Identification and Distribution of Phenylacetic Acid in the Brain of the Rat

Abstract: Phenylacetic acid, the major metabolite of phenylethylamine, has been identified and quantitated in rat brain regions by capillary column high-resolution gas chromatography mass spectrometry. Its distribution is heterogeneous and correlates with that of phenylethylamine. The values obtained were (ng/g ± SEM): whole brain, 31.2 ± 2.7; caudate nucleus, 64.6 ± 6.5; hypothalamus, 60.1 ± 7.4; cerebellum, 31.3 ± 2.9; brainstem, 33.1 ± 3.3, and the "rest," 27.6 ± 3.0.     

Thrombocyte substitution in acute leukemia. Effect of histocompatibility on the clinical efficacy

Thrombocyte substitution is an essential prerequisite for intensive cytoreductive therapy in acute leukemia. Evaluating 228 thrombocyte transfusions in 17 patients shows that the clinical effectiveness of thrombocyte concentrates can be increased by making the coordination of HLA antigens of donor and receiver as good as possible. When measured in the corrected increment (CI) 24 hours after transfusion, the effectiveness of A3/B1 match preparations (CI = 7.0 +/- 1.6) is significantly higher than that of random preparations (CI = 3.0 +/- 0.5). With the presence of HLA antibodies an effective substitution (CI24 greater than or equal to 4.5) can only be achieved by A3/B1 match thrombocytes. This can only be realized by applying the fourfold thrombapheresis of single donors.     

In vivo suppression of phytochrome aggregation by the GroE chaperonins in Escherichia coli

When expressed in Escherichia coli, a truncated form of phytochrome (oat PHYA AP3 residues 464-1129) self associates to form a series of products ranging in size from monomers to aggregates of greater than 20 subunits. When these same phytochrome sequences are coexpressed with the chaperonins GroEL and GroES, the truncated phytochrome migrates as a native-like dimer in size exclusion chromatography and no higher-order aggregates were detected. GroEL and GroES inhibition of phytochrome aggregation in E. coli presumably occurs via the suppression of folding pathways leading to incorrectly folded phytochrome.     

Biosynthesis of man-β-G1cNAc-G1cNAc-pyrophosphoryl-polyprenol by a solubilized enzyme from aorta

The particulate enzyme fraction from pig aorta was treated with Triton X-100 or Nonidet P-40 to yield a soluble enzyme preparation. This solubilized enzyme catalyzed the transfer of mannose from GDP-[¹⁴C]mannose, but not from [¹⁴C]mannosyl-phosphoryl-polyprenol, to G1cNAc-G1cNAc-pyrophosphoryl-polyprenol to form the trisaccharide-lipid, Man-β-GlcNAc-GlcNAc-pyrophosphoryl-polyprenol. The trisaccharide-lipid formed in these reactions was isolated by solvent fractionation and was subjected to mild acid hydrolysis to release the [¹⁴C]trisaccharide. Essentially all of the radioactivity was released from this trisaccharide as mannose upon treatment with β-mannosidase while α-mannosidase had no effect.     

Tranylcypromine: stimulation of eating through alpha-adrenergic neuronal system in the paraventricular nucleus.

Injection of norepinephrine (NE) into the paraventricular nucleus of the hypothalamus has been shown to elicit eating in satiated rats. In the present study, the monoamine oxidase inhibitor tranylcypromine (TCP), which is known to release endogenous NE, was found to produce a similar effect in rats maintained and tested on a palatable milk-mash combination diet. The effect of this antidepressant drug was positively correlated in magnitude with the NE effect in the same animals. It was selectively antagonized by central injection of drugs which block α-adrenergic receptors. Local pretreatment with NE synthesis inhibitors similarly blocked the TCP eating response but had no effect on eating elicited by exogenous NE. It is suggested from these results that TCP is stimulating eating through the release of endogenous NE from adrenergic neurons innervating the region of the paraventricular nucleus.     

Oxidation-reduction potential dependence of low-temperature photoreactions of chloroplast Photosystem II

The light-induced free-radical signal of Photosystem II (observed after illumination at 77 °K) has been studied in chloroplasts as a function of the oxidation-reduction potential established prior to freezing. The intensity of the light-induced signal is unchanged in the potential region of +590 mV to +760 mV. At higher potential (+850 mV), there is a 30% decrease in signal intensity. The light-induced signal decreases to zero in the low-potential region, with a midpoint potential of +475 mV. These results are considered in terms of a Photosystem II reaction-center complex in which the light-induced free-radical signal arises from the oxidized form of the reaction-center chlorophyll, and this chlorophyll molecule is capable of being reduced at liquid-nitrogen temperature by a secondary electron donor which has a midpoint oxidation-reduction potential of +475 mV.
