Liver hypoxia still represents an important cause of liver injury during shock and liver transplantation. We have investigated the protective effects of beta-alanine against hypoxic injury using isolated perfused rat livers and isolated rat hepatocyte suspensions. Perfusion with hypoxic Krebs-Henseleit buffer increased liver weight and caused a progressive release of lactate dehydrogenase (LDH) in the effluent perfusate. The addition of 5 mmol/l beta-alanine to the perfusion buffer completely prevented both weight increase and LDH leakage. These findings were confirmed by histological examinations showing that beta-alanine blocked the staining by trypan blue of either liver parenchymal and sinusoidal cells. Studies performed in isolated hepatocytes revealed that beta-alanine exerted its protective effects by interfering with Na+ accumulation induced by hypoxia. The addition of gamma-amino-butyric acid, which interfered with beta-alanine uptake by the hepatocytes or of Na+/H+ ionophore monensin, reverted beta-alanine protection in either hepatocyte suspensions or isolated perfused livers. We also observed that liver receiving beta-alanine were also protected against LDH leakage and weight increase caused by the perfusion with an hyposmotic (205 mosm) hypoxic buffer obtained by decreasing NaCl content from 118 to 60 mmol/l. This latter effect was not reverted by blocking K+ efflux from hepatocyte with BaCl(2) (1mmol/l). Altogether these results indicated that beta-alanine protected against hypoxic liver injury by preventing Na+ overload and by increasing liver resistance to osmotic stress consequent to the impairment of ion homeostasis during hypoxia.  相似文献

Beta-D-galactofuranosidase is a good chemotherapeutic target for the design of inhibitors, since beta-D-galactofuranose is a constituent of important parasite glycoconjugates but is not present in the host mammals. With this aim, we have synthesized for the first time alkyl, benzyl and aryl 1-thio-beta-D-galactofuranosides by condensation of penta-O-benzoyl-alpha,beta-D-galactofuranose with the corresponding thiols, in the presence of SnCl4as catalyst. The complete chemical and spectroscopical characterization of these compounds showed that the reaction was stereoselective. Debenzoylation with sodium methoxide afforded the beta-S-galactofuranosides in high yield. The thioglycosides were tested as inhibitors of the beta-D- galactofuranosidase of Penicillium fellutanum, using for the first time 4-nitrophenyl-beta-D-galactofuranoside as chromogenic substrate. The 4- aminophenyl-1-thio-beta-D-galactofuranoside, obtained by catalytic hydrogenation of the nitrophenyl derivative, was the best inhibitor being then an adequate ligand for the preparation of an affinity phase aimed at the isolation of beta-d-galactofuranosidases from different sources. Background

Morphological and functional differences of the right and left ventricle are apparent in the adult human heart. A differential contribution of cardiac fibroblasts and smooth muscle cells (populations of epicardium-derived cells) to each ventricle may account for part of the morphological-functional disparity. Here we studied the relation between epicardial derivatives and the development of compact ventricular myocardium.

Results

Wildtype and Wt1CreERT2/+ reporter mice were used to study WT-1 expressing cells, and Tcf21lacZ/+ reporter mice and PDGFRα-/-;Tcf21LacZ/+ mice to study the formation of the cardiac fibroblast population. After covering the heart, intramyocardial WT-1+ cells were first observed at the inner curvature, the right ventricular postero-lateral wall and left ventricular apical wall. Later, WT-1+ cells were present in the walls of both ventricles, but significantly more pronounced in the left ventricle. Tcf21-LacZ + cells followed the same distribution pattern as WT-1+ cells but at later stages, indicating a timing difference between these cell populations. Within the right ventricle, WT-1+ and Tcf21-lacZ+ cell distribution was more pronounced in the posterior inlet part. A gradual increase in myocardial wall thickness was observed early in the left ventricle and at later stages in the right ventricle. PDGFRα-/-;Tcf21LacZ/+ mice showed deficient epicardium, diminished number of Tcf21-LacZ + cells and reduced ventricular compaction.

Conclusions

During normal heart development, spatio-temporal differences in contribution of WT-1 and Tcf21-LacZ + cells to right versus left ventricular myocardium occur parallel to myocardial thickening. These findings may relate to lateralized differences in ventricular (patho)morphology in humans.  相似文献

Background  

Elucidation of the communal behavior of microbes in mixed species biofilms may have a major impact on understanding infectious diseases and for the therapeutics. Although, the structure and the properties of monospecies biofilms and their role in disease have been extensively studied during the last decade, the interactions within mixed biofilms consisting of bacteria and fungi such as Candida spp. have not been illustrated in depth. Hence, the aim of this study was to evaluate the interspecies interactions of Pseudomonas aeruginosa and six different species of Candida comprising C. albicans, C. glabrata, C. krusei, C. tropicalis, C. parapsilosis, and C. dubliniensis in dual species biofilm development.  相似文献

Although molluscs feature prominently in the semi-popular and academic literature on marine biodiversity, field surveys largely ignore the small and rare species that form the majority of marine molluscan diversity. As a result of a massive effort to sample the benthic molluscs of a complex tropical coastal environment, 23,238 gastropod specimens representing 259 species of Triphoridae—a family with most adult species ranging from 2 to 10 mm—were obtained from a 45,000 hectares study area off the island of Espiritu Santo, Vanuatu. Most species are represented by fewer than 20 specimens and, despite the intensity of the sampling effort, 13% of the species are unique singletons. Spatial heterogeneity was high: out of 416 sampling events, 187 contained triphorids, and 42% of the species occurred at fewer than 5 stations. Most species were small (68% below 5 mm) or very small (22% below 3 mm). A faunal turnover was documented at around 10 m, and another at around 60 m, at the onset of the “twilight zone” that is particularly difficult to sample. On the order of 70% of the species are probably new to science. When dealing with taxonomically difficult groups a morphospecies segregation approach is operationally appropriate to detect patterns of richness, rarity and spatial turn-over. Very few, if any, conservation surveys have the human and funding resources to carry out baseline surveys of the intensity that generated the results presented here. However, as species numbers are often used to promote the conservation interest of a reef, a bay or a stretch of coast, it is essential to know how the numbers were generated: absolute numbers of species are meaningless unless sampling effort and techniques, area surveyed, and size classes targeted are described. Constructed wetland systems built to handle nutrient contaminants are often efficient at removing nitrogen, but ineffective at reducing phosphorus (P) loads. Incorporating a clay-based substrate can enhance P removal in subsurface-flow constructed wetland systems. We evaluated the potential of crushed brick, a recycled building product, and two particle sizes of a palygorskite–bentonite industrial mineral aggregate (calcined clay) to sorb P from simulated nutrient-rich plant nursery effluent. The three substrates were screened for P sorbing behavior using sorption, desorption, and equilibration experiments. We selected one substrate to evaluate in an 8-month field trial to compare field sorption capacity with laboratory sorption capacity. In the laboratory, coarse calcined clay average sorption capacity was 497 mg kg−1 and it sorbed the highest percentage of P supplied (76%), except at exposure concentrations >100 mg L−1 where the increased surface area of fine calcined clay augmented its P sorption capacity. Subsurface-flow mesocosms were filled with coarse calcined clay and exposed to a four and seven day hydraulic retention time treatment. Phosphorus export was reduced by 60 to 74% for both treatments until substrate P-binding sites began to saturate during month seven. During the eight month experiment, the four and seven day treatments fixed 1273 ± 22 mg kg−1 P and 937 ± 16 mg kg−1 P, respectively. Sequential extractions of the P saturated clay indicated that P could desorb slowly over time from various pools within the calcined clay; thus, if the calcined clay were recycled as a soil amendment, most P released would be slowly available for plant uptake and use. This study demonstrated the viability of using coarse calcined clay as a root bed substrate in subsurface-flow treatment wetlands remediating phosphorus from plant nursery runoff.  相似文献

The germination of seeds of Eichhornia crassipes in locations distant from the foci of infestation may be a means of dispersal of this invasive plant. Nonetheless, no modern studies have examined the influence of single components on germination, although the influence of temperature, oxygen and redox potential was examined in earlier studies. The effects of pH, conductivity, nitrate, phosphate, potassium, calcium, iron and boron on germination of E. crassipes were determined in seed germination tests. The results showed that external input of nutrients influences germination, since neither pH nor conductivity alone had any effect, but there was a significant increase in germination in growth media containing phosphorus at 3.95 mg·l(-1) (P/L; 95.5%) and boron at 10 mg·l(-1) (B/L; 97.5%); at higher concentrations, the latter was toxic. These findings contribute to knowledge of factors controlling the germination of E. crassipes seeds. Consequently, E. crassipes seeds would find very good conditions for germination in water classified as hypereutrophic, which may play a decisive role in expansion of this plant.  相似文献
