The IVS9-675C > A polymorphism of the HIF-1alpha gene was analyzed in patients with acute ischemic stroke and in a control group. The genotype and allele frequencies proved to significantly differ between the two groups. Nonsymmetrical polymethine dyes are shown to be applied as a new approach in the studies of phospholipid membrane microviscosity. The method requires determination of the intensity ratio for the long-wave length (Ig) and short-wave length (Ik) bands of fluorescence spectra in the region of 730-770 nm at exitation 600 nm. It allows determination of microviscosity by comparative measurements of the fluorescence parameter Ig/Ik in the model medium of the known viscosity (glycerol) and the object under study. Microviscosity in egg phosphatidylcholine vesicules (liposomes) is found to be 0.6-1.2 P. It depends on the surface curvature (size of vesicle), cholesterol content and temperature. It the studies of dimiristoylphosphatidyl choline liposomes the changes in microviscosity at the phase transition temperature are shown to be from 4.5 to 1.1 P. The transition temperature is 24.5 degrees C, the transition range being 2.2 degrees C. The results of this work demonstrate the advantages of the suggested approach to study mobility in phospholipid membranes and confirm it to be promising to study natural membranes and whole cells.  相似文献

Melanin synthesis affects melanoma behavior and tumor responsiveness to therapy; therefore, we investigated metabolic changes in melanoma cells after induction of melanogenesis. Amelanotic and melanotic melanoma cells were labeled with 13C precursors and changes in their metabolism was analyzed by high-resolution magic angle spinning (HRMAS) nuclear magnetic resonance (NMR). HRMAS NMR demonstrated clear differences in the pattern of metabolic intermediates between amelanotic and melanotic cells. Although the exact nature of the metabolites requires further investigations, our comparative studies clearly show that induction of melanogenesis is associated with changes of glucose and sodium acetate metabolism, demonstrating HRMAS NMR as a powerful and noninvasive technique to investigate such process.  相似文献

Adaptive deep brain stimulation (aDBS) has the potential to improve the treatment of Parkinson''s disease by optimizing stimulation in real time according to fluctuating disease and medication state. In the present realization of adaptive DBS we record and stimulate from the DBS electrodes implanted in the subthalamic nucleus of patients with Parkinson''s disease in the early post-operative period. Local field potentials are analogue filtered between 3 and 47 Hz before being passed to a data acquisition unit where they are digitally filtered again around the patient specific beta peak, rectified and smoothed to give an online reading of the beta amplitude. A threshold for beta amplitude is set heuristically, which, if crossed, passes a trigger signal to the stimulator. The stimulator then ramps up stimulation to a pre-determined clinically effective voltage over 250 msec and continues to stimulate until the beta amplitude again falls down below threshold. Stimulation continues in this manner with brief episodes of ramped DBS during periods of heightened beta power.Clinical efficacy is assessed after a minimum period of stabilization (5 min) through the unblinded and blinded video assessment of motor function using a selection of scores from the Unified Parkinson''s Rating Scale (UPDRS). Recent work has demonstrated a reduction in power consumption with aDBS as well as an improvement in clinical scores compared to conventional DBS. Chronic aDBS could now be trialed in Parkinsonism.  相似文献

We measured the thermoregulatory characteristics of a common, tropical murid rodent, Sundamys muelleri. Subcutaneous body temperatures and evaporative water loss increased at ambient temperatures above 33°C, but no increases in metabolic rate were observed as high as 38.2°C, indicating tropical rodents may be resilient as climate change progresses.
