The ability of S. putrefaciens to reduce Fe(III) complexed by a variety of ligands has been investigated. All of the ligands tested caused the cation to be more susceptible to reduction by harvested whole cells than when uncomplexed, although some complexes were more readily reduced than others. Monitoring rates of reduction by a ferrozine assay for Fe(II) formation proved inadequate using Fe(III) ligands giving Fe(II) complexes of low kinetic lability (e.g. EDTA). A more suitable assay for Fe(III) reduction in the presence of such ligands proved to be the observation of associated cytochrome oxidation and re-reduction. Where possible, an assay for Fe(III) reduction based upon the disappearance of Fe(III) complex was also employed. Reduction of all Fe(III) complexes tested was totally inhibited by the presence of O2, partially inhibited by HQNO and slower in the absence of a physiological electron donor. Upon cell fractionation, Fe(III) reductase activity was detected exclusively in the membranes. Using different physiological electron donors in assays on membranes, relative reduction rates of Fe(III) complexes complemented the data from whole cells. The differences in susceptibility to reduction of the various complexes are discussed, as is evidence for the respiratory nature of the reduction.  相似文献

Shallow intertidal habitats are recognised as critical for larval and juvenile fish, and are often assumed to function as refuge areas where predation risk is reduced. Yet there is growing evidence that suggest these areas may also be regularly inhabited by large bodied fish and be the site of high levels of juvenile fish predation. In the present study we examined the use of an intermittently available surf zone habitat in tropical northern Australia by a diverse community of large-bodied teleosts (mean total length 444 mm), sharks (mean total stretched length 658 mm) and rays (mean total stretched length 1,108 mm). Drawing on the methods and ecological knowledge of a local commercial fishery, gillnets were used to capture fish as they entered the surf zone on the flooding tide. Monthly surveys over a one-year period revealed a dynamic assemblage consisting of 30 species of teleosts (mostly caught as adults) and 14 species of sharks and rays (mostly caught as juveniles or young of the year). Although it is unclear why these fish use this habitat, we conclude that it may support a broad range of biological benefits including spawning for teleosts, parturition for elasmobranchs, as well as foraging and refuge. The unique findings of this study highlight existing knowledge gaps and the need to better understand what fish use the intertidal zone and why. This should be a high priority given the increasing anthropogenic pressures on coastal margins.  相似文献

The Langendorff isolated rat heart was adapted to the study of minute-to-minute percentage changes in bulk protein degradation by using non-recirculating perfusion. Hearts were perfused at 8 ml/min at 35 degrees C with Krebs-Henseleit buffer containing 11 mM-glucose, and only hearts with regular ventricular rhythm were employed. Proteins were labelled by infusion of [3H]leucine for 0.5 h in vitro. A complete amino acid mixture was then added at 3 times normal rat extracellular concentrations. After labelling, the re-incorporation of [3H]leucine was competitively inhibited by addition of either 4 mM-leucine or 20 microM-cycloheximide. The residual unincorporated radioactivity and the preferentially labelled rapid-turnover proteins were eliminated during a 3 h preliminary perfusion period. The basal rate of release of [3H]leucine and percentage changes were then determined at 1 min intervals, by using each heart as its own control. Leucine metabolism was inconsequential to results. Exchange of intracellular leucine pools with extracellular leucine and subsequent release in effluent perfusate was 95% complete within approx. 2 min. The basal rate of protein degradation was unchanged by electrical stimulation of the heart rate to 360 beats/min or cessation of contractile activity by membrane depolarization under 25 mM-KCl. Infusion of the beta-agonist isoprenaline at 5-500 nM caused a graded inhibition of myocardial protein degradation within 5-6 min, with a maximum inhibition of 30%. This inhibition was sustained for at least 1 h of drug administration and was reversed within 4-6 min of cessation of isoprenaline or simultaneous infusion of 1 microM of the beta-receptor antagonist propranolol. Minute-to-minute adrenergic proteolytic control was a simultaneous co-variable with beta-receptor-mediated inotropic changes in right-intraventricular systolic pressure. Stoppage of the heart in asystole by the Ca2+-channel blocker nifedipine (0.7 microM) delayed the onset, but did not cause sustained reversal, of adrenergic-inhibited degradation, indicating the absence of a direct obligatory mechanistic linkage between the events of the contraction-relaxation cycle and protein degradation in this preparation.  相似文献

  1. The disappearance of nitrate from suspensions of intact, washed cells of Rhodopseudomonas capsulata strain N22DNAR+ was measured with an ion selective electrode. In samples taken from phototrophic cultures grown to late exponential phase, nitrate disappearance was partially inhibited by light but was not affected by the presence of ammonium. Nitrate disappearance from samples from low density cultures in the early exponential phase of growth was first inhibited and later stimulated by light. In these cells ammonium ions inhibited the light-dependent but not the dark disappearance of nitrate. It is concluded that cells in the early exponential phase of growth possess both an ammonium-sensitive, assimilatory pathway for nitrate reduction (NRI) and an ammonium-insensitive pathway for nitrate reduction (NRII) which is linked to respiratory electron flow and energy conservation. In cells harvested in late exponential phase only the respiratory pathway for pitrate reduction is detectable.

Nitrate reduction, as judged by the oxidation of reduced methyl viologen by anaerobic cell suspensions, was measured at high rates in those strains of R. capsulata (AD2, BK5, N22DNAR+) which are believed to possess NRII activity but not in those strains (Kbl, R3, N22) which only manifest the ammonium-sensitive NRI pathway. On this basis we have used nitrate-dependent oxidation of reduced methyl viologen as a diagnostic test for the nitrate reductase of NRII in cells harvested from cultures of R. capsulata strain AD2. The activity was readily detectable in cells from cultures grown aerobically in the dark with ammonium nitrate as source of nitrogen. When the oxygen supply to the culture was withdrawn, the level of methyl viologen-dependent nitrate reductase increased considerably and nitrite accumulated in the culture medium. Upon reconnecting the oxygen supply, methyl viologen-dependent nitrate reductase activity decreased and the reduction of nitrate to nitrite in the culture was inhibited. It is concluded that the respiratory nitrate reductase activity is regulated by the availability of electron transport pathways that are linked to the generation of a proton electrochemical gradient.

We investigated the effects of Tween emulsifier fatty acid chain length on the shear stability and crystallization behavior of 35 wt% partially crystalline oil-in-water emulsions prepared with and without 1 wt% sodium caseinate. Emulsions containing sodium caseinate and Tween 20, 40, 60 or 80 varied in shear stability, degree of supercooling and crystallization behavior depending on the type and concentration of Tween as well as the presence of protein. Generally, emulsions containing the unsaturated emulsifier Tween 80 were the most shear sensitive followed by the saturated emulsifiers Tween 20, 40 and 60 in order of increasing fatty acid chain length. Long chain saturated Tween emulsifiers (40 and 60) improved shear stability regardless of whether sodium caseinate was present indicating that alone, these emulsifiers form more robust interfacial films compared to the saturated short chain length Tween 20 and Tween 80. In emulsions prepared with sodium caseinate, the degree of supercooling decreased and the crystallization rate diminished with increasing saturated fatty acid chain length but only negligible changes were found without sodium caseinate. Together, these findings indicate that long chain saturated Tween emulsifiers provide better emulsion stability regardless of the presence of sodium caseinate but with sodium caseinate, stability may also be affected by changes to fat crystallization. These novel findings provide guidance on how combinations of proteins and emulsifiers can be used to modify and control the stability of partially crystalline oil-in-water emulsions through their combined effects on the properties of the interfacial film and fat crystallization.  相似文献

Endometriosis is a common condition in reproductive-aged women characterized by ectopic endometrial lesions of varied appearance, including red, white, blue, black or powder burn coloration, which contribute to chronic pain and infertility. The immunoconjugate molecule (Icon) targets Tissue Factor, a transmembrane receptor for Factor VII/VIIa that is aberrantly expressed in the endothelium supporting ectopic endometrial tissue. Icon has been shown to cause regression of endometriosis in a murine model of disease but prior to this study had not been tested in non-human primates. This study evaluated Icon as a novel treatment for endometriosis in non-human primates (Papio anubis) using an adenoviral vector (AdIcon) delivery system. Female baboons (n?=?15) underwent surgical induction of endometriosis. After laparoscopic confirmation of endometriosis lesions 6-weeks post-surgery, the treatment group (n?=?7) received weekly intraperitoneal injections of viral particles carrying the sequence for Icon, resulting in expression of the protein, while the control group (n?=?8) received no treatment. Icon preferentially reduced the number and volume of red vascularized lesions. Icon may present a novel treatment for endometriosis by degrading red vascularized lesions, likely by targeting tissue factor aberrantly expressed in the lesion vasculature.  相似文献

Dengue is a major global disease requiring improved treatment and prevention strategies. The recently licensed Sanofi Pasteur Dengvaxia vaccine does not protect children under the age of nine, and additional vaccine strategies are thus needed to halt this expanding global epidemic. Here, we employed a molecular engineering approach and plant expression to produce a humanized and highly immunogenic poly‐immunoglobulin G scaffold (PIGS) fused to the consensus dengue envelope protein III domain (cEDIII). The immunogenicity of this IgG Fc receptor‐targeted vaccine candidate was demonstrated in transgenic mice expressing human FcγRI/CD64, by induction of neutralizing antibodies and evidence of cell‐mediated immunity. Furthermore, these molecules were able to prime immune cells from human adenoid/tonsillar tissue ex vivo as evidenced by antigen‐specific CD4+ and CD8+ T‐cell proliferation, IFN‐γ and antibody production. The purified polymeric fraction of dengue PIGS (D‐PIGS) induced stronger immune activation than the monomeric form, suggesting a more efficient interaction with the low‐affinity Fcγ receptors on antigen‐presenting cells. These results show that the plant‐expressed D‐PIGS have the potential for translation towards a safe and easily scalable single antigen‐based tetravalent dengue vaccine.  相似文献
