Asparagus officinalis seedlings treated with either thiocarbamatesor carbamates are induced to flower within 2-3 weeks of application.SDS-PAGE and 2-D PAGE resolutions of total soluble proteinsof treated seedlings showed the accumulation of an acidic 17kDa polypeptide. Partialmino acid sequence of the acidic proteinshowed homology to the wound-responsive A. officinalisPR 1,AoPRl. We named this protein EFP, early flowering protein, owingto its advanced appearance during the early transition fromvegetative to floral phase. Carbamates and thiocarbamates thatinduced more than 80% flowering rates corresponded with increasedlevel of EFP while compounds that are weak flower inducers showedreduced amount of EFP. A new TEMPO-mediated oxidation with catalytic amounts of TEMPO and NaClO, and NaClO2 as the primary oxidant under aqueous conditions at pH 3.5–6.8 was used to prepare water-soluble β-(1 → 4)-linked polyglucuronic acid Na salts (cellouronic acids, CUAs) with high molecular weight in good yield. When regenerated cellulose with original degree of polymerization (DP) of 680 was oxidized by the 4-acetamide-TEMPO/NaClO/NaClO2 system at pH 5.8 and 40 °C for 3 days, CUA with weight average DP (DPw) of 490 was obtained quantitatively. No peaks other than six signals from β-(1 → 4)-linked anhydroglucuronic acid units of CUA were detected in the solution-state 13C NMR spectra of the oxidized products. Although the oxidized product prepared under the above conditions contained about 20% unoxidized cellulose particles, the non-CUA fraction was separable from CUAs by filtration or salt precipitation. The DPw values and yields of CUAs after the filtration or salt precipitation treatment were 250–380 and 45–71%, respectively. 相似文献

The germination of spores of the endophytic brown algae Laminariocolax aecidioides and Laminarionema elsbetiae (Ectocarpales sensu lato) on their host Laminaria saccharina was studied using scanning electron microscopy. Zoids were the infective units in both taxa. Despite a large difference in size between spores of the species studied, the initial steps of infection were similar. Zoids settled on the host surface, and during secretion of adhesive material from their anterior end, the spores became elongated to rod-shaped and stood erect on the host. A single germ tube developed at the proximal end and penetrated the intact surface of the host. Sharp edges around the entrance hole and the absence of inward deformation of the host surface around the settled zoids suggest an enzymatic rather than a mechanical penetration mechanism. 相似文献

Genes encoding 2-deoxy-d-ribose-5-phosphate aldolase (DERA) homologues from two hyperthermophiles, the archaeon Pyrobaculum aerophilum and the bacterium Thermotoga maritima, were expressed individually in Escherichia coli, after which the structures and activities of the enzymes produced were characterized and compared with those of E. coli DERA. To our surprise, the two hyperthermophilic DERAs showed much greater catalysis of sequential aldol condensation using three acetaldehydes as substrates than the E. coli enzyme, even at a low temperature (25 degrees C), although both enzymes showed much less 2-deoxy-d-ribose-5-phosphate synthetic activity. Both the enzymes were highly resistant to high concentrations of acetaldehyde and retained about 50% of their initial activities after a 20-h exposure to 300 mM acetaldehyde at 25 degrees C, whereas the E. coli DERA was almost completely inactivated after a 2-h exposure under the same conditions. The structure of the P. aerophilum DERA was determined by X-ray crystallography to a resolution of 2.0 A. The main chain coordinate of the P. aerophilum enzyme monomer was quite similar to those of the T. maritima and E. coli enzymes, whose crystal structures have already been solved. However, the quaternary structure of the hyperthermophilic enzymes was totally different from that of the E. coli DERA. The areas of the subunit-subunit interface in the dimer of the hyperthermophilic enzymes are much larger than that of the E. coli enzyme. This promotes the formation of the unique dimeric structure and strengthens the hydrophobic intersubunit interactions. The ligation reaction is widely used in molecular biology. There are several kits available that complete the ligation reaction
very rapidly but they are rather expensive. In this study, we successfully modified the ligation buffer with much lower cost
than existing kits. The ligation reaction can be completed in 10 min using very low activities such as 0.01 U T4 DNA ligase,
and costs only $1 for 100 reactions of 20 μl scale. We name this ligation system the “Coffee Break Ligation” system; one can
complete ligation reaction while drinking a cup of coffee, and perform 100 reactions by spending money equivalent to a cup

Cu in blood has been believed to transport into cell via albumin and some amino acids. To shed light on the Cu transport process we studied the reaction of the Cu(II)-peptide with the amino acid by absorption and CD spectra. Albumin mimic peptides GlyGly-L-HisGly (GGHG) and penta-Gly(G5) formed stable 4N coordinated Cu(II) complexes, but in the reaction with histidine (His) and penicillamine (Pes) the ternary Cu(II) complex formations were observed different by the kinetic study. Cu(II)-G5 complexes reacted with Pes to form the ternary complex Cu(H(-1)G5)(Pes(-)) which was subsequently transformed to the binary complex Cu(Pes(-))(2). In the system with GGHG the Cu(II) was also transported from GGHG to Pes, but the ternary Cu(H(-1)GGHG)(Pes(-)) complex as the intermediate was detected a trace. The ternary complex would be spontaneously transformed to Cu(Pes(-))(2) upon forming, because the rate constant of the ternary complex formation k(1+)= approximately 2M(-1)s(-1) was less than k(2+)= approximately 5 x 10(2)M(-1)s(-1) for the Cu(Pes(-))(2) formation at physiological pH. In the Cu(II)-GGHG-His system the ternary Cu(H(-1)GGHG)(His) complex was also hardly identified because the formation constant K(1) and k(1+) were very small and the equilibrium existed between Cu(H(-2)GGHG) and Cu(His)(2) and its overall equilibrium constant beta(2) for Cu(His)(2) was very small to be 1.00+/-0.05 M(-1) at pH 9.0. These results indicated that the ternary complex is formed in the Cu transport process from the albumin to the amino acid, but His imidazole nitrogen in the fourth-binding site of Cu(II) strongly resists the replacement by the incoming ligand. 相似文献

This study undertook the analysis of tissue cadmium (Cd) distribution using in-air micro-particle-induced X-ray emission (PIXE)
and the examination of the involvement of metal ions in parenteral Cd toxicity. A mouse was injected intraperitoneally with
3 mg/kg body weight of CdCl2 thrice weekly. After 27 wk, the liver and kidney were excised and fixed in 10% formalin solution for 4 h and then embedded
in paraffin. Thin paraffin sections were used to analyze trace elements with in-air micro-PIXE and to examine metallothionein
protein and histological changes. Cd distribution was determined by micro-PIXE in the liver and renal cortex of the Cd-exposed
mouse, and the net Cd count was higher in the liver than in the renal cortex. The net iron (Fe) count was higher in the liver
of the Cd-exposed mouse compared to the control, and an opposite tendency was observed in the renal cortex. Wide cellular
Cd distribution was demonstrated in the liver and renal cortex of the chronic Cd-exposed mouse compared to the control. Metallothionein
staining was increased by chronic exposure to Cd both in the liver and kidney, and nephrotoxicity was more apparent than hepatotoxicity.
The modification of tissue Fe and calcium distribution by an intraperitoneal injection of Cd might be involved in Cd-induced
