CFTR is a monomer: biochemical and functional evidence 总被引:3,自引:0,他引:3
Although the CFTR protein alone is sufficient to generate a regulated chloride channel, it is unknown how many of the polypeptides form the channel. Using biochemical and functional assays, we demonstrate that the CFTR polypeptide is a monomer. CFTR sediments as a monomer in a linear, continuous sucrose gradient. Cells co-expressing different epitope-tagged CFTR provide no evidence of co-assembly in immunoprecipitation and nickel affinity binding experiments. Co-expressed wild-type and DF508 CFTR are without influence on each other in their ability to progress through the secretory pathway, suggesting they do not associate in the endoplasmic reticulum. No hybrid conducting single channels are seen in planar lipid bilayers with which membrane vesicles from cells co-expressing similar amounts of two different CFTR conduction species have been fused. 相似文献

The interactions between an oligomeric heat-shock protein, alpha-crystallin, and its individual subunits with unfolded proteins were monitored by surface plasmon resonance. Immobilization at the sensor chip allowed us for the first time to study isolated alpha-crystallin subunits under physiological conditions. We observe that these subunits, in contrast to alpha-crystallin oligomers, do not bind unfolded protein. Our data indicate that quaternary structure of alpha-crystallin is necessary for its chaperone-like activity. 相似文献

After phosphorylation by protein kinase A, gating of the cystic fibrosis transmembrane conductance regulator (CFTR) chloride channel is regulated by the interaction of ATP with its nucleotide binding domains (NBDs). Models of this gating regulation have proposed that ATP hydrolysis at NBD1 and NBD2 may drive channel opening and closing, respectively (reviewed in Nagel, G. (1999) Biochim. Biophys. Acta 1461, 263-274). However, as yet there has been little biochemical confirmation of the predictions of these models. We have employed photoaffinity labeling with 8-azido-ATP, which supports channel gating as effectively as ATP to evaluate interactions with each NBD in intact membrane-bound CFTR. Mutagenesis of Walker A lysine residues crucial for azido-ATP hydrolysis to generate the azido-ADP that is trapped by vanadate indicated a greater role of NBD1 than NBD2. Separation of the domains by limited trypsin digestion and enrichment by immunoprecipitation confirmed greater and more stable nucleotide trapping at NBD1. This asymmetry of the two domains in interactions with nucleotides was reflected most emphatically in the response to the nonhydrolyzable ATP analogue, 5'-adenylyl-beta,gamma-imidodiphosphate (AMP-PNP), which in the gating models was proposed to bind with high affinity to NBD2 causing inhibition of ATP hydrolysis there postulated to drive channel closing. Instead we found a strong competitive inhibition of nucleotide hydrolysis and trapping at NBD1 and a simultaneous enhancement at NBD2. This argues strongly that AMP-PNP does not inhibit ATP hydrolysis at NBD2 and thereby questions the relevance of hydrolysis at that domain to channel closing. 相似文献

The development of defense reaction was studied in the wildlife and experimentally in 7 broods of altricial pied flycatcher (Ficedula hypoleuca) nestlings. Field studies demonstrated that passive-defense response first appeared on the 4th day of the nest life. It developed from the cessation of begging in young relatively satiated nestlings to characteristic freezing response independent of the level of feeding motivation in older nestlings. Older nestlings also acquire the defense reaction in response to novel visual stimuli. The efficiency of the natural stimulus for defense behavior (species-specific alarm call) nongradually changes during the nest life attaining the 100% level only on the 11th posthatching day. During the initial phase of defense behavior development, the reaction can be induced by different rhythmically organized stimuli. Later it becomes considerably more selective and other rhythmic and acoustic signals become much less effective than the alarm call. 相似文献

Data are presented on the distribution of centromeric heterochromatin of the human X-chromosome in the interphase nucleus of a moving cell. The in situ hybridization made it possible to obtain some results leading to the following conclusions: in moving fibroblasts centromeric heterochromatin of the X-chromosome is located in end regions of the interphase nucleus; there was no preferential localization noted of the centromeric region of the X-chromosome in the front or back areas of the nucleus as to the direction of the movement. 相似文献

Mössbauer spectroscopy was used to investigate the distribution of iron in rat organs and its localisation in liver subcellular fractions. A 57Fe-sucrose complex solution was injected by 0.5 ml doses into tail veins of anmals every day, during a 6-day period. Mössbauer spectra were measured in spleen, blood, liver and liver subcellular fractions. The Mössbauer spectrum of a spleen sample has two symmetrical doublets, one with δ=0.6 mm/s and Δ=0.7 mm/s, and the other with δ=1.0 mm/s and Δ=2.35 mm/s. The Mössbauer spectrum of blood has parameters which are close to those for carboxyhemoglobin and oxyhemoglobin complexes. After the addition of sodium citrate, the proportion of the carboxyhemoglobin complexes increases. The Mössbauer spectrum of liver has a two-component pattern with two symmetrical doublets, the first with δ=0.6 mm/s and Δ=0.63 mm/s and the second with δ=1.4 mm/s and Δ=3.45 mm/s. The first component, which was identified as ferritin, is present in all subcellular fractions (800 × gav sediment fraction, mitochondrial/lysosomal, microsomal and supernatant fractions), with its content in microsomal fraction. After the addition of NaBH4 to mitochondrial/lysosomal fraction, about 20 % of the iron contained in ferritin was reduced. In the Mössbauer spectrum this is reflected by an appearance of a doublet with δ=0.85 mm/s and Δ=3.7 mm/s. 相似文献

The influence of asynchronous actuation of the Angara-5-1 facility modules on the implosion symmetry of the wire array plasma in the Z-pinch mode is studied. It is shown that an increase in the r.m.s. jitter in the start times of the modules leads to an appreciable azimuthal asymmetry of magnetic field penetration inside the wire array and, as a consequence, to an increase in the duration of the soft X-ray pulse, a decrease in its power, and a shift of the Z-pinch with respect to the array axis. Necessary conditions for axisymmetric pinch implosion are determined. Experimental data on the magnitude and azimuthal distribution of the current (magnetic field) inside the wire array were obtained from magnetic probe measurements. The position of the Z-pinch with respect to the wire array axis was determined from two-dimensional X-ray images and radial optical streak images. 相似文献
