Any immunological response is the end result of the equilibrium between many positive and negative regulatory factors. It has been recently demonstrated that histamine receptor-bearing T lymphocytes could play a role in this regulation. This work aims to study the effects of different cell populations after incubation with histamine on the proliferative response of normal lymphocytes. The histamine-incubated peripheral blood lymphocytes (PBL) lower the proliferative response of normal cells toward mitogens (phytohemagglutinin, concanavalin A) and antigens (mixed lymphocyte culture). In order to precise the cell subpopulations involved in this suppression, PBL have been depleted of adherent cells and B and T lymphocytes have been purified by a standard rosette technique. The enriched B cells do not suppress the normal response but the suppressor activity of T cells, as well as adherent cell-depleted PBL, are significantly reduced compared to the one of PBL. The initial suppressor activity is restored by addition of 1% adherent cells (and not 5 or 10%) to adherent cell-depleted lymphocytes and 10% adherent cells (not 1 or 5%) to T-enriched population. These observations suggest a role for adherent cells in this regulation.  相似文献

The properties of cyclic nucleotide phosphodiesterase were studied in soluble and particulate fractions from the central nervous system of Manduca sexta (Lepidoptera: Sphingidae). It was determined that: (1) The highest levels of phosphodiesterase occur in nervous tissue. (2) The total and specific enzyme activities of larval and adult brains are greater than those of the remaining ganglia. (3) Specific central nervous sy stem phosphodiesterase activities of the adult are lower than those of the larva, but both protein and total phosphodiesterase contents are considerably greater in the adult central nervous system. (4) Mg2+ is not absolutely required for either cyclic AMP-phosphodiesterase or cyclic GMP-phosphodiesterase activity. (5) Phosphodiesterase is inhibited by a variety of physiological and non-physiological compounds, nucleoside triphosphates being particularly effective; Some potent inhibitors of mammalian phosphodiesterase are comparatively ineffective toward Manduca sexta phosphodiesterase. (6) Kinetic analyses of soluble and particulate phosphodiesterase revealed non-linear double-reciprocal plots for the hydrolysis of both cyclic AMP and cyclic GMP, with Michaelis constants of approximately 10 mu M and 20 mu M; (7) The hydrolysis of both cyclic nucleotides appears in part to be the function of a single enzyme or related enzymes in the insect central nervous system. It follows that the intracellular level of one cyclic nucleotide may influence the concentration of the other by inhibiting its DEGRADATION.  相似文献

Wistar rats were submitted to portacaval anastomosis (PCA). Control rats were sham-operated and pair-fed (SOPF). After 3 weeks, PCA led to the hypertrophy of right atrium (+50%), left atrium (+67%) and both ventricles (+26%). The response of adenylate cyclase activity to secretin was specifically and markedly decreased in membranes from atria (-51 to 59%) and ventricles (-68 to 69%). These data suggest a decrease in the number of functional secretin receptors in heart considering that: the half-maximal stimulatory secretin concentration was unchanged; glucagon stimulations were unaltered and D,L-isoproterenol stimulations were hardly affected; the Gpp(NH)p-, NaF-, and forskolin-stimulated adenylate cyclase activities were moderately decreased (in ventricles, by 14-28%) or unchanged (in atria).  相似文献

Professional workers and consumers frequently use hard-surface cleaning products and these products may contain glycol ethers (GEs), such as 2-Butoxyethanol (2-BE). Governmental agencies have set exposure limits for some chemicals used in cleaning products and these exposure limits have been used as guides to protect human health. The study objectives were to determine realistic inhalation exposures for professional workers performing multiple, sequential cleaning tasks and compare the exposures to the acute reference exposure level (REL), which California established for 2-BE. The ConsExpo model was acceptable for evaluating exposure based on a comparison of its predictions to experimentally measured 2-BE vapor concentrations from hard-surface cleaning. The typical worker exposure was predicted for a cleaning scenario consisting of three bathrooms and three kitchens (or three bedrooms) in a 1-h period. This exposure scenario would not be expected to result in significant health consequences because the predicted exposure was much lower than the REL. The predicted chronic and aggregate exposures were also acceptable. This analysis identified two important variables that affect inhalation exposure: cleaners should be used with adequate ventilation and wet wiping towels should be properly disposed so that they are not a source of continuing exposure.  相似文献

Among the identified risk factors of age-related macular degeneration, sunlight is known to induce cumulative damage to the retina. A photosensitive derivative of the visual pigment, N-retinylidene-N-retinylethanolamine (A2E), may be involved in this phototoxicity. The high energy visible light between 380 nm and 500 nm (blue light) is incriminated. Our aim was to define the most toxic wavelengths in the blue-green range on an in vitro model of the disease. Primary cultures of porcine retinal pigment epithelium cells were incubated for 6 hours with different A2E concentrations and exposed for 18 hours to 10 nm illumination bands centered from 380 to 520 nm in 10 nm increments. Light irradiances were normalized with respect to the natural sunlight reaching the retina. Six hours after light exposure, cell viability, necrosis and apoptosis were assessed using the Apotox-Glo Triplex™ assay. Retinal pigment epithelium cells incubated with A2E displayed fluorescent bodies within the cytoplasm. Their absorption and emission spectra were similar to those of A2E. Exposure to 10 nm illumination bands induced a loss in cell viability with a dose dependence upon A2E concentrations. Irrespective of A2E concentration, the loss of cell viability was maximal for wavelengths from 415 to 455 nm. Cell viability decrease was correlated to an increase in cell apoptosis indicated by caspase-3/7 activities in the same spectral range. No light-elicited necrosis was measured as compared to control cells maintained in darkness. Our results defined the precise spectrum of light retinal toxicity in physiological irradiance conditions on an in vitro model of age-related macular degeneration. Surprisingly, a narrow bandwidth in blue light generated the greatest phototoxic risk to retinal pigment epithelium cells. This phototoxic spectrum may be advantageously valued in designing selective photoprotection ophthalmic filters, without disrupting essential visual and non-visual functions of the eye.  相似文献
