The activities of plasma membrane associated sialidase Neu3, total β-glucosidase, CBE-sensitive β-glucosidase, non-lysosomal
β-glucosyl ceramidase GBA2, β-galactosidase, β-hexosaminidase and sphingomyelinase were determined at three different stages
of differentiation of murine neural stem cell cultures, corresponding to precursors, commited progenitors, and differentiated
cells. Cell immunostaining for specific markers of the differentiation process, performed after 7 days in culture in presence
of differentiating agents, clearly showed the presence of oligodendrocytes, astrocytes and neurons. Glial cells were the most
abundant. Sialidase Neu3 after a decrease from progenitors to precursors, showed an increase parallel to the differentiation
process. All the other glycosidases increased their activity along differentiation. The activity of CBE-sensitive β-glucosidase
and GBA2 were very similar at the precursor stage, but CBE-sensitive β-glucosidase increased 7 times while GBA2 only two in
the differentiated cells. In addition, we analysed also sphingomyelinase as enzyme specifically associated to sphingolipids.

Deregulation of the receptor tyrosine kinase c-Met has been implicated in several human cancers and is an attractive target for small molecule drug discovery. Herein, we report the discovery of a structurally diverse series of carbon-linked quinoline triazolopyridinones, which demonstrates nanomolar inhibition of c-Met kinase activity. This novel series of inhibitors exhibits favorable pharmacokinetics as well as potent inhibition of HGF-mediated c-Met phosphorylation in a mouse liver pharmacodynamic model. 相似文献

The shelf-life of fresh-cut tomatoes mainly depends on loss of tissue integrity and firmness that occurs also in intact fruits after long-term cold storage due to chilling injury. Round-fruit tomatoes (Solanum lycopersicum L.) cv. Jama were stored in 1.1-L plastic (polyethylene) fresh-cut produce containers as 10.0-mm-thick tomato slices and as intact tomatoes at 4 ± 0.5 °C. The aim of this work was to study the loss of membrane integrity and biochemical processes involved in membrane disruption. Electrolyte leakage and lipid peroxidation were studied at different stages of maturity: mature green, pink (PK), fully ripe and two different storage temperatures: 4 and 15 °C. The tomato slices of PK stage stored at 4 °C did not show changes for both parameters, while significant increase in membrane leakage and lipid peroxidation was observed at 15 °C, especially after 24 h of storage. The enzymes showed a simultaneous increase in their activities with a rise in electrolyte leakage and lipid peroxidation after 7 days of storage. Finally, phospholipase C (PLC) and phospholipase D (PLD) were investigated for intact fruit and tomato slices stored at 4 °C. The PLC had higher activity compared with PLD. In conclusion, the loss of membrane integrity in fresh-cut tomatoes is mainly affected by ripening stages, storage temperature and duration. The wounds enhance the PLC and PLD activities and they play a role late during storage. 相似文献

In this article, we review the relevant results obtained during almost 60 years of research on a specific aspect of stereochemistry, namely handedness preference and switches between right‐handed and left‐handed helical peptide structures generated by protein amino acids or appropriately designed, side‐chain modified analogs. In particular, we present and discuss here experimental and theoretical data on three categories of those screw‐sense issues: (i) right‐handed/left‐handed α‐helix transitions underwent by peptides rich in Asp, specific Asp β‐esters, and Asn; (ii) comparison of the preferred conformations adopted by helical host–guest peptide series, each characterized by an amino acid residue (e.g. Ile or its diastereomer aIle) endowed with two chiral centers in its chemical structure; and (iii) right‐handed (type I)/left‐handed (type II) poly‐(Pro)n helix transitions monitored for peptides rich in Pro itself or its analogs with a pyrrolidine ring substitution, particularly at the biologically important position 4. The unique modular and chiral properties of peptides, combined with their relatively easy synthesis, the chance to shape them into the desired conformation, and the enormous chemical diversity of their coded and non‐coded α‐amino acid building blocks, offer a huge opportunity to structural chemists for applications to bioscience and nanoscience problems. Satellite‐derived indices of photosynthetic activity are the primary data source used to study changes in global vegetation productivity over recent decades. Creating coherent, long‐term records of vegetation activity from legacy satellite data sets requires addressing many factors that introduce uncertainties into vegetation index time series. We compared long‐term changes in vegetation productivity at high northern latitudes (>50°N), estimated as trends in growing season NDVI derived from the most widely used global NDVI data sets. The comparison included the AVHRR‐based GIMMS‐NDVI version G (GIMMSg) series, and its recent successor version 3g (GIMMS3g), as well as the shorter NDVI records generated from the more modern sensors, SeaWiFS, SPOT‐VGT, and MODIS. The data sets from the latter two sensors were provided in a form that reduces the effects of surface reflectance associated with solar and view angles. Our analysis revealed large geographic areas, totaling 40% of the study area, where all data sets indicated similar changes in vegetation productivity over their common temporal record, as well as areas where data sets showed conflicting patterns. The newer, GIMMS3g data set showed statistically significant (α = 0.05) increases in vegetation productivity (greening) in over 15% of the study area, not seen in its predecessor (GIMMSg), whereas the reverse was rare (<3%). The latter has implications for earlier reports on changes in vegetation activity based on GIMMSg, particularly in Eurasia where greening is especially pronounced in the GIMMS3g data. Our findings highlight both critical uncertainties and areas of confidence in the assessment of ecosystem‐response to climate change using satellite‐derived indices of photosynthetic activity. Broader efforts are required to evaluate NDVI time series against field measurements of vegetation growth, primary productivity, recruitment, mortality, and other biological processes in order to better understand ecosystem responses to environmental change over large areas. 相似文献

Synaptic transmission and plasticity mediated by NMDA receptors (NMDARs) could modulate the severity of multiple sclerosis (MS). Here the role of NMDARs in MS was first explored in 691 subjects carrying specific allelic variants of the NR1 subunit gene or of the NR2B subunit gene of this glutamate receptor. The analysis was replicated for significant SNPs in an independent sample of 1548 MS subjects. The C allele of rs4880213 was found to be associated with reduced NMDAR-mediated cortical excitability, and with increased probability of having more disability than the CT/TT MS subjects. MS severity was higher in the CC group among relapsing-remitting MS (RR-MS) patients, while primary progressive MS (PP-MS) subjects homozygous for the T allele had more pronounced clinical worsening. Mean time to first relapse, but not to an active MRI scan, was lower in the CC group of RR-MS patients, and the number of subjects with two or more clinical relapses in the first two years of the disease was higher in CC compared to CT/TT group. Furthermore, the percentage of relapses associated with residual disability was lower in subjects carrying the T allele. Lesion load at the MRI was conversely unaffected by the C or T allele of this SNP in RR-MS patients. Axonal and neuronal degeneration at the optical coherence tomography was more severe in the TT group of PP-MS patients, while reduced retinal nerve fiber thickness had less consequences on visual acuity in RR-MS patients bearing the T allele. Finally, the T allele was associated with preserved cognitive abilities at the Rao’s brief repeatable neuropsychological battery in RR-MS. Signaling through glutamate NMDARs enhances both compensatory synaptic plasticity and excitotoxic neurodegeneration, impacting in opposite ways on RR-MS and PP-MS pathophysiological mechanisms. 相似文献

The first evidence that plants represent a valid, safe and cost-effective alternative to traditional expression systems for
large-scale production of antigens and antibodies was described more than 10 years ago. Since then, considerable improvements
have been made to increase the yield of plant-produced proteins. These include the use of signal sequences to target proteins
to different cellular compartments, plastid transformation to achieve high transgene dosage, codon usage optimization to boost
gene expression, and protein fusions to improve recombinant protein stability and accumulation. Thus, several HIV/SIV antigens
and neutralizing anti-HIV antibodies have recently been successfully expressed in plants by stable nuclear or plastid transformation,
and by transient expression systems based on plant virus vectors or Agrobacterium-mediated infection. The current article gives an overview of plant expressed HIV antigens and antibodies and provides an
account of the use of different strategies aimed at increasing the expression of the accessory multifunctional HIV-1 Nef protein

The Polynucleobacter-Euplotes association is an obligatory symbiotic system between a monophyletic group of ciliate species belonging to the genus Euplotes and bacteria of the species Polynucleobacter necessarius (Betaproteobacteria). Both organisms are unable to survive independently. Several studies revealed the existence of free-living populations of Polynucleobacter bacteria which are phylogenetically closely related to the endosymbiotic ones, but never share associations with Euplotes in the natural environment. Hence, following the most parsimonious explanation on the origin of the association, this symbiosis should represent a synapomorphic character for the hosts' clade. Nevertheless, phylogenetic analyses performed on an increased number of strains here presented suggest that Euplotes species, during their evolution, recruited Polynucleobacter bacteria as symbionts more than once. Moreover, in three cases, we observed different bacteria as obligate symbionts. These symbionts are the first characterized representatives of a phylogenetic lineage branching in a basal position with respect to the genus Polynucleobacter. The hypothesis that the original obligate symbionts belonged to this newly discovered clade and that, only subsequently, in most cases they have been replaced by Polynucleobacter bacteria recruited from the environment is proposed and discussed. The evolutionary path of this association seems anyway to have been more complex than so far supposed. 相似文献
