Yeast and bovine cytochrome c oxidases (COX) are composed of 12 and 13 different polypeptides, respectively. In both cases, the three subunits constituting the catalytic core are encoded by mitochondrial DNA. The other subunits are all products of nuclear genes that are translated on cytoplasmic ribosomes and imported through different transport routes into mitochondria. Biogenesis of the functional complex depends on the expression of all the structural and more than two dozen COX-specific genes. The latter impinge on all aspects of the biogenesis process. Here we review the current state of information about the functions of the COX-specific gene products and of their relationship to human COX deficiencies. 相似文献

The synthesis of new zirconium and hafnium mixed ligand phthalocyanine complexes PcM(β-ketoester)2, where M-Zr (IV), Hf (IV); Pc - the dianion of phthalocyanine, and β-ketoester - the out planed ligand, is reported. The obtained complexes are characterized by 1H NMR, IR, UV-Vis spectroscopy and cyclic voltammetry. 1H NMR and elemental analysis confirm the substitution of two Cl atoms for two β-ketoester fragments to the central atom of the macrocycle. The data of 1H NMR, UV-Vis spectroscopy have allowed us to conclude that two β-ketoester ligands are in the cis geometry to the phthalocyanine plane. X-ray crystallography for bis(isopropyl 3-oxobutanoato)hafnium(IV)phthalocyanine confirms this conclusion. The central macrocycle of the phthalocyanine ligand is not exactly planar (deviations from the least-square plane exceed 0.15 Å) and has the conformation of an essentially flattened crown. The Hf(1) atom is 1.349(3) above this least-square plane. Cyclic voltammetry investigation shows that the introduction of two β-ketoester ligands to the central atom of phthalocyanine complex leads to both chemical and electrochemical stabilization of the whole Pc system. 相似文献

Activity-dependent validation of excitatory versus inhibitory synapses by neuroligin-1 versus neuroligin-2 总被引:8,自引:0,他引:8
Neuroligins enhance synapse formation in vitro, but surprisingly are not required for the generation of synapses in vivo. We now show that in cultured neurons, neuroligin-1 overexpression increases excitatory, but not inhibitory, synaptic responses, and potentiates synaptic NMDAR/AMPAR ratios. In contrast, neuroligin-2 overexpression increases inhibitory, but not excitatory, synaptic responses. Accordingly, deletion of neuroligin-1 in knockout mice selectively decreases the NMDAR/AMPAR ratio, whereas deletion of neuroligin-2 selectively decreases inhibitory synaptic responses. Strikingly, chronic inhibition of NMDARs or CaM-Kinase II, which signals downstream of NMDARs, suppresses the synapse-boosting activity of neuroligin-1, whereas chronic inhibition of general synaptic activity suppresses the synapse-boosting activity of neuroligin-2. Taken together, these data indicate that neuroligins do not establish, but specify and validate, synapses via an activity-dependent mechanism, with different neuroligins acting on distinct types of synapses. This hypothesis reconciles the overexpression and knockout phenotypes and suggests that neuroligins contribute to the use-dependent formation of neural circuits. 相似文献

Telomeres are DNA-protein complexes at the ends of chromosomes that control genomic integrity but appear to become shorter with age and stress. To test whether stress causes telomere attrition, we exposed the offspring of wild-caught house mice (Mus musculus) to stressful conditions and examined the changes in telomere length over six months. We found that females exposed to males and reproductive stress (either with or without crowding) had significantly shorter telomeres than controls, and males exposed to crowding stress had shorter telomeres than males that were not crowded. Our results indicate that stress alters telomere dynamics, causing attrition and hindering restoration, and these effects are sex dependent. Telomeres may thus provide a biomarker for assessing an individual's cumulative exposure or ability to cope with stressful conditions. 相似文献

Since the 1970s the area under sugarcane in Brazil has increased from 2 million to over 5 million ha (M ha), and it is expected
to pass the 7 M ha mark in 2007. More than half of the cane is harvested to produce bioethanol as a fuel for light vehicles.
The distilleries produce approximately 13 L of distillery waste (vinasse) for each litre of ethanol produced. In the 1980s
there was considerable concern over the long-term effects of the disposal of this material (containing about 1% carbon and
high in K) on cane yields if it was applied to the field. At the same time there was a growing movement to abandon the practice
of pre-harvest burning and some research was showing that some Brazilian varieties of sugar cane were able to obtain significant
contributions of N from plant-associated biological nitrogen fixation (BNF). For these reasons an experiment was installed
on a cane plantation in the state of Pernambuco, NE Brazil to investigate the long-term effects of vinasse and N fertiliser
additions and the practice of pre-harvest burning on crop and sugar yield, soil fertility parameters, N balance and soil C
stocks. The results showed that over a 16-year period, trash conservation (abandonment of burning) increased cane yields by
25% from a mean of 46 to 58 Mg ha−1. Vinasse applications (80 m3 ha−1 crop−1) increased mean cane and sugar yield by 12 to 13% and the application of 80 kg N ha−1 as urea increased cane yields by 9%, but total sugar yield by less than 6% (from 7.0 to 7.4 Mg ha−1 crop−1). The total N balance for the soil/plant system when only the surface 20 cm of the soil was considered was positive in plots
where no N fertiliser was added. However, the data indicated that during the 16 years of the study considerable quantities
of soil organic matter were accumulated below 20 cm depth such that the N balance considering the soil to 60 cm depth was
strongly positive, except where N fertiliser was added. The data indicated that there were considerable BNF inputs to the
system, which was consistent with its low response to N fertiliser and low N fertiliser-use-efficiency. There were no significant
effects of vinasse or urea addition, or trash conservation on soil C stocks, although the higher yields proportioned by trash
conservation had potentially significant benefits for increased mitigation of CO2 emissions where the main use of the cane was for bioethanol production. 相似文献

We have recently reported about a new class of Aurora-A inhibitors based on a bicyclic tetrahydropyrrolo[3,4-c]pyrazole scaffold. Here we describe the synthesis and early expansion of CDK2/cyclin A-E inhibitors belonging to the same chemical class. Synthesis of the compounds was accomplished using a solution-phase protocol amenable to rapid parallel expansion. Compounds with nanomolar activity in the biochemical assay and able to efficiently inhibit CDK2-mediated tumor cell proliferation have been obtained. 相似文献

- Marked populations of Limulus (=Xiphosura) polyphemus reveal that in Cold Spring Harbor, New York, they consisted of 10,000–18,000 adults in 1957 and 1961. The sex ratio in 1957 was about 4 males: 1 female. Pairs may remain attached for as long as 9 days. An undisturbed female may lay as many as 12,000 eggs in one nest.
- The Cold Spring Harbor populations appear to be rather sedentary: none of the 1,000 animals marked on the north edge of the sandspit in 1961 were detected in the outer harbor either at Laurel Hollow Beach or the peninsula adjacent to the Cold Spring Harbor Yacht Club 500–800 meters from the tagging site (see Fig. 1), nor were they found in the small beach adjacent to the Biological Laboratory in the inner harbor. Similarly, none of the 300 animals marked at this last site were found at the north edge of the sandspit.
- The phenotype of the compound eye varies from black to pigmentless. Samples observed in Cold Spring Harbor and in the Marine Biological Laboratory, Woods Hole, Massachusetts (separated by Long Island Sound and a distance of 150 miles) differ in the frequency of the various phenotypes scored, but the mode of inheritance of eye color remains obscure.
- The available evidence indicates Limulus has considerable phenotypic variation in regard to body size, eye color, and other characters believed to be inherited, with the result that demes or physiological races are created. It is argued that the belief that this organism is stable and has not changed since the Triassic 200 million years ago has foundation only in regard to the pattern of the body of Limulus, but not in regard to its genotype. Limulus does not seem to be different from other organisms for which considerable genetic evidence is available, and thus the statement that DNA is fairly stable and has remained so for 200 million years is open to question.
