Transformed root tissue of Beta vulgaris (Detroit Dark Red) was permeabilized to stimulate the release of intracellularly stored betanin without adverse affects on tissue viability as measured by biomass accumulation. Product release of up to 15% (w/w) was achieved by heat treatment at 42°C for 45 min with minimal effect on viability. Higher levels of product release were obtained with increasing temperature and exposure, but at the expense of viability. Viability was measured by comparing dry weight increases of permeabilized tissue 3 days after treatment vs non-permeabilized tissue over the same time interval. Recovery of heat-treated tissue was improved by addition of CaCl2 (20 mm for 10 min) post-heat treatment. Betanin release up to 15% was also obtained at ambient temperature (25°C) by addition of up to 20 mm (NH4)2SO4 in the presence of 1 mm ethylenediaminetetraacetic acid (EDTA). Correspondence to: A. A. This volume follows failures out into the world, exploring how they unfold ethnographically. Taking a longer view shows how objects, narratives, and diagnoses of failures may be crafted, acted on, suffered, resisted – unmade or recomposed. Thus while tropes and diagnoses of failure can temporarily (re)organize, narrate, and stabilize the world, the kinds of failures explored here also indicate a mode of uncontainable excess that refuses the boundedness of knowledge objects, temporalities, and spaces. This volume offers three main interventions. The first concerns knowledge production: how objects of failure are crafted through selective ways of knowing that occlude both other modes of apprehension at different scales and failure's many affective valences. The second thinks through the knotted temporalities – whether pasts, futures, suspended presents, or repetition and sedimentation – that make and are made by failure. Finally, writing about unfurling failures requires careful attention to non-linear reverberations and traces as well as to open-ended and mobile narratives that produce different social and material effects.  相似文献

Zervamicin IIB is a member of the alpha-aminoisobutyric acid containing peptaibol antibiotics. A new procedure for the biosynthetic preparation of the uniformly 13C- and 15N-enriched peptaibol is described This compound was isolated from the biomass of the fungus-producer Emericellopsis salmosynnemata strain 336 IMI 58330 obtained upon cultivation in the totally 13C, 15N-labelled complete medium. To prepare such a medium the autolysed biomass and the exopolysaccharides of the obligate methylotrophic bacterium Methylobacillus flagellatus KT were used. This microorganism was grown in totally 13C, 15N-labelled minimal medium containing 13C-methanol and 15N-ammonium chloride as the only carbon and nitrogen sources. Preliminary NMR spectroscopic analysis indicated a high extent of isotope incorporation (> 90%) and led to the complete 13C- and 15N-NMR assignment including the stereospecific assignment of Aib residues methyl groups. The observed pattern of the structurally important secondary chemical shifts of 1H(alpha), 13C=O and 13C(alpha) agrees well with the previously determined structure of zervamicin IIB in methanol solution.  相似文献

One of the oldest known helcionelloid molluscs, Anabarella Vostokova, is redescribed on the basis of type material from the lower Cambrian of north Siberia. Published records of the type species, Anabarella plana Vostokova, show a very wide range in morphology, but studies of variation through ontogeny and in taphonomy confirm assignment to a single variable species. A current challenge in computational neuroscience is to elucidate the role of cortical circuitry in information processing and in generating motor output. Our understanding of the functional significance of specifically organized feedback connections is progressing rapidly as researchers establish the equivalence of theoretical models to biological neural circuits. Modeling studies of different neural structures, along with quantitative comparisons of model performance to biological data, have recently helped to identify the basic features of synaptic connectivity that may play important roles in cortical operations.  相似文献

Light-triggered membrane potential changes in cells of a liverwort Anthoceros are greatly enhanced by the ionophorous uncouplers nigericin and monesin. Stimulation of the light-triggered electrical response (LTER) by nigericin occurred concomitantly with inhibition of a slow decline in the chlorophyll fluorescence, which suggests that the transmembrane pH gradient in thylakoids is not essential for generation of LTER at the plasma membrane. The extent of monensin-stimulated LTER remained high under a diminished driving force for the ionophore-induced proton-cation exchange across the plasma membrane (elevation of the external Na+ concentration from 1 to 50 m M ), which indicates that energy uncoupling in chloroplasts is more related to the electric response enhancement than the induction of the H+/K+(Na+) exchange at the plasma membrane. Enhancement of LTER by ionophores occurs in parallel with stimulation of light-triggered pH changes (alkalinization) in the vicinity of the cell surface, which suggests an association of trans-membrane H+ fluxes with LTER. The results are consistent with the hypothesis that illumination produces a temporary inhibition of the plasma membrane H+ pump with a subsequent activation of gated channels and transient rapid depolarization of the cell.  相似文献
