RNA-dependent RNA polymerase (RdRp) activity was detected in the crude microsomal fraction of rice cultured cells that contain a 14 kbp double-stranded RNA (dsRNA). RdRp activity is maximal in the presence of all four nucleotide triphosphates and Mg2+ ion and is resistant to inhibitors of DNA-dependent RNA polymerases (actinomycin D and alpha-amanitin). RdRp activity increases approximately 2.5-fold in the presence of 0.5% deoxycholate. Treatment of purified microsomal fraction with proteinase K plus deoxycholate suggests that the RdRp enzyme complex with its own 14 kb RNA template is located in vesicles. The RdRp enzyme complex was solubilized with Nonidet P-40 and purified by glycerol gradient centrifugation, then exogenous RNA templates were added. Results indicate that exogenous dsRNA reduces RNA synthesis from the endogenous 14 kb RNA template.  相似文献

The activity of ribosomal RNA (rRNA) genes as judged by nucleolar size and rRNA synthesis has been shown to depend upon the phase of diapause in the eggs of Bombyx mori. In the present study, we found that nucleolar size in diapausing eggs was enlarged at a very early stage during cold treatment, a procedure necessary for the termination of diapause. In contrast, the intrinsic capacity of ribosomal RNA synthesis in the chilled eggs, as examined at 25°C by radioactive precursor incorporation into rRNA, increased much later, in parallel with the break of diapause. The early phase of cold treatment is the period when the eggs undergo some important changes (the so-called diapause development), preparing for diapause termination. Thus we infer that the above mentioned increase in nucleolar size may be one of the features of diapause development.  相似文献

Rice CEBiP recognizes chitin oligosaccharides on the fungal cell surface or released into the plant apoplast, leading to the expression of plant disease resistance against fungal infection. However, it has not yet been reported whether CEBiP is actually required for restricting the growth of fungal pathogens. An automated image-matching technique is presented to assess alignment of the entire lower extremity for normal and implanted knees and the positioning of implants with respect to bone. Sawbone femur and tibia and femoral and tibial components of a total knee arthroplasty system were used. Three spherical markers were attached to each sawbone and each component to define the local coordinate system. Outlines of the three-dimensional (3D) bone models and component computer-aided design (CAD) models were projected onto extracted contours of the femur, tibia, and implants in frontal and oblique X-ray images. Three-dimensional position of each model was recovered by minimizing the difference between the projected outline and the contour. Median values of the absolute error in estimating relative positions were within 0.5 mm and 0.6° for the femur with respect to the tibia, 0.5 mm and 0.5° for the femoral component with respect to the tibial component, 0.6 mm and 0.6° for the femoral component with respect to the femur, and 0.5 mm and 0.4° for the tibial component with respect to the tibia, indicating significant improvements when compared to manually obtained results.  相似文献

The requirement of Ca2+ for the hydrolysis of phosphatidylinositol 4,5-bisphosphate (PtdInsP2) or the accumulation of inositol 1,4,5-trisphosphate (InsP3) in macrophages stimulated with fMet-Leu-Phe was examined using [32P]Pi or [3H]inositol-labeled cells. The dependence on Ca2+ of inositol-trisphosphate phosphatase was also examined. The application of 1 X 10(-8) M fMet-Leu-Phe caused a rapid decrease in the amount of PtdInsP2 to 70% of the control within 10 s, and the decrease was reverted to the control level by prolonged incubation. The decrease in the amount of PtdInsP2 accompanied the accumulation of phosphatidic acid and of InsP3, indicating that the loss of PtdInsP2 is due to phosphodiesteric breakdown. The dose-dependence of fMet-Leu-Phe or its analog on the hydrolysis of PtdInsP2 was much the same as that of the increase in intracellular free Ca2+ concentration in macrophages. The loss of PtdInsP2 as induced by fMet-Leu-Phe was similarly observed in macrophages treated with ionophore A23187 in the absence of external Ca2+ for 10 min. InsP3 was degraded by the particulate or cytosol fraction prepared from macrophages, and the activity of inositol-trisphosphate phosphatase in the particulate fraction was higher than that in the cytosol fraction. The enzyme in the cytosol fraction required Mg2+ for activity, and was activated by free Ca2+ concentrations ranging from 10(-7) to 10(-6) M in the presence of 1 mM MgCl2.  相似文献

The morphologic changes in PMNs induced by an i.p. injection of PSK, a polysaccharide from the mycelia ofCoriolus versicolor, and tumor cells undergoing cell death, were evaluated by immunohistochemical staining and electron microscopy. Male C3H/He mice, 8–10-weeks old, received an i.p. injection of 125 mg/kg of PSK. Their PMNs were obtained 6 h after the PSK injection by peritoneal lavage. N-CWS (Nocardia rubra cell wall skeleton) was added at the start of the chromium release assay using the MM46 mammary carcinoma cell line, which is syngeneic to C3H/He mice, as target cells. During the cytotoxic assay, the cells were fixed at various time points. The MM46 cells expressed ICAM-1 while the PMNs expressed both ICAM-1 and LFA-1 as determined by immunohistochemical staining and immunoelectron microscopy using anti-ICAM-1 and anti-LFA-1 antibodies. PMNs with ruffle-like microvilli adhered to the MM46 tumor cells 30 min after the addition of N-CWS. Immunoelectron microscopic findings suggested that the adhesion molecules were LFA-1 on the PMNs and ICAM-1 on the MM46 tumor cells, but cell fusion between the PMNs and tumor cells was not observed. The MM46 tumor cells gradually lost their microvilli, which showed cell damage, and died 6–7 h after the addition of the N-CWS. This time course of tumor cell death is compatible with the results of the cytotoxic assay. Pretreatment of PMNs by anti-LFA-1 antibody suppressed % lysis of MM46 tumor cells from 90 % to 10 %(p<0.01). These data suggest that adhesion molecule on the surface of PMNs such as LFA-1 might play an important role on signal transduction of these PMNs cytotoxic function in this experimental system.  相似文献

The movements of the infective third-stage larvae (L3) of a rodent parasitic nematode Strongyloides ratti were examined on a sodium chloride (NaCl) gradient set up on agarose plates. The movements of larvae were followed by observing their tracks on the surface of the agarose. The direction of movement depended on the NaCl concentration at the point of their initial placement on the gradient. Larvae placed at between 230 and 370 mM NaCl tended to migrate towards areas of lower concentration. On the other hand, when placed at concentrations less than 20 mM NaCl, larvae tended to migrate initially towards higher concentrations but did not linger in areas where the concentration was over approximately 80 mM NaCl. It seems that S. ratti L3, tested in vitro, prefer regions with a concentration of NaCl below 80 mM NaCl. Two typical chemokinetic behaviors are seen; a unidirectional avoidance movement when initially placed in unfavorable environmental conditions and a random dispersal movement when placed within an area of favorable conditions. Track patterns were straight in the avoidance movement but included multiple changes of direction and loops in the dispersal movement. This study introduces an assay system suitable for studying chemokinetic behavior of larvae of Strongyloides ratti.  相似文献

Genomic DNA for a class IV chitinase was cloned from yam (Dioscorea opposita Thunb) leaves and sequenced. The deduced amino acid sequence shows 50 to 59% identity to class IV chitinases from other plants. The yam chitinase, however, has an additional sequence of 8 amino acids (a C-terminal extension) following the cysteine that was reported as the last amino acid for other class IV chitinases; this extension is perhaps involved in subcellular localization. A homology model based on the structure of a class II chitinase from barley was used as an aid to interpreting the available data. The analysis suggests that the class IV enzyme recognizes an even shorter segment of the substrate than class I or II enzymes. This observation might help to explain why class IV enzymes are better suited to attack against pathogen cell walls.  相似文献

1 Diversity patterns of small mammals were studied along an elevational transect on Mount Kinabalu, the highest mountain in South‐east Asia, utilizing data from previously existing sources and a new field study. A mark‐and‐release study (conducted during wet and dry seasons between November 1994 and April 1995) resulted in captures of 12 small mammal species, including two species of squirrels, two tree shrews, seven murid rodents and one gymnure. 2 Based on data compiled from this survey, museum specimens, and published and unpublished literature (analysed by locally weighted sums of squares and quadratic polynomial regressions), species richness of small mammals formed a middle elevation bulge, highest at about 1200–1400 m and declining at lower and higher elevations. Trapping during two seasons did not change the assessment of the pattern. 3 A cluster analysis of these data indicated that there are two elevationally associated faunas, one in the highlands and another in the lowlands. The transition between these two assemblages is at 1700–1800 m elevation. The lowland faunal assemblage has the highest number of species, with maximum species richness at about 1300 m for total small mammal species, about 1200 m for arboreal species and about 1400 m for terrestrial species. 4 The areas where much overlapping of species occurs are the elevations where climate and vegetation change rapidly from lowland to montane types. Tree species, gymnosperms, orchids and ferns showed a similar curvilinear pattern along the same elevational gradient, with maximum species richness at about 1400–1500 m. Temperature declined progressively with increasing elevation, but rainfall and humidity reached their highest levels at about 1700 m. 5 Maximum diversity of small mammals thus occurred at the elevation where a highland and a lowland assemblage overlapped, where several types of plants reached their maximum diversity, and where rainfall and humidity reached their maxima. Similar patterns have been documented for small mammals, plants, and climate at sites scattered in Indo‐Australia from Taiwan to New Guinea.  相似文献

An experimental method is proposed for detecting the effects of positive natural selection on DNA polymorphisms. Since beneficial mutations are expected to increase in frequency faster than neutral mutations, variants which have reached high frequencies in a relatively short period could be linked to some beneficial mutation. D. melanogaster has a cosmopolitan polymorphic inversion -In(2L)t- whose age in some local populations has been estimated. Setting the age of In(2L)t as the upper limit for the age of variants, we searched for variants whose frequencies were possibly influenced by positive natural selection. We detected a single candidate whose frequency and distribution met the requirements imposed by our method.  相似文献
