Advanced scientific knowledge on arbuscular mycorrhizal symbioses recently enhanced potential for implementation of mycorrhizal biotechnology in horticulture and agriculture plant production, landscaping, phytoremediation and other segments of the plant market. The advances consist in significant findings regarding:—new molecular detection tools for tracing inoculated fungi in the field;—the coexistence mechanisms of various fungi in the single root system;—new knowledge on in vitro physiology of the AM fungi grown in root organ cultures;—mechanisms of synergistic interactions with other microbes like PGPR or saprotrophic fungi; discovery of mycorrhiza supportive compounds such as strigolactones. Scientific knowledge has been followed by technological developments like novel formulations for liquid applications or seed coating, mycorrhiza stimulating compounds or new application modes. Still the missing components of biotechnology are appropriate, cheap, highly reproducible and effective methods for inocula purity testing and quality control. Also there is a weak traceability of the origin of the mycorrhizal fungi strains used in commercial inocula. Numerous poor quality products can still be found on the markets claiming effective formation mycorrhiza which have very low capacity to do so. These products usually rely in their effects on plant growth not on support of host plants via formation of effective mycorrhizal symbiosis but on fertilizing compounds added to products. There is growing number of enterprises producing mycorrhiza based inocula recently not only in developed world but increasingly in emerging markets. Also collaboration between private sector and scientific community has an improving trend as the development of private sector can fuel further research activities. Last but not least there is apparent growing pull of the market and increasing tendency of reduction of agrochemical inputs and employment of alternative strategies in planting and plant production. These circumstances support further developments of mycorrhizal inocula production and applications and maturation of the industry.  相似文献

Each of the three plasma membrane Ato proteins is involved in ammonium signalling and the development of yeast colonies. This suggests that although these proteins are homologous, they do not functionally substitute for each other, but may form a functional complex. Here, we present a detailed combined FRET, FLIM and photobleaching study, which enabled us to detect interactions between Ato proteins found in distinct compartments of yeast cells. We thus show that the proteins Ato1p and Ato2p interact and can form complexes when present in the plasma membrane. No interaction was detected between Ato1p and Ato3p or Ato2p and Ato3p. In addition, using specially prepared strains, we were able to detect an interaction between molecules of the same Ato protein, namely Ato1p-Ato1p and Ato3p-Ato3p, but not Ato2p-Ato2p.  相似文献

Regulated exocytosis is a process that strongly depends on the formation and stability of the fusion pore. It was indicated experimentally and theoretically that narrow and highly curved fusion pore may be stabilized by accumulation of anisotropic membrane components possessing orientational ordering. On the other hand, narrow fusion pore may also undergo repetitive opening and closing, disruption in the so called kiss and run process or become completely opened in the process of full fusion of the vesicle with the membrane. In this paper we attempt to elucidate the subtle interplay between the stabilizing and destabilizing processes in the fusion neck. A possible physical mechanism which may lead to disruption of the stable fusion pore or complete fusion of the vesicle with the membrane is discussed. It is indicated that topologically driven defects of the in-plane orientational membrane ordering in the region of the fusion pore may disrupt the fusion. Alternatively, it may facilitate repetitive opening and closing of the fusion pore or induce full fusion of the vesicle with the target membrane.  相似文献

Fucose-containing sulfated polysaccharides can be extracted from the brown seaweed, Sargassum sp. It has been reported that fucose-rich sulfated polysaccharides from brown seaweeds exert different beneficial biological activities including anti-inflammatory, anticoagulant, and anti-viral effects. Classical extraction of fucose-containing sulfated polysaccharides from brown seaweed species typically involves extended, multiple-step, hot acid, or CaCl2 treatments, each step lasting several hours. In this work, we systematically examined the influence of acid concentration (HCl), time, and temperature on the yield of fucose-containing sulfated polysaccharides (FCSPs) in statistically designed two-step and single-step multifactorial extraction experiments. All extraction factors had significant effects on the fucose-containing sulfated polysaccharides yield, with the temperature and time exerting positive effects, and the acid concentration having a negative effect. The model defined an optimized single-step FCSPs extraction procedure for Sargassum sp. (a brown seaweed). A maximal fucose-containing sulfated polysaccharides yield of ~7% of the Sargassum sp. dry matter was achieved by the optimal extraction procedure of: 0.03?M HCl, 90°C, 4?h. HPAEC-PAD analysis confirmed that fucose, galactose, and glucuronic acid were the major constituents of the polysaccharides obtained by the optimized method. Lower polysaccharide yield, but relatively higher fucose content was obtained with shorter extraction time. The data also revealed that classical multi-step extraction with acid ≥0.2?M HCl at elevated temperature and extended time had a detrimental effect on the FCSPs yield as this treatment apparently disrupted the structural integrity of the polymer and evidently caused degradation of the carbohydrate chains built up of fucose residues.  相似文献

Complex aquatic systems of karst harbour a rich but little‐investigated biodiversity. In Croatia and Bosnia–Herzegovina karst, temporal springs are inhabited by a group of minnow‐like fishes that retreat to the associated ground water during dry seasons and spend several months underground. The most abundant species in this group is Delminichthys adspersus (Heckel 1843), which also has the most fragmented distribution range. To determine the population composition and dispersal patterns, and to detect potential underground migration, a large genetic data set comprising 544 specimens of D. adspersus covering most of its distribution area was analysed. Analysis of mitochondrial cytochrome b sequences (~1000 bp) and eight microsatellite loci showed that D. adspersus comprises at least three subpopulations with gene flow occurring among them. Coalescent‐based analysis revealed a complex migration pattern, with several unidirectional dispersal paths, including between temporal springs that share no surface connection. The results of this study suggest the existence of recurrent underground migration of fish in a karst environment and demonstrate the complexity of its hydrological network. The findings are relevant to conservation strategies for endemic karst organisms and karst ecosystems as a whole.  相似文献

Habitat loss and degradation can be considered as major threats to freshwater invertebrates. These often irreversible processes lead to reduction of habitat patch quality and cause local extinctions of dragonflies, notably of habitat specialists. However, the biodiversity of specific secondary habitats is very high. Here, we present findings from a 10-year study that intensively monitored odonate fauna in the Upper Silesian industrial coal region having many secondary habitats characterized by very frequent disturbances due to soil instability. We evaluated qualitative changes in the dragonfly assemblages on 10 patches using a modified dragonfly biotic index. Data analysis was supplemented by a model examining population dynamics of the threatened dragonfly Leucorrhinia pectoralis, using the capture-mark-recapture method, as an effective indicator of habitat quality. We show that dynamics of environmental conditions in secondary habitats are reflected in population dynamics of dragonfly populations and assemblages. As frequency of L. pectoralis population extinctions within the patch is considerable and independent of size and spatial isolation of single habitats, these can be regarded as ecological traps. Nevertheless, the metapopulation dynamics may be a key adaptation of dragonflies to frequent freshwater habitat disturbances. We suggest that local extinctions are effectively balanced with (re-)colonization of newly emerging freshwater habitats. These findings have implications for potential conservation management of specific human-made habitats, because secondary habitats with a great diversity of succession stages arising directly as a consequence of environmental instability may be considered as partial alternatives to natural habitats in cultural landscapes.  相似文献

Phragmites australis and Glyceria maxima are fast-growing littoral grasses often competing for similar wetland habitats. Eutrophication affects their competitiveness, but the outcome is not generally predictable due to the complexity of interrelated factors. We hypotheses that pore water N:P and NH4 +:NO3 ? modify their growth and metabolic responses to the trophic status of the habitat. The hypothesis was tested under standardized conditions of long-term sand cultures. Application of N?+?P up to extreme levels in combination with N:P?<?10 and NH4 +:NO3 ??<?1 triggered positive growth response in both species. In contrast, similar N levels applied in N:P?>?90 and NH4 +:NO3 ??=?4 caused lower productivity, changes in resource allocation, morphology and metabolic relations (e.g. high shoot density, low shoot diameters and heights, reduced root and rhizome growth). Observed signs of stress resembled the factors associated with the reed retreat at the die-back sites. Unbalanced N levels obviously alter plant susceptibility to stresses (altering, e.g. ventilation efficiency, plant anchorage or below-ground storage capacity). The positive effect of sufficient P supply was pronounced in Glyceria. It might therefore favour Glyceria in competition with Phragmites at highly fertile habitats rich in P.  相似文献

The anaerobic dehalogenation of organohalides is catalyzed by the reductive dehalogenase (RdhA) enzymes produced in phylogenetically diverse bacteria. These enzymes contain a cobamide cofactor at the active site and two iron-sulfur clusters. In this study, the tetrachloroethene (PCE) reductive dehalogenase (PceA) of the Gram-positive Desulfitobacterium hafniense strain Y51 was produced in a catalytically active form in the nondechlorinating, cobamide-producing bacterium Shimwellia blattae (ATCC 33430), a Gram-negative gammaproteobacterium. The formation of recombinant catalytically active PceA enzyme was significantly enhanced when its dedicated PceT chaperone was coproduced and when 5,6-dimethylbenzimidazole and hydroxocobalamin were added to the S. blattae cultures. The experiments were extended to D. hafniense DCB-2, a reductively dehalogenating bacterium harboring multiple rdhA genes. To elucidate the substrate spectrum of the rdhA3 gene product of this organism, the recombinant enzyme was tested for the conversion of different dichlorophenols (DCP) in crude extracts of an RdhA3-producing S. blattae strain. 3,5-DCP, 2,3-DCP, and 2,4-DCP, but not 2,6-DCP and 3,4-DCP, were reductively dechlorinated by the recombinant RdhA3. In addition, this enzyme dechlorinated PCE to trichloroethene at low rates.  相似文献
