Soil invertebrates and heavy metal concentrations are heterogeneously distributed in the soil of steppe plots surrounding an iron mining enterprise in southern Russia. This study assesses whether patches of high soil invertebrate abundance coincide with patches of low concentrations of pollutants. For this aim, spatial analysis by distance indices (SADIE) was applied. Three valleys in Belogorye Nature Reserve were chosen. One valley faced the tailing pond to the north and the other two faced south-east or south-west. Two sampling plots were chosen in each valley, 60 m apart from each other. On every plot 16 soil cores were collected from a grid of 4 × 4 units with a 5-m distance between each sample unit. Each soil core had an area of 76 cm2 and was 12–15 cm deep. All macroinvertebrates were hand-sorted and identified to family. Abundance of soil invertebrates was not controlled by patches of metal concentration in the soil. Epigaeic groups, like insects and other invertebrates inhabiting the litter layer, were not directly associated with local parameters of the soil. On the contrary, belowground invertebrate abundance (elaterid larvae and earthworms) showed significant dissociation with some heavy metal (Fe, Pb, Zn) concentrations in the soil. The patchiness of soil pollution may act as a leading factor of belowground soil invertebrate distribution. The spatial structure of animal populations in industrially transformed soils needs further research.  相似文献

Background, aim and scope  

In spite of a number of lingering issues, life cycle assessment (LCA) is widely recognised as one of the most powerful tools to investigate the environmental performance of a product or service. Carbon footprint (CF) analysis can also be considered a subset of LCA, limited to a single impact category (i.e. global warming potential (GWP)). However, the inherent complexity of a full LCA or CF analysis often stands in the way of their widespread application in the industry and policy-making sectors. For these latter ambits, this paper advocates the adoption of tailor-made streamlined approaches, with reduced inventory requirements and impact assessment scope. Two such examples are provided, respectively addressing the evaluation of GWP in the development of new product standards and the GWP savings attainable through the use of recycled materials.  相似文献

Aim Quaternary palaeopalynological records collected throughout the Iberian Peninsula and species distribution models (SDMs) were integrated to gain a better understanding of the historical biogeography of the Iberian Abies species (i.e. Abies pinsapo and Abies alba). We hypothesize that SDMs and Abies palaeorecords are closely correlated, assuming a certain stasis in climatic and topographic ecological niche dimensions. In addition, the modelling results were used to assign the fossil records to A. alba or A. pinsapo, to identify environmental variables affecting their distribution, and to evaluate the ecological segregation between the two taxa. Location The Iberian Peninsula. Methods For the estimation of past Abies distributions, a hindcasting process was used. Abies pinsapo and A. alba were modelled individually, first calibrating the model for their current distributions in relation to the present climate, and then projecting it into the past—the last glacial maximum (LGM) and the Middle Holocene periods—in relation to palaeoclimate simulations. The resulting models were compared with Iberian‐wide fossil pollen records to detect areas of overlap. Results The overlap observed between past Abies refugia—inferred from fossil pollen records—and the SDMs helped to construct the Quaternary distribution of the Iberian Abies species. SDMs yielded two well‐differentiated potential distributions: A. pinsapo throughout the Baetic mountain Range and A. alba along the Pyrenees and Cantabrian Range. These results propose that the two taxa remained isolated throughout the Quaternary, indicating a significant geographical and ecological segregation. In addition, no significant differences were detected comparing the three projections (present‐day, Mid‐Holocene and LGM), suggesting a relative climate stasis in the refuge areas during the Quaternary. Main conclusions Our results confirm that SDM projections can provide a useful complement to palaeoecological studies, offering a less subjective and spatially explicit hypothesis concerning past geographic patterns of Iberian Abies species. The integration of ecological‐niche characteristics from known occurrences of Abies species in conjunction with palaeoecological studies could constitute a suitable tool to define appropriate areas in which to focus proactive conservation strategies.  相似文献

Background

Aberrant promoter DNA methylation has been shown to play a role in acute myeloid leukemia (AML) pathophysiology. However, further studies to discuss the prognostic value and the relationship of the epigenetic signatures with defined genomic rearrangements in acute myeloid leukemia are required.

Methodology/Principal Findings

We carried out high-throughput methylation profiling on 116 de novo AML cases and we validated the significant biomarkers in an independent cohort of 244 AML cases. Methylation signatures were associated with the presence of a specific cytogenetic status. In normal karyotype cases, aberrant methylation of the promoter of DBC1 was validated as a predictor of the disease-free and overall survival. Furthermore, DBC1 expression was significantly silenced in the aberrantly methylated samples. Patients with chromosome rearrangements showed distinct methylation signatures. To establish the role of fusion proteins in the epigenetic profiles, 20 additional samples of human hematopoietic stem/progenitor cells (HSPC) transduced with common fusion genes were studied and compared with patient samples carrying the same rearrangements. The presence of MLL rearrangements in HSPC induced the methylation profile observed in the MLL-positive primary samples. In contrast, fusion genes such as AML1/ETO or CBFB/MYH11 failed to reproduce the epigenetic signature observed in the patients.

Conclusions/Significance

Our study provides a comprehensive epigenetic profiling of AML, identifies new clinical markers for cases with a normal karyotype, and reveals relevant biological information related to the role of fusion proteins on the methylation signature.  相似文献

The D5 domain of TrkC receptors is a docking site for Neurotrophin‐3 (NT‐3), but other domains may be relevant for function or harmonizing signals with p75NTR coreceptors. We report a monoclonal antibody (mAb) 2B7 targeting the juxtamembrane domain of TrkC. mAb 2B7 binds to murine and human TrkC receptors and is a functional agonist that affords activation of TrkC, AKT, and MAPK. These signals result in cell survival but not in cellular differentiation. Monomeric 2B7 Fabs also affords cell survival. Binding of 2B7 mAb and 2B7 Fabs to TrkC are blocked by NT‐3 in a dose‐dependent manner but not by pro‐NT‐3. Expression of p75NTR coreceptors on the cell surface block the binding and function of mAb 2B7, whereas NT‐3 binding and function are enhanced. mAb 2B7 defines a previously unknown neurotrophin receptor functional hot spot; that exclusively generates survival signals; that can be activated by non‐dimeric ligands; and potentially unmasks a site for p75‐TrkC interactions. © 2009 Wiley Periodicals, Inc. A number of aliphatic diamines and amino-alcohols and several of their alkyl, acyl and carbamoyl derivatives, have been synthesised and evaluated in vitro on cultures of Leishmania spp. In general, diamine derivatives resulted to be more potent than their amino-alcohol or amino-ether analogues. Two diamine derivatives (8b and 9d) and one amino-alcohol (6a) showed a fair inhibition of parasite growth, at concentrations below 10 microg/mL, with potencies close to that of the reference drug, amphotericin B. Artificial viruses consisting of DNA plasmid packaged in vitro into virus-like particles (VLPs) are new vehicles for gene transfer. We therefore investigated the ability of nine human papillomavirus (HPV) VLPs to interact with heterologous DNA and transfer genes. HPV 16, 18, 31, 33, 39, 45, 58, 59, and 68 VLPs were able to bind heterologous DNA and to transfer genes into Cos-7 cells. Inhibition of gene transfer by preincubation of the pseudovirions with heparin confirmed that heparan sulfate on the cell surface plays a role as cell receptor for HPVs. As HPV neutralizing antibodies are mainly type-specific, gene transfer with different HPV pseudovirions offers the possibility of their sequential use in vivo for a greater efficacy.  相似文献

Three new series of benzo[d]isothiazole, benzothiazole and thiazole Schiff bases were synthesized and tested in vitro with the aim of identifying novel lead compounds active against emergent and re-emergent human and cattle infectious diseases (AIDS, hepatitis B and C, tuberculosis, bovine viral diarrhoea) or against drug-resistant cancers (leukaemia, carcinoma, melanoma, MDR tumors) for which no definitive cure or efficacious vaccine is available at present. In particular, these compounds were evaluated in vitro against representatives of different virus classes, such as a HIV-1 (Retrovirus), a HBV (Hepadnavirus) and the single-stranded RNA(+) viruses Yellow fever virus (YFV) and Bovine viral diarrhoea virus (BVDV), both belonging to Flaviviridae. Title compounds were also tested against representatives of Gram-positive and Gram-negative bacteria (Staphylococcus aureus, Salmonella spp.), various atypic mycobacterial strains (Mycobacterium fortuitum and Mycobacterium smegmatis), yeast (Candida albicans) and mould (Aspergillus fumigatus). None of the compounds showed antiviral or antimicrobial activity. The benzo[d]isothiazole compounds showed a marked cytotoxicity (CC(50)=4-9 microM) against the human CD4(+) lymphocytes (MT-4) that were used to support HIV-1 growth. For this reason, the most cytotoxic compounds of this series were evaluated for their antiproliferative activity against a panel of human cell lines derived from haematological and solid tumors. The results highlighted that all the benzo[d]isothiazole derivatives inhibited the growth of leukaemia cell lines, whereas only one of the above mentioned compounds (1e) showed antiproliferative activity against two solid tumor-derived cell lines.  相似文献
