Summary The woody species kiwi (Actinidia deliciosa A. Chev.), a male and late flowering clone of the cv Hayward, has been transformed by a T-DNA fragment encompassing rol A, B, C genes of A. rhizogenes. Transgenic plants, regenerated from leaf disc callus, showed the typical hairy root phenotype as described in herbaceous species. Explants from these plants (both leaf discs or 3 to 4 node leafy microcuttings) showed an increased ability to produce roots. Since root formation is one of the limiting factors in the vegetative propagation of woody species, the results have been discussed in relation to the use of A. rhizogenes rol genes in improving root morphogenesis in trees.  相似文献

Peptide T (ASTTTNYT) is a promising molecule to prevent the neuropsychometric symptoms of patients suffering AIDS and for the treatment of psoriasis. In order to fully prove its therapeutic benefits, efforts were put forward to design peptidomimetics of the peptide. In this direction, in a recent computational study the natural product amygdalin was identified as a prospective peptidomimetic of the peptide and later proved to exhibit a similar chemotactic profile to the peptide. However, the cyanide moiety of amygdalin provides to the molecule a toxic profile. The present study reports the synthesis of a set of amygdalin analogs lacking the cyanide group with improved chemotactic profiles.  相似文献

Unless metastatic or compressing the pancreatic duct, carcinoid of the pancreas are asymptomatic showing normal levels of serotonine and its metabolites in plasma and urine, thus resulting in delayed diagnosis and a consequent poor prognosis. Nitrophorins (NPs) are nitric oxide (NO)-carrying heme proteins found in the saliva of the blood-sucking insect Rhodnius prolixus. Though NP7 exhibits a large sequence resemblance with other NPs, two major differential features are the ability to interact with negatively charged cell surfaces and the presence of a specific N-terminus composed of three extra residues (Leu1-Pro2-Gly3). The aim of this study is to examine the influence of the N-terminus on the ligand binding, and the topological features of inner cavities in closed and open states of NP7, which can be associated to the protein structure at low and high pH, respectively. Laser flash photolysis measurements of the CO rebinding kinetics to NP7 and its variant NP7(Δ1–3), which lacks the three extra residues at the N-terminus, exhibit a similar pattern and support the existence of a common kinetic mechanism for ligand migration and binding. This is supported by the existence of a common topology of inner cavities, which consists of two docking sites in the heme pocket and a secondary site at the back of the protein. The ligand exchange between these cavities is facilitated by an additional site, which can be transiently occupied by the ligand in NP7, although it is absent in NP4. These features provide a basis to explain the enhanced internal gas hosting capacity found experimentally in NP7 and the absence of ligand rebinding from secondary sites in NP4. The current data allow us to speculate that the processes of docking to cell surfaces and NO release may be interconnected in NP7, thereby efficiently releasing NO into a target cell. To analyze the information provided about individual visual stimuliin the responses of single neurons in the primate temporal lobevisual cortex, neuronal responses to a set of 65 visual stimuli wererecorded in macaques performing a visual fixation task and analyzedusing information theoretical measures. The population of neuronsanalyzed responded primarily to faces. The stimuli included 23 facesand 42 nonface images of real-world scenes, so that the function ofthis brain region could be analyzed when it was processing relativelynatural scenes.It was found that for the majority of the neurons significantamounts of information were reflected about which of several of the23 faces had been seen. Thus the representation was not local, forin a local representation almost all the information available canbe obtained when the single stimulus to which the neuron respondsbest is shown. It is shown that the information available about anyone stimulus depended on how different (for example, how manystandard deviations) the response to that stimulus was from theaverage response to all stimuli. This was the case for responsesbelow the average response as well as above.It is shown that the fraction of information carried by the lowfiring rates of a cell was large—much larger than that carried bythe high firing rates. Part of the reason for this is that theprobability distribution of different firing rates is biased towardlow values (though with fewer very low values than would bepredicted by an exponential distribution). Another factor is thatthe variability of the response is large at intermediate and highfiring rates.Another finding is that at short sampling intervals (such as 20 ms)the neurons code information efficiently, by effectively acting asbinary variables and behaving less noisily than would be expectedof a Poisson process.  相似文献

The single and triple helical organization of protein chains strongly influences the mechanical properties of gelatin-based materials. A chemical method for obtaining different degrees of helical organization in gelatin is covalent functionalization, while a physical method for achieving the same goal is the variation of the drying conditions of gelatin solutions. Here we explored how the introduction of desaminotyrosine (DAT) and desaminotyrosyl tyrosine (DATT) linked to lysine residues of gelatin influenced the kinetics and thermodynamic equilibrium of the helicalization process of single and triple helices following different drying conditions. Drying at a temperature above the helix-to-coil transition temperature of gelatin (T > T(c), called v(short)) generally resulted in gelatins with relatively lower triple helical content (X(c,t) = 1-2%) than lower temperature drying (T < T(c), called v(long)) (X(c,t) = 8-10%), where the DAT(T) functional groups generally disrupted helix formation. While different helical contents affected the thermal transition temperatures only slightly, the mechanical properties were strongly affected for swollen hydrogels (E = 4-13 kPa for samples treated by v(long) and E = 120-700 kPa for samples treated by v(short)). This study shows that side group functionalization and different drying conditions are viable options to control the helicalization and macroscopic properties of gelatin-based materials.  相似文献
