According to sympatric speciation theory, adaptation to different host plants is expected to pleiotropically lead to assortative mating, an important factor in the reduction of gene flow between the diverging subpopulations. This scenario predicts mating on and oviposition preference for the respective hosts in both the diverging subpopulations and recently originated species. Here, we test both predictions in the oligophagous Yponomeuta padellus (L.) and the monophagous Yponomeuta cagnagellus (Hübner) (Lepidoptera: Yponomeutidae), two closely related small ermine moth species from the western European clade of Yponomeuta for which speciation in sympatry has been proposed. Mating location and adult host acceptance were evaluated under both semi-field (in a large outdoor cage with a choice of host and non-host plants) and field conditions. In the semi-field experiment, only Y. cagnagellus showed some preference for mating on its own host (16% of all mating pairs) over non-host plants (3% of all mating pairs). However, in both species, more than 80% of the mating pairs were not formed on a plant but instead on the cage itself. Further examination of the mating site of Y. cagnagellus in the field revealed no preference for host plants over non-host plants in the two consecutive years of observation. Yponomeuta padellus females, collected from and reared on Prunus spinosa L. (Rosaceae), showed an oviposition preference for the alternative host Crataegus monogyna Jacq. (Rosaceae) in the semi-field experiment. We thus found no evidence that host-plant fidelity (in terms of mating site) has been the driving force in the speciation process of these Yponomeuta species, nor did we find evidence of host race formation in the tested population of the oligophagous Y. padellus .  相似文献

Despite recent modelling approaches integrating the effects of niche-based processes and dispersal-based processes on local plant species composition, their relative importance is still not clear. We test whether the predictability of local species composition from environmental conditions is influenced by dispersal traits. We analyzed a large database with co-occurrence data, using ordination techniques (DCA and CCA) to identify the major environmental determinants of species composition. The percentage of explained variance in occurrence was quantified for individual species with CCA. Effects of life-history traits on the predictability of occurrence patterns were tested by means of regression analysis, using a generalized linear models approach. The results reveal close correlations between species composition and environmental conditions, implying that the predictability of the set of species that might occur in a given environmental setting ("habitat species pool") is high. The habitat species pool, however, reflects the potential species composition, and not the actual local situation. At the level of individual species, a large proportion (>90%) of the variation in occurrence remained unexplained. Predictability of species occurrence patterns was increased by a greater capacity for long-distance dispersal, greater adult longevity and the capacity to build a persistent seed bank. The results indicate that the predictability of species composition from environmental conditions is reduced by a few orders of magnitude by dispersal limitation and that poor dispersers are underrepresented.  相似文献

The volatile organic compounds of different ground hay samples from Austria, Italy and Switzerland were collected at 50 degrees C on a Supelco Carbowax Divinylbenzene headspace solid-phase microextraction fibre, separated by capillary gas chromatography on an HP5-ms column running a temperature programme and using helium as carrier gas, detected with a mass sensitive detector and studied with principal component analyses after autoscaling selected variables. The analytes, mainly mono- and sesquiterpenes, were able to cluster differences resulting from the site of production. Coumarin can be used to differentiate hay grown north and south of the main chain of the Alps. Acetic acid is appropriate for distinguishing between hay from Kastelruth and Passeier Valley, two South Tyrolean regions. The average linalool content in aftermath is higher than in hay.  相似文献

Background

Small interfering (si)RNA mediated inhibition of oncogenes or viral genes may offer great opportunities for the treatment of several diseases such as hepatocellular carcinoma and viral hepatitis. However, the development of siRNAs as therapeutic agents strongly depends on the availability of safe and effective intracellular delivery systems. Poly(β‐amino esters) (PbAEs) are, in contrast to many other cationic polymers evaluated in siRNA delivery, biodegradable into smaller, nontoxic molecules.

Methods and Results

We show for the first time that PbAE : siRNA complexes, containing 1,4‐butanediol (PbAE1) or 1,6‐hexanediol (PbAE2) diacrylate‐based polymers, induced efficient gene silencing in both hepatoma cells and primary hepatocytes without causing significant cytotoxicity. Furthermore, carriers that slowly release the siRNA into the cytoplasm and hence induce a prolonged gene silencing are of major clinical interest, especially in fast dividing tumour cells. Therefore, we also studied the duration of gene silencing in the hepatoma cells and found that it was maintained for at least 5 days after siRNA delivery with PbAE2, the polymer with the slowest degradation kinetics.

Conclusions

From the time‐dependent cellular distribution of these PbAE : siRNA complexes, we suggest that the slowly degrading PbAE2 causes a sustained endosomal release of siRNA during a much longer period than PbAE1. This may support the hypothesis that the endosomal release mechanism of PbAE : siRNA complexes is based on an increase of osmotic pressure in the endosomal vesicles after polymer hydrolysis. In conclusion, our results show that both PbAEs, and especially PbAE2, open up new perspectives for the development of efficient biodegradable siRNA carriers suitable for clinical applications. Pattern recognition receptors (PRRs) may contribute to inflammatory bowel diseases (IBD) development due to their microbial-sensing ability and the unique microenvironment in the inflamed gut. In this study, the PRR mRNA expression profile together with T cell-associated factors in the colon was examined using a chronic colitis mice model. 8–12 week old C57BL/6 mice were exposed to multiple dextran sodium sulfate (DSS) treatments interspersed with a rest period to mimic the course of chronic colitis. The clinical features and histological data were collected. The mRNA expressions of colonic PRRs, T cell-associated components were measured. Finally, the colons were scored for Foxp3+ cells. During chronic colitis, the histological data, but not the clinical manifestations demonstrated characteristic inflammatory symptoms in the distal colon. In contrast to acute colitis, the expression of all Toll-like receptors (Tlrs), except Tlr5 and Tlr9, was unaffected after repeated DSS treatments. The expression of Nod1 was decreased, while Nod2 increased. After third DSS treatment, only the expressions of Tlr3 and Tlr4 were significantly enhanced. Unlike other PRRs, decreased Tlr5 and increased Tlr9 mRNA expression persisted during the chronic colitis period. As the colitis progress, only the mRNA expression of Ifnγ and Il17 staid increased during chronic colitis, while the acute colitis-associated increase of Il23, and Il10 and Il12 was abolished. Finally, increased histological score of Foxp3+ cell in colon was found during the chronic colitis period. This study provides an expression pattern of PRRs during chronic colitis that is accompanied by a Th1- and Th17 cell-mediated immune response.  相似文献

The microspatial distribution of carabid beetles in a pristine alluvial Quercus-Ulmus forest along the river Elbe (north Germany) was investigated using 152 pitfall traps over a study period of 28 d. Traps were set in an area of only 100 × 120 m which comprised a wide range of microhabitats due to an extremely heterogeneous microrelief. Composition of the ground beetle assemblage was strongly influenced by microclimatic parameters and vegetation structure within a few metres: specifically soil moisture, light intensity. pH as well as cover of leaf litter and herbs. TWINSPAN analyses indicated a sensitive segregation of carabids into distinct microhabitats on a small scale. PCA and subsequent RDA distinguished two distinct species groups: species of wet deciduous woodland and species of wet open marsh habitats. The fringes of small temporary waters within the alluvial forest were of particular value for nature conservation, providing habitat for a high number of rare species. Although there was a shift in species composition over time and the assemblages reorganised spatially, the main environmental parameters governing the community remained the same as indicated by discriminant analyses, namely soil moisture, leaf litter cover and light intensity.  相似文献

Staphylococcus aureus is frequently detected in patients with sepsis and thus represents a major health burden worldwide. CD4+ T helper cells are involved in the immune response to S. aureus by supporting antibody production and phagocytosis. In particular, Th1 and Th17 cells secreting IFN-γ and IL-17A, are involved in the control of systemic S. aureus infections in humans and mice.To investigate the role of T cells in severe S. aureus infections, we established a mouse sepsis model in which the kidney was identified to be the organ with the highest bacterial load and abundance of Th17 cells. In this model, IL-17A but not IFN-γ was required for bacterial control. Using Il17aCre × R26YFP mice we could show that Th17 fate cells produce Th17 and Th1 cytokines, indicating a high degree of Th17 cell plasticity. Single cell RNA-sequencing of renal Th17 fate cells uncovered their heterogeneity and identified a cluster with a Th1 expression profile within the Th17 cell population, which was absent in mice with T-bet/Tbx21-deficiency in Th17 cells (Il17aCre x R26eYFP x Tbx21-flox). Blocking Th17 to Th1 transdifferentiation in Th17 fate cells in these mice resulted in increased S. aureus tissue loads.In summary, we highlight the impact of Th17 cells in controlling systemic S. aureus infections and show that T-bet expression by Th17 cells is required for bacterial clearance. While targeting the Th17 cell immune response is an important therapeutic option in autoimmunity, silencing Th17 cells might have detrimental effects in bacterial infections.  相似文献

Abstract. Species richness in calcareous grassland is discussed against the background of historical dispersal processes associated with traditional land-use management such as grazing, but also the artificial establishment by hayseed. Important vectors in the traditionally man-made landscape were sheep and cattle or other livestock such as goats. Calcareous grasslands were not only connected to each other but also to other habitats such as villages, forests, arable fields and heathlands by these vectors which could cover large distances (e.g. transhumance shepherding), which is not the case in the current man-made landscape. Species richness after restoration management of abandoned and afforested calcareous grasslands was predicted by using characters of dispersability in space and time. These were the persistence of the species in the vegetation and the diaspore bank after abandonment or afforestation and the dispersal capacity through wind and sheep. The results reveal that reintroduction of sheep grazing is necessary to reestablish the original species richness. The first validation of the prediction of the succession on clear-cut sites and a comparison with data of species composition in abandoned quarries and the surroundings made it obvious that a species' own dispersal capacity in space is very low except for some well wind-dispersed species. Therefore, it is recommended to include and to simulate dispersal processes in future management to be able to restore the original species richness.  相似文献

Summary Formaldehyde-induced fluorescence (Falck-Hillarp technique) provided histochemical evidence for the presence of catecholamines in the sensory epithelia (macula and crista) of the Octopus statocyst. A specific bright green fluorescence occurred in the neuronal plexus beneath the receptor cell layers of the epithelia and in the appropriate nerves. The histochemical findings are discussed with reference to the well-known neuronal and synaptic organization of the epithelia and to relevant results in cephalopods as well as in other molluscs. All data support the hypothesis that in the receptor systems of the Octopus statocyst catecholamines (probably dopamine and/or noradrenaline) act as neurotransmitters in the efferent fibre system.  相似文献
