Trehalose is a natural non-reducing sugar that is found in the vast majority of organisms such as bacteria, yeasts, invertebrates and even in plants. Regarding its features, it is considered as a unique compound. It plays a key role as a carbon source in lower organisms and as an osmoprotectant or a stabilizing molecule in higher animals and plants. Although in plants it is present in a minor quantity, its levels rise upon exposure to abiotic stresses. Trehalose is believed to play a protective role against different abiotic stressful cues such as temperature extremes, salinity, desiccation. Moreover, it regulates water use efficiency and stomatal movement in most plants. Detectable endogenous trehalose levels are vital for sustaining growth under stressful cues. Exogenously applied trehalose in low amounts mitigates physiological and biochemical disorders induced by various abiotic stresses, delays leaf abscission and stimulates flowering in crops. External application of trehalose also up-regulates the stress responsive genes in plants exposed to environmental cues. The genetically modified plants with trehalose biosynthesis genes exhibit improved tolerance against stressful conditions. An increased level of trehalose has been observed in transgenic plants over-expressing genes of microbial trehalose biosynthesis. However, these transgenic plants display enhanced tolerance to heat, cold, salinity, and drought tolerance. Due to multiple bio-functions of this sugar, it has gained considerable ground in various fields. However, exogenous use of this bio-safe sugar would only be possible under field conditions upon adopting strategies of low-cost production of trehalose. In short, trehalose is a unique chemical that preserves vitality of plant life under harsh ecological conditions. Certainly, the new findings of this disaccharide will revolutionize a wide array of new avenues.
Fetal exposure to hyperglycemia impacts negatively kidney development and function.
Objective
Our objective was to determine whether fetal exposure to moderate hyperglycemia is associated with epigenetic alterations in DNA methylation in peripheral blood cells and whether those alterations are related to impaired kidney function in adult offspring.
Design
Twenty nine adult, non-diabetic offspring of mothers with type 1 diabetes (T1D) (case group) were matched with 28 offspring of T1D fathers (control group) for the study of their leukocyte genome-wide DNA methylation profile (27,578 CpG sites, Human Methylation 27 BeadChip, Illumina Infinium). In a subset of 19 cases and 18 controls, we assessed renal vascular development by measuring Glomerular Filtration Rate (GFR) and Effective Renal Plasma Flow (ERPF) at baseline and during vasodilatation produced by amino acid infusion.
Results
Globally, DNA was under-methylated in cases vs. controls. Among the 87 CpG sites differently methylated, 74 sites were less methylated and 13 sites more methylated in cases vs. controls. None of these CpG sites were located on a gene known to be directly involved in kidney development and/or function. However, the gene encoding DNA methyltransferase 1 (DNMT1)—a key enzyme involved in gene expression during early development–was under-methylated in cases. The average methylation of the 74 under-methylated sites differently correlated with GFR in cases and controls.
Conclusion
Alterations in methylation profile imprinted by the hyperglycemic milieu of T1D mothers during fetal development may impact kidney function in adult offspring. The involved pathways seem to be a nonspecific imprinting process rather than specific to kidney development or function. 相似文献
The acrosome is an organelle that is central to sperm physiology and a defective acrosome biogenesis leads to globozoospermia, a severe male infertility. The identification of the actors involved in acrosome biogenesis is therefore particularly important to decipher the molecular pathogeny of globozoospermia. We recently showed that a defect in the DPY19L2 gene is present in more than 70% of globozoospermic men and demonstrated that Dpy19l2, located in the inner nuclear membrane, is the first protein involved in the attachment of the acrosome to the nuclear envelope (NE). SUN proteins serve to link the nuclear envelope to the cytoskeleton and are therefore good candidates to participate in acrosome-nucleus attachment, potentially by interacting with DPY19L2. In order to characterize new actors of acrosomal attachment, we focused on Sun5 (also called Spag4l), which is highly expressed in male germ cells, and investigated its localization during spermatogenesis. Using immunohistochemistry and Western blot experiments in mice, we showed that Sun5 transits through different cellular compartments during meiosis. In pachytene spermatocytes, it is located in a membranous compartment different to the reticulum. In round spermatids, it progresses to the Golgi and the NE before to be located to the tail/head junction in epididymal sperm. Interestingly, we demonstrate that Sun5 is not, as initially reported, facing the acrosome but is in fact excluded from this zone. Moreover, we show that in Dpy19l2 KO spermatids, upon the detachment of the acrosome, Sun5 relocalizes to the totality of the NE suggesting that the acrosome attachment excludes Sun5 from the NE facing the acrosome. Finally, Western-blot experiments demonstrate that Sun5 is glycosylated. Overall, our work, associated with other publications, strongly suggests that the attachment of the acrosome to the nucleus does not likely depend on the formation of SUN complexes. 相似文献
Karst aquifers have a high degree of heterogeneity and anisotropy in their geologic and hydrogeologic properties which makes predicting their behavior difficult. This paper evaluates the application of the Equivalent Porous Media (EPM) approach to simulate groundwater hydraulics and contaminant transport in karst aquifers using an example from the North Coast limestone aquifer system in Puerto Rico. The goal is to evaluate if the EPM approach, which approximates the karst features with a conceptualized, equivalent continuous medium, is feasible for an actual project, based on available data and the study scale and purpose. Existing National Oceanic Atmospheric Administration (NOAA) data and previous hydrogeological U. S. Geological Survey (USGS) studies were used to define the model input parameters. Hydraulic conductivity and specific yield were estimated using measured groundwater heads over the study area and further calibrated against continuous water level data of three USGS observation wells. The water-table fluctuation results indicate that the model can practically reflect the steady-state groundwater hydraulics (normalized RMSE of 12.4%) and long-term variability (normalized RMSE of 3.0%) at regional and intermediate scales and can be applied to predict future water table behavior under different hydrogeological conditions. The application of the EPM approach to simulate transport is limited because it does not directly consider possible irregular conduit flow pathways. However, the results from the present study suggest that the EPM approach is capable to reproduce the spreading of a TCE plume at intermediate scales with sufficient accuracy (normalized RMSE of 8.45%) for groundwater resources management and the planning of contamination mitigation strategies. 相似文献
A practical evaluation of one- and two-dimensional applications of electric fields for in situ extraction of contaminants is provided. The evaluation is based on contaminant transport by electroosmosis and ion migration. Parameters evaluated include electrode requirements, effectiveness of electric field distribution, remediation time, and energy expenditure. Formulation is provided for calculating cost components of the process, including electrode, energy, chemicals, posttreatment, fixed, and variable costs. Equations are also provided for evaluating optimum electrode spacings based on energy and time requirements. The derivations show that spacing between same-polarity electrodes is as significant in cost calculations and in process effectiveness as that between anodes and cathodes. Decreasing the same-polarity electrode spacing to half the anode-cathode spacing will result in a 100% increase in electrode requirements, but will decrease the area of the ineffective electric field by one half. Selection of the voltage gradient impacts the optimum electrode spacing. The analysis show that a minimum exists in the cost versus electrode spacings relationship. 相似文献
The current research was performed to evaluate the effect of Bacillus megaterium MCR-8 on mitigation of nickel (Ni) stress in Vinca rosea grown on Ni-contaminated soil (50, 100, and 200 mg Ni kg?1 soil). The treated plants exhibited reduced growth, biomass, gas exchange capacity, and chlorophyll (Chl) content under Ni stress. The inoculated plants growing in Ni-contaminated media exhibited relatively higher growth, total soluble protein, and proline contents. Similarly, bacterial inoculation improved the activity of antioxidant enzymes including superoxide dismutase (SOD), catalase (CAT), peroxidase (POD), and ascorbate peroxidase (APX) under Ni stress. The Ni stress alleviation in inoculated plants was attributed to the reduced level of malondialdehyde (MDA) and hydrogen peroxide (H2O2), enhanced synthesis of protein, proline, phenols, and flavonides in conjunction with improved activity of antioxidant enzymes. The growth-promoting characteristics of microbe such as 1-aminocyclopropane-1-carboxylate deaminase (ACCD) and phosphate solubilization activity, siderophore, and auxin production capability also improved the growth and stress mitigation in inoculated plants. Furthermore, the inoculated plants exhibited higher value for bioconcentration factor (BCF), translocation factor (TF), and resulted in higher loss of Ni content from soil. The current results exhibited the beneficial role of B. megaterium MCR-8 regarding stress alleviation and Ni phytoextraction by V. rosea. 相似文献
A study was conducted to find out the role of ascorbic acid (AsA) in modulating growth and different physio-biochemical attributes of canola plants under well-watered as well as water-deficit conditions. Drought stress imposed on 60 % field capacity significantly decreased the shoot and root fresh and dry weights, leaf chlorophyll contents, shoot and root P, root K+, and activity of CAT enzyme, while increased chlorophyll a/b contents, MDA, NPQ, leaf total phenolics, free proline and GB contents in both canola cultivars. Foliar-applied varying levels (50, 100 and 150 mg L?1) of AsA enhanced shoot and root fresh and root dry weights, qN, NPQ, shoot and root P, AsA as well as the activity of POD enzyme particularly under drought stress conditions. Of both canola cultivars, cv. Dunkeld was higher in shoot fresh weights, ETR and Fv/Fm, MDA, proline and GB contents, and POD activity, however, cv. Cyclone in total phenolics and qN under well-watered and water-deficit conditions. Overall, the foliar-applied AsA had a positive effect, though not marked, on salt sensitive cv. Cyclone in terms of improved growth and other attributes, whereas exogenously applied AsA had a non-significant effect on relatively salt tolerant cv. Dunkeld. 相似文献
Primary objective: This study uses numerical analysis and validation against clinical data to develop a method to correct intraocular pressure (IOP) measurements obtained using the Corvis Tonometer for the effects of central corneal thickness (CCT), and age. Materials and Methods: Finite element analysis was conducted to simulate the effect of tonometric air pressure on the intact eye globe. The analyses considered eyes with wide variations in IOP (10–30 mm Hg), CCT (445–645 microns), R (7.2–8.4 mm), shape factor, P (0.6–1) and age (30–90 years). In each case, corneal deformation was predicted and used to estimate the IOP measurement by Corvis (CVS-IOP). Analysis of the results led to an algorithm relating estimates of true IOP as a function of CVS-IOP, CCT and age. All other parameters had negligible effect on CVS-IOP and have therefore been omitted from the algorithm. Predictions of corrected CVS-IOP, as obtained by applying the algorithm to a clinical data-set involving 634 eyes, were assessed for their association with the cornea stiffness parameters; CCT and age. Results: Analysis of CVS-IOP measurements within the 634-large clinical data-set showed strong correlation with CCT (3.06 mm Hg/100 microns, r2 = 0.204) and weaker correlation with age (0.24 mm Hg/decade, r2 = 0.009). Applying the algorithm to IOP measurements resulted in IOP estimations that became less correlated with both CCT (0.04 mm Hg/100 microns, r2 = 0.005) and age (0.09 mm Hg/decade, r2 = 0.002). Conclusions: The IOP correction process developed in this study was successful in reducing reliance of IOP measurements on both corneal thickness and age in a healthy European population. 相似文献
