In order to identify pathogenic correlates of refractory rheumatoid arthritis (RA), antibodies against anti-cyclic citrullinated protein (ACPAs) were investigated in RA patients in whom the dysregulated immune system had been ablated by high-dose chemotherapy (HDC) and autologous haematopoietic stem cell transplantation (HSCT). Six patients with refractory RA were extensively characterized in terms of levels of total immunoglobulins, RA-specific autoantibodies (ACPAs and rheumatoid factor) and antibodies against rubella, tetanus toxoid (TT) and phosphorylcholine before and after HDC plus HSCT. Additionally, the avidity of ACPAs was measured before and after treatment and compared with the avidity of TT antibodies following repeated immunizations. Synovial biopsies were obtained by arthroscopy before HDC plus HSCT, and analyzed by immunohistochemistry. In the three patients with clinically long-lasting responses to HDC plus HSCT (median 423 days), significant reductions in ACPA-IgG levels after therapy were observed (median level dropped from 215 to 34 arbitrary units/ml; P = 0.05). In contrast, stable ACPA-IgG levels were observed in three patients who relapsed shortly after HDC plus HSCT (median of 67 days). Clinical responders had ACPA-IgG of lower avidity (r = 0.75; P = 0.08) and higher degree of inflammation histologically (r = 0.73; P = 0.09). Relapse (after 38 to 530 days) in all patients was preceded by rising levels of low avidity ACPA-IgG (after 30 to 388 days), in contrast to the stable titres of high avidity TT antibodies. In conclusion, humoral autoimmune responses were differentially modulated by immunoablative therapy in patients with synovial inflammation and low avidity ACPA-IgG autoantibodies as compared with patients with high levels of high avidity ACPA-IgG. The distinct clinical disease course after immunoablative therapy based on levels and avidity of ACPA-IgG indicates that refractory RA is not a single disease entity.  相似文献

The cyclin D1 gene encodes a regulatory subunit of the holoenzyme that phosphorylates and inactivates the pRb tumor suppressor to promote nuclear DNA synthesis. cyclin D1 is overexpressed in human breast cancers and is sufficient for the development of murine mammary tumors. Herein, cyclin D1 is shown to perform a novel function, inhibiting mitochondrial function and size. Mitochondrial activity was enhanced by genetic deletion or antisense or small interfering RNA to cyclin D1. Global gene expression profiling and functional analysis of mammary epithelial cell-targeted cyclin D1 antisense transgenics demonstrated that cyclin D1 inhibits mitochondrial activity and aerobic glycolysis in vivo. Reciprocal regulation of these genes was observed in cyclin D1-induced mammary tumors. Cyclin D1 thus integrates nuclear DNA synthesis and mitochondrial function.  相似文献

The addition of nocturnal, Hoplias malabaricus, and diurnal, Crenicichla alta, predatory fishes downstream of barrier waterfalls increases predation threat for a killifish, Rivulus hartii, in Trinidadian streams. We hypothesized that the diel patterning of predation risk would affect prey movement rates, and tested this hypothesis by comparing movement in river sites/zones containing both the nocturnal and diurnal predator with movement in river sites/zones containing only the nocturnal taxon. We evaluated this prediction in the framework of an intermediate threat hypothesis (ITH) that holds that movement will be highest at some intermediate level of threat. We marked prey fish in study sites in two watersheds of a river, each with waterfalls that divided the river into three zones: a predator absent zone (P0), a zone with one nocturnal predator (P1), and a zone with one nocturnal and one diurnal predator (P2), and tested the ITH prediction that movement will be ordered as P0<P1>P2. The single predator promoted longitudinal movement by Rivulus (P0<P1), while zones with the two predators retarded movement for small Rivulus (P1>P2) as predicted by the ITH. However, movement by larger, less vulnerable Rivulus remained elevated (P1=P2 or P2>P1). A displacement experiment in each zone found that threat tended to reduce the probability of a displaced fish reaching home, but the two predator zones did not differ from one another in their effect on this probability. Hence, the prediction that predator activity over the full 24 h diel cycle would retard movement, P2<P1, was not supported with respect to homing. Because habitat and predator communities change predictably from headwater streams to larger rivers in many lotic ecosystems, we present a conceptual model for predicting fish movement behavior along this continuum. The model posits an important role for predation threat, and the size and spacing of refuge patches, suggesting that human alterations of these factors will affect the natural movement of fish in streams.  相似文献

Background  

A polarized light scattering technique was used to monitor the performance of a continuously operated foam fractionation process. The S 11 and S 12 parameters, elements of the light scattering matrix, combined together (S 11 +S 12) have been correlated with the bubble size and liquid content for the case of a freely draining foam. The performance of a foam fractionation column is known to have a strong dependence on the bubble size distribution and liquid hold up in foam. In this study the enrichment is used as a metric, representative of foam properties and column performance, and correlated to the S 11 +S 12 parameter.  相似文献

The adenovirus E1A protein interferes with regulators of apoptosis and growth by physically interacting with cell cycle regulatory proteins including the retinoblastoma tumor suppressor protein and the coactivator proteins p300/CBP (where CBP is the CREB-binding protein). The p300/CBP proteins occupy a pivotal role in regulating mitogenic signaling and apoptosis. The mechanisms by which cell cycle control genes are directly regulated by p300 remain to be determined. The cyclin D1 gene, which is overexpressed in many different tumor types, encodes a regulatory subunit of a holoenzyme that phosphorylates and inactivates PRB. In the present study E1A12S inhibited the cyclin D1 promoter via the amino-terminal p300/CBP binding domain in human choriocarcinoma JEG-3 cells. p300 induced cyclin D1 protein abundance, and p300, but not CBP, induced the cyclin D1 promoter. cyclin D1 or p300 overexpression inhibited apoptosis in JEG-3 cells. The CH3 region of p300, which was required for induction of cyclin D1, was also required for the inhibition of apoptosis. p300 activated the cyclin D1 promoter through an activator protein-1 (AP-1) site at -954 and was identified within a DNA-bound complex with c-Jun at the AP-1 site. Apoptosis rates of embryonic fibroblasts derived from mice homozygously deleted of the cyclin D1 gene (cyclin D1(-/-)) were increased compared with wild type control on several distinct matrices. p300 inhibited apoptosis in cyclin D1(+/+) fibroblasts but increased apoptosis in cyclin D1(-/-) cells. The anti-apoptotic function of cyclin D1, demonstrated by sub-G(1) analysis and annexin V staining, may contribute to its cellular transforming and cooperative oncogenic properties.  相似文献

The movement strategies of birds and mammals are often closely linked to their mating system, but few studies have examined the relationship between mating systems and movement in fishes. We examined the movement patterns of the guppy (Poecilia reticulata) in the Arima river of Trinidad and predicted that sexual asymmetry in reproductive investment would result in male-biased movement. Since male guppies maximize their reproductive success by mating with as many different females as possible, there should be strong selection for males to move in search of mates. In agreement with our prediction, the percentage of fish that emigrated from release pools was higher for males than females (27.3% vs. 6.9%, respectively). Sex ratio was highly variable among pools and may influence a male's decision to emigrate or continue moving. We also detected a positive relationship between body length and the probability of emigration for males and a significant bias for upstream movement by males. Among the few females that did emigrate, a positive correlation was observed between body length and distance moved. Sex-biased movement appears to be related to mating systems in fishes, but the evidence is very limited. Given the implications for ecology, evolution, and conservation, future studies should explicitly address the influence of sex and mating systems on movement patterns.  相似文献

Largemouth bass ( Micropterus salmoides ) with an average weight of 45.5 g were used to study ration level effects on nitrogen excretion, nitrogen retention, and gross efficiency of utilization of nitrogen for growth. Bass were starved 3 to 4 days and then each bass was placed into an aquarium containing a known volume of water. One day after the fishes were placed in aquaria, nitrogen excretion rates were determined; this rate is the maintenance nitrogen excretion. Each fish was then fed one or more shiners ( Notropis cornutus ); fish were fed only once. Nitrogen excretion measurements were made daily until the rates were similar to maintenance rates. The nitrogen excretion rates for each day after feeding which were above the maintenance nitrogen excretion were combined and reflect the total nitrogen excretion for a given ration level. All ration levels were converted to nitrogen consumption in mg and nitrogen absorption was calculated from subtracting the average faecal nitrogen from nitrogen consumption. From data on nitrogen consumption, nitrogen absorption, nitrogen excretion, and faecal nitrogen, calculations were made for nitrogen retention and gross efficiency of utilization of nitrogen for growth.
As ration level and nitrogen absorption increased, nitrogen excretion increased and is described by the equation, Y = 8.56+0.40 X , where Y is total nitrogen excretion and X is nitrogen absorption. Nitrogen retention also increased with nitrogen absorption and is described by the equation, Y =–8.57+0.60 X , where Y is nitrogen retention and X is nitrogen absorption. Efficiency increases rapidly above maintenance, but levels off at higher ration levels and approaches an asymptote of 60%.  相似文献
