D-3 phosphorylated inositides are a peculiar class of lipids, synthesized by phosphatidylinositol 3-kinase (PtdIns 3-K), which are also present in the nucleus. In order to clarify a possible role for nuclear D-3 phosphorylated inositides during human erythroid differentiation, we have examined the issue of whether or not, in K562 human erythroleukemia cells, erythropoietin (EPO) may generate nuclear translocation of an active PtdIns 3-K. Immunoprecipitation with an anti-p85 regulatory subunit of PtdIns 3-K, revealed that both the intranuclear amount and the activity of the kinase increased rapidly and transiently in response to EPO. Enzyme translocation was blocked by the specific PtdIns 3-K pharmacological inhibitor, LY294002, which also inhibited erythroid differentiation. In vivo, intranuclear synthesis of phosphatidylinositol (3,4,5) trisphosphate (PtdIns (3,4,5)P(3)) was stimulated by EPO. Almost all PtdIns 3-K that translocated to the nucleus was highly phosphorylated on tyrosine residues of the p85 regulatory subunit. These findings strongly suggest that an important step in the signaling pathways that mediate EPO-induced erythroid differentiation may be represented by the intranuclear translocation of an active PtdIns 3-K.  相似文献

The enzymatic hydrolysis of wheat straw was carried out in bi-phasic media prepared with acetate esters and Na-acetate buffer. The volume percentage of the organic chemicals was 75%. The biomass was pretreated in a steam explosion plant at 217°C and for 3 min. A cellulase complex from commercial source was utilised and the experiments were run at 45°C and at constant enzyme to biomass weight ratio (0.06). Biomass loadings ranged from 6.25 to 100 g per litre of reactor. The amount of glucose formed per litre of reactor and hour and the glucose yield (grams of product per gram of biomass) were close to the values attained in pure buffer. The glucose concentration in the aqueous phase was in bi-phasic media much higher than in pure buffer and reached the value of 146 g lH2O−1 during 72 h of saccharification. The results were poorly dependent on the physical–chemical properties of the solvents. Nevertheless, butyl acetate could be slightly preferred to propyl and i-amyl acetate. The use of bi-phasic media did not require stirring rate higher than in pure buffer. The presence of acetate ester traces did not alter markedly the production of ethanol in the fermentation stage, but determined the extension of the lag phase.  相似文献

Nocardia globerula strain 432 was able to synthesize triacylglycerols (TAG) during cultivation on 2,6,10,14-tetramethyl pentadecane (pristane) under nitrogen-limiting conditions. Within these cells, 4,8,12-trimethyl tridecanoic acid was the major fatty acid detected. Fatty acids with an odd number of carbon atoms and minor amounts of even-numbered fatty acids were also observed. Experiments carried out with acrylic acid, an inhibitor of beta-oxidation, suggested that odd-numbered fatty acids such as C15:0, C17:0 and 10-methyl C17:0 were synthesized de novo using propionyl-CoA, the beta-oxidation product, as precursor. Although N. globerula 432 incorporated mainly straight chain fatty acids into TAG, the branched fatty acid 4,8,12-trimethyl tridecanoic acid also appeared, to some extent, in the acylglycerols. The importance of TAG biosynthesis by pristane-grown cells of N. globerula strain 432 is discussed.  相似文献

The synthesis and a preliminary biological characterization of a new class of N-benzyl-aminoalcohols which have serotonin (5-HT2) and dopamine (D2) receptor affinity is described. In vitro competition binding studies were conducted with the new molecules and 3H-spiperone on crude membrane preparation from rat striatum and frontal cortex. One of these compounds, 3-benzylamino-1-(4-fluoro-2-iodophenyl)-propan-1-ol (6f), whose IC50 values are in the micromolar range for both the D2 and 5-HT2 receptors, was prepared in iodine-125 labelled form (6i) by nucleophilic substitution of the bromine atom of 3-benzylamino-1-(2-bromo-4-fluorophenyl)-propan-1-ol (6d). In the in vivo studies, conducted on rats, the radiolabelled molecule 6i shows a good capacity to cross the blood–brain barrier (BBB) with a mean value of first pass cerebral extraction (E) of ca. 50% when the regional cerebral blood flow, measured with microsphere technique, is in the experimental animal's physiologic range (0.8–1 mL/min/g). A preliminary in vitro autoradiographic distribution on coronal rat brain slices of the radioiodinated molecule showed that it was preferentially localized in the striatum and in the cerebral regions rich in dopamine- and serotonin receptors, even if a high non-specific binding was observed.  相似文献

Simulation is used to facilitate new learning in a variety of situations. One application of simulation could be to help therapists gain therapeutic skills prior to seeing clients. This particular study was interested in measuring changes in stress response by looking at subjective and objective measures of distress (as measured by SUDS, HR, and HRV) over three sessions of simulated therapy. 16 second year psychology students participated in three sessions, and had their HR and HRV measured by Polar watches. Over the three sessions, there was a decrease in perceived distress, as measured by SUDS ratings. During and between sessions, there was inconclusive change in physiological parameters.  相似文献
