In this article the authors argue that the study of the ongoing significance of significant dreams necessarily goes beyond quantitative methods for analyzing dream content to a qualitative study of how the dream experience influences the dreamer's meaning-making processes. A set of concepts from narrative psychology is introduced as being potentially valuable in this regard. The paper describes a sensitive and rapid zymogram technique for detection of transglycosylating activity (XET) of xyloglucan endotransglycosylase/hydrolase (XTH; EC 2.4.1.207) in polyacrylamide isoelectric focusing gels. After the electrophoresis, the separating gel was overlaid and incubated with an agarose detection gel containing XET substrates: tamarind-seed xyloglucan as the glycosyl donor and sulphorhodamine-labeled xyloglucan-derived oligosaccharides (XGO-SRs) as the glycosyl acceptors. The transglycosylation catalyzed by XTH caused incorporation of the fluorescent label into the high-M(r) polysaccharide. Selective removal of unreacted XGO-SRs from the agarose replicas by washing with organic solvents revealed the zones corresponding to XET activity as bright pink fluorescent spots under UV-light. The method appears suitable for a number of purposes such as analysis of the isoenzyme composition of XTHs with XET activity in crude extracts from various plants and plant organs, monitoring the enzyme expression at various stages of plant development and/or for checking enzyme purity in the course of its isolation procedure. 相似文献

Plumbous ion has been shown to catalyze the degradation of RNA, and the effects of lead-catalyzed depolymerization on some of the functions of tRNA has been investigated. Exposure to Pb2+ brings about a rapid decrease but not a complete loss of amino acid acceptance capacity. The tRNA molecules responsible for this residual activity have not been degraded and represent a class of molecules that are resistant to Pb2+. The ratio of susceptible to resistant tRNA increases when the temperature is raised. The effect of varying the Pb2+ concentration was studied, and there is a threshold Pb2+ concentration below which the tRNA is not inactivated. 相似文献

Summary Manganese added to the growth medium of Saccharomyces cerevisiae to a final concentration of 4–8 mM induces not only mitochondrial respiratory-deficient mutations (Fig. 1), but also mitochondrial mutations to chloramphenicol- and erythromycin-resistance (Fig. 1, Tables 1–3). This is the first mutagen shown to be capable of inducing mitochondrial antibiotic resistance mutations in yeast.
