Purpose

Fuel economy and emissions of heavy-duty trucks greatly vary based on vehicular/environmental conditions. Large-scale infrastructure construction projects require a large amount of material/equipment transportation. Single-parameter generic hauling models may not be the best option for an accurate estimation of hauling contribution in life cycle assessment (LCA) involving construction projects; therefore, more precise data and parameterized models are required to represent this contribution. This paper discusses key environmental/operational variables and their impact on transportation of materials and equipment; a variable-impact transportation (VIT) model accounting for these variables was developed to predict environmental impacts of hauling.

Methods

The VIT model in the form of multi-nonlinear regression equations was developed based on simulations using the U.S. Environmental Protection Agency (EPA)’s Motor Vehicle Emission Simulator (MOVES) to compute all the impact categories in EPA’s TRACI 2.1 and energy consumption of transportation. Considering actual driving cycles of hauling trucks recorded during a pavement rehabilitation project, the corresponding environmental impacts were calculated, and sensitivity analyses were performed. In addition, an LCA case study based on historical pavement reconstruction projects in Illinois was conducted to analyze the contribution of transportation and variability of its impacts during the pavements’ life cycle.

Results and discussion

The importance of vehicle driving cycles was realized from simulation results. The case study results showed that considering driving cycles using the VIT model could increase the contribution of hauling in total life cycle Global Warming Potential (GWP) and total life cycle GWP itself by 2–4 and 3–5%, respectively. In addition to GWP, ranges of other hauling-related impact categories including Smog, Ozone Depletion, Acidification, and Primary Energy Demand from fuel were presented based on the case study. Ozone Depletion ranged from 9 to 45%, and Smog ranged from 11 to 48% of the total relevant life cycle impacts. The GWP contribution of hauling in pavement LCA ranged between 5 and 32%. The results indicate that the contribution of hauling transportation can be significant in pavement LCA.

Conclusions

For large-scale roadway infrastructure construction projects that need a massive amount of material transportation, high fidelity models and data should be used especially for comparative LCAs that can be used as part of decision making between alternatives. The VIT model provides a simple analytical platform to include the critical vehicular/operational variables without any dependence on an external software; the model can also be incorporated in those studies where some of the transportation activity data are available.

The oncofetal H19 gene transcribes a long non-coding RNA(lncRNA) that is essential for tumor growth. Here we found that numerous established inducers of epithelial to mesenchymal transition(EMT) also induced H19/miR-675 expression. Both TGF-β and hypoxia concomitantly induced H19 and miR-675 with the induction of EMT markers. We identified the PI3K/AKT pathway mediating the inductions of Slug, H19 RNA and miR-675 in response to TGF-β treatment, while Slug induction depended on H19 RNA. In the EMT induced multidrug resistance model, H19 level was also induced. In a mouse breast cancer model, H19 expression was tightly correlated with metastatic potential. In patients, we detected high H19 expression in all common metastatic sites tested, regardless of tumor primary origin. H19 RNA suppressed the expression of E-cadherin protein. H19 up-regulated Slug expression concomitant with the suppression of E-cadherin protein through a mechanism that involved miR-675. Slug also up-regulated H19 expression and activated its promoter. Altogether, these results may support the existence of a positive feedback loop between Slug and H19/miR-675, that regulates E-cadherin expression. H19 RNA enhanced the invasive potential of cancer cells in vitro and enhanced tumor metastasis in vivo. Additionally, H19 knockdown attenuated the scattering and tumorigenic effects of HGF/SF. Our results present novel mechanistic insights into a critical role for H19 RNA in tumor progression and indicate a previously unknown link between H19/miR-675, Slug and E-cadherin in the regulation of cancer cell EMT programs.  相似文献

Conservation of Anatidae in North Africa is hindered by lack of information concerning population size, population trends, and species-habitat relationships. In this work, we used a 2-year survey data in 25 wetlands distributed throughout Morocco to model duck spatial distribution and to assess the relevance of a set of environmental and anthropogenic factors in predicting site occupancy, by means of Generalized Linear Mixed Models (GLMM). Mallards and Ferruginous ducks were the most commonly detected species, whereas White-Headed Ducks and Common Pochards were the least detected ones. An inter-annual variation in site occupancy was recorded for Ruddy Shelducks, Red-Crested Pochards and White-Headed Ducks. Geographical location (measured mainly as distance to the coastline and altitude) was the major predictor of the occurrence probability of Ruddy Shelducks, Marbled Teals and Red-Crested Pochards, while human presence and habitat features were the most relevant factors in shaping Mallard’s distribution. However, none of the considered environmental and anthropogenic factors explained the distribution patterns of the Ferruginous Duck, Common Pochard and Gadwall. The results of this study clearly show that there are still gaps in our knowledge on factors driving wetland occupancy by breeding Anatidae in Morocco. The pursuit of the investigations, while considering other explanatory factors such as water quality (limnological data), diet, predation, and conservation status, is of great importance to more profoundly understand the dynamics of Moroccan duck populations.  相似文献

A quantitative method for measuring simultaneously the flavor and water contents in model spray-dried flavor delivery systems was developed using spectroscopic techniques and chemometrics. Nine encapsulated systems were prepared, consisting of a solid carrier (maltodextrin and gum arabic) and varying the amounts of water and flavor. The model flavors used in this work were a hydrophobic (limonene) and a more hydrophilic (2,5-dimethylpyrazine) single components. Near-infrared (NIR) and low-field time-domain nuclear magnetic resonance (low field TD-NMR) data were acquired on each system and analyzed using multivariate chemometric techniques to develop optimal prediction models. Partial least squares regression models showed good predictive ability, with coefficients of determination (R2) between 0.81 and 1.00 and low root mean square error of cross-validation values compared to the range of concentrations. The predictive ability of the chemometric models computed using the NIR spectra improved significantly when data were pre-processed using multiplicative signal correction. The development of good prediction models (i.e., robust models resulting in accurate predictions for water and flavor content) from the NMR relaxation data spectra was successful only for the hydrophobic limonene systems, yielding prediction models whose performance was better than the models obtained using the NIR data. Overall, NIR spectroscopy and NMR relaxometry were identified as complementary techniques rather than competitive methods in the characterization of encapsulated flavor systems.  相似文献

Two endothermic peaks could be observed for five commercial samples of bovine serum albumin (BSA). The smaller peak observed by differential scanning calorimetry (DSC) corresponded to enthalpy relaxation. This peak was followed on storage of BSA, in its glassy state, after it had been heated above its denaturation temperature. Enthalpy and peak temperature increased with duration of storage. On storage for one week at 60 degrees C, a sample at 8.3% moisture showed a peak at 100 degrees C with an energy value of approximately 2 J per g protein. BSA samples were heated within the DSC sufficiently to eliminate the lower enthalpy peak but without altering the denaturation enthotherm. The amount of physical aging shown by these BSA samples was similar to that of the heat-denatured samples. It was concluded that the heating endotherms of dry BSA reflect both the storage and thermal history of the sample. Possible implications of the enthalpy relaxation of BSA on the behavior of this important protein are considered.  相似文献

Tall fescue EST-SSR markers with transferability across several grass species   总被引:26,自引:0,他引:26  
Tall fescue (Festuca arundinacea Schreb.) is a major cool season forage and turf grass in the temperate regions of the world. It is also a close relative of other important forage and turf grasses, including meadow fescue and the cultivated ryegrass species. Until now, no SSR markers have been developed from the tall fescue genome. We designed 157 EST-SSR primer pairs from tall fescue ESTs and tested them on 11 genotypes representing seven grass species. Nearly 92% of the primer pairs produced characteristic simple sequence repeat (SSR) bands in at least one species. A large proportion of the primer pairs produced clear reproducible bands in other grass species, with most success in the close taxonomic relatives of tall fescue. A high level of marker polymorphism was observed in the outcrossing species tall fescue and ryegrass (66%). The marker polymorphism in the self-pollinated species rice and wheat was low (43% and 38%, respectively). These SSR markers were useful in the evaluation of genetic relationships among the Festuca and Lolium species. Sequencing of selected PCR bands revealed that the nucleotide sequences of the forage grass genotypes were highly conserved. The two cereal species, particularly rice, had significantly different nucleotide sequences compared to the forage grasses. Our results indicate that the tall fescue EST-SSR markers are valuable genetic markers for the Festuca and Lolium genera. 6-(2-Phenylethyl)nicotine (1b; K(i)=15 nM) was unexpectedly found to bind at alpha4beta2 nicotinic cholinergic (nACh) receptors. Although this compound failed to produce nicotine-like agonist action in several functional assays, 1b antagonized the antinociceptive effects of nicotine (mouse tail-flick assay) in a dose-dependent fashion when administered via an intrathecal route.  相似文献
