 hr) of calcite and monohydrocalcite seeds in the presence of dissolved iron. Our transformation experiments suggest that the formation of siderite is promoted when carbonate seeds are present.  相似文献

In the present work, a positive effect was obtained by using low molecular weight chitosan compounds in combination with synthetic fungicides. Antifungal activity against Botrytis cinerea, determined by the radial growth method, was more than 75%, with a 25?×?10??10 g/L concentration of fludioxonil or difenoconazole in compounds. Metabolic activity of B. cinerea fungus was about 15% when using a chitosan compound containing fludioxonil at a concentration of 25?×?10??7 g/L. The combined action of chitosan with difenoconazole at a fungicide concentration of 25?×?10??4 g/L is 2–3 times more effective than the action of each component separately. Results of studies for artificially inoculated B. cinerea tomato fruit when treated with low molecular chitosan and chitosan conjugate with gallic acid reduced the frequency of rotting fruit by 50 and 83%, respectively. Chitosan-gallic acid conjugate were obtained from chitosans with Mw of 28 kDa (Ch28GA) was proved to be effective as a preventive treatment for 3 days and can potentially be used as a biofungicide against B. cinerea on tomatoes in the post-harvest period.
