The structural basis of the TIM10 chaperone assembly   总被引:5,自引:0,他引:5  
Tim9 and Tim10 are essential components of the "small Tim" family of proteins that facilitate insertion of polytopic proteins at the inner mitochondrial membrane. The small Tims are themselves imported from the cytosol and are organized in specific translocation assemblies in the intermembrane space. Their conformational properties and how these influence the mechanism of assembly remain poorly understood. Moreover, the three-dimensional structure of the TIM10 complex is unknown. We have characterized the structural properties of these proteins in their free and assembled states using NMR, circular dichroism, and small angle x-ray scattering. We show that the free proteins are largely unfolded in their reduced assembly-incompetent state and molten globules in their oxidized assembly-competent state. Tim10 appears less structured than Tim9 in their respective free oxidized forms and undergoes a larger structural change than Tim9 upon complexation. The NMR data here demonstrates unequivocally that only the oxidized states of the Tim9 and Tim10 proteins are capable of forming a complex. Zinc binding stabilizes the reduced state against proteolysis without significantly affecting the secondary structure. Solution x-ray scattering was used to obtain a molecular envelope for the subunits individually and for their fully functional TIM10 complex. Ab initio shape reconstructions based on the scattering data has allowed us to obtain the first low resolution three-dimensional structure of the TIM10 complex. This is a novel structure that displays extensive surface hydrophobicity. The structure also provides an explanation for the escorting function of this non-ATP-powered chaperone particle.  相似文献

The bot fly Cuterebra austeni exhibits year-to-year consistency in its landmark-based hilltopping mating system. Flies were found on some of the same localized ridgetop perching areas that were known to be mate encounter sites 22 years previously. In both 1980 and 2002, males were drawn preferentially to locally elevated, largely vegetation-free sites. In addition, a substantial proportion of individuals exhibited site fidelity in both years, as demonstrated by their willingness to return to the site where marked after being captured and handled. In both years of observation, a smaller proportion returned the second day after marking and only 10–20% of marked males were seen on the third day. Thus males apparently have only a very limited window of opportunity to secure a given perching territory. In both years, a number of males (residents) succeeded in monopolizing a perching area over a substantial part of the brief morning flight period. One unexplained difference across the decades: residents appeared on a lower proportion of days of observation in 2002 than in 1980.  相似文献

Adaptationist analyses of animal contests have contributed much to our understanding of behavioral evolution. One class of contest, however, the war of attrition, has proven difficult to interpret. In wars of attrition involving aerial displays, there is evidence that asymmetries in performance parameters such as flight energetics may be important determinants of contest resolution. This paradigm is not universal, however, and we presently lack a framework for understanding why certain biophysical parameters are important only in some cases. One possibility is that the relevance of these parameters is determined by evolutionarily conserved life-history-scale patterns of resource allocation and acquisition. We evaluated this hypothesis by investigating the correlates of competitive success in two territorial insects that exemplify markedly different lifetime patterns of resource utilization. We found that in the bot fly Cuterebra austeni, an extreme capital breeding species that depends entirely on energy acquired during its immature stages, territorial residency was most strongly correlated with a size-independent measure of energetic availability. In contrast, residency in the tarantula hawk wasp Hemipepsis ustulata was best predicted by variation in body size per se. Adult H. ustulata are able to supplement their larval-derived nutrient capital in the manner of an income breeder, and fuel reserves were independent of age and actually correlated negatively with residency in this species. These results underscore how the study of sexually selected phenomena may be enriched by an explicit consideration of life-history principles.  相似文献

In central Arizona, receptive females of the pipevine swallowtail, Battus philenor, are widely scattered in time and space, and in this region the butterfly's mating system is one in which males patrol mountain peaks. Hilltopping males engage intruding males in ascending flights that appear to be ritualized aerial combat with individuals defending patrolling sites for relatively short periods on any given day. The day-to-day appearance of marked males is irregular, unlike the site fidelity shown by males of many other hilltopping insects. The distinctive pattern of male territoriality in B. philenor may be partly a response to very low male and female density in the observed population. Males at the hilltop chase, court, and attempt to copulate with virgin females released near them. Males assess the receptivity of females rapidly, and receptive females permit a lengthy copulation to occur after a courtship that lasts less than 30 s. During an initial copulation males pass a spermatophore that weighs about 6% of their body mass, with partners on following days receiving a smaller but still substantial donation.  相似文献

Large numbers of males of the bee Centris pallida Fox have been observed patrolling areas in which females are emerging. Males locate specific sites at which a buried bee is about to emerge and dig down to meet the other individual, male or female. If it is a female, mating is initiated when she scrambles into the excavation pit created by the male. Males fight intensely with one another for possession of digging sites and for unburied virgin females. Experiments indicate that males locate conspecifics beneath the surface on the basis of extremely non-specific olfactory cues; they are capable of locating buried honey bees and other insects. The evolution of digging behaviour is traced to selection favouring males that are first to reach a virgin female which will mate just once in her lifetime. A number of examples are given of other insects that have evolved similar abilities, apparently in response to similar selection pressures.  相似文献

We have studied the effects of iontophoretic injection of the quaternary ammonium compounds tetraethylammonium (TEA) and tetrabutylammonium (TBA) in cardiac purkinje fibers. We find that TBA(+) is a more effective blocker than TEA(+), but injection of either compound reduces the time-dependent outward plateau currents, transient outward current (I(to)), and the delayed rectifier (I(x)). Our findings provide evidence that these outward cardiac currents are carried by channels that in some respects are pharmacologically similar to squid axon potassium channels. We demonstrate that this procedure is a new tool that can be useful in the analysis of membrane currents in the heart.  相似文献

Inductively coupled plasma-mass spectrometry (ICP-MS) is a powerful tool for both quantitative multielement analyses of inorganic elements and measurement of isotope ratios (IRs). The main disadvantage of this technique is the existence of polyatomic isobaric interferences at some key masses. Zinc has been investigated for such potential interferences in serum or plasma. The Zn isotopes,66Zn and68Zn, have no apparent interferences, but32S16O2 and32S2 are isobaric with64Zn. The possible effects of S and other major components of blood plasma—Na, K, Cl, P, Ca—on Zn IRs were investigated using a series of mineral solutions which simulated human plasma with respect to these elements. The mixture of all mineral elements interfered only with64Zn (6.66 ng/mL) and70Zn (8.51 ng/mL). Interferences to66Zn,67Zn, and68Zn were minimal containing 0.90, 0.94, and 0.39 ng/mL, respectively. The copresence of Na or S shifted35Cl16O2 (atomic mass 67 coming from Cl solution) to35Cl2 which reduced the contribution to67Zn. The hypothesis that Zn IRs obtained from plasma at various intervals after the intravenous administration of enriched67Zn to humans would reflect those obtained after extraction of Zn was therefore tested. To compare the two pretreatment methods, “extraction” versus “nonextraction,” specimens were collected from 10 human subjects at intervals of 5 min to 24 h postinjection, and in 4 subjects from 5 min to 9 d postinjection. Two separate aliquots of plasma from each time-point were dried and digested with hydrogen peroxide, and the residue dissolved in nitric acid. One specimen was subjected to zinc extraction using ammonium diethyldithiocarbamate chelate followed by back extraction into nitric acid. The matching aliquot received no further pretreatment. The normalized IRs obtained from67Zn/66Zn and67Zn/68Zn in both the “extracted” and “nonextracted” samples agreed well(r 2 = 0.976 andr 2 = 0.985, respectively) compared to those from other ratios (r 2 = 0.838 for67Zn/64Zn andr 2 = 0.747 for67Zn/70Zn). Considering the minimum possibility of isobaric interferences in plasma samples,67Zn/68Zn obtained from “nonextracted” samples is sufficient for routine Zn kinetic analysis by ICP-MS.  相似文献
