The gene locus of Machado-Joseph disease (MJD) has recently been mapped within a 29-cM subregion of 14q chromosome. We did a linkage study of 24 multigenerational MJD Japanese pedigrees, in an attempt to narrow the candidate region of this gene. Pairwise and multipoint linkage analysis, together with haplotype segregation analysis, led to the conclusion that the MJD gene is located at the 6.8-cM interval between D14S256 and D14S81 (Zmax = 24.78, multipoint linkage analysis). D14S291 and D14S280, located at the center of this interval, showed no obligate recombination with the MJD gene (Zmax = 5.93 for D14S291 and 9.99 for D14S280). A weak, but significant, linkage disequilibrium of MJD gene was noted with D14S81 (P < .05) but not with D14S291 or D14S280. These results suggest that a 3.6-cM interval flanked by D14S291/D14S280 and D14S81 is the most likely location of the MJD gene and that it is closest to D14S81.  相似文献

Schizophyllan exists in aqueous solution as a triple helix, which is intact at room temperature. Its aqueous solution forms some ordered structure at low temperatures but undergoes a sharp transition to a disordered structure as the temperature is raised. The transition temperature Tc is about 7 and 18°C for H2O and D2O solutions, respectively. This transition was followed by time-domain reflectometry to investigate dynamic aspects of the transition. In addition to a major peak around 10 GHz, the dielectric dispersion curve of a 20 wt % schizophyllan in D2O exhibited a small peak around 100 MHz below Tc and around 10 MHz above Tc. The major peak is due to bulk water, whereas the 100 MHz peak is assigned to “bound” or “structured” water, and that around 10 MHz to side-chain glucose residues. However, unlike usual bound water reported for biopolymer solutions, this “structured” water disappears abruptly when the temperature becomes close to Tc without accompanying a conformational transition of the main chain. The above assignment is consistent with the structure of the ordered phase derived from previous static data that it consists of side-chain glucose residues along with nearby water molecules surrounding the helix core that are interacting with each other loosely through hydrogen bonds, and spreads radially only a layer of one or two water molecules but a long distance along the helix axis. © 1995 John Wiley & Sons, Inc.  相似文献

A colorless mutant of Chlorella kessleri, grown in darknessin a medium that contained nitrate and glucose, took up ammoniamore efficiently than nitrate. Irradiation with blue light inhibitedthe uptake of ammonia but, conversely, the uptake of nitratewas enhanced by blue light. These effects were not observedunder illumination with red or far-red light. The inhibitionby blue light of the uptake of ammonia was abolished in thepresence of nitrate. The charge compensation for the uptakeof ammonia was achieved by the immediate release of potassiumions and this release was followed by release of protons, therate of the latter process being strongly reduced by blue light. Changes in intracellular levels of Chl a precursors were examinedin relation to changes in the PSI/PSII stoichiometry in thecyanophyte Synechocystis PCC 6714. Protochlorophyllide (Pchlide)accumulated markedly in cells with a low PSI/PSII stoichiometrygrown under light that is absorbed by Chl a (PSI light) whereasno accumulation occurred in cells with a high PSI/PSII stoichiometrygrown under light absorbed by phycobilisomes (PSII light). Levelsof Pchlide in cells grown under PSI light decreased rapidlyupon a shift to PSII light. The rapid decrease in Pchlide accompanieda transient increase in chlorophyllide a, indicating that reductionof Pchlide was enhanced by shift to PSII light. The action spectrumindicated that the Pchlide decrease upon the shift to PSII lightdepended on excitation of Pchlide, suggesting that the accumulationof Pchllide was due to limited excitation of Pchlide, so thatPchlide photoreduction, under PSI light. However, comparisonof levels of Pchlide and the photosystem complexes in wild-typePlectonema boryanum with those in a mutant that lacked the darkPchlide reductase (YFC 1004) indicated that dark reduction compensatedfor the limited photoreduction under PSI light. Similar compensationby dark reduction was confirmed with Synechocystis PCC 6714.In cultures of Synechocystis under conditions where Pchlidecould not be photoreduced, accumulation of Pchlide and low PSI/PSIIstoichiometry occurred only when cells were illuminated withlight that preferentially excited PSI. The results indicatethat the low PSI/PSII stoichiometry in cells grown under PSIlight is not a result of inefficient synthesis of Chl a witha reduced rate of Pchlide photoreduction. Abstract In vivo induction of cytokines by a monoclonal antibody (mAb) against T-cell receptor (TCR) αβ and the protective effect induced by the mAb on a lethal infection with Listeria monocytogenes were studied. Injection of anti-TCR αβ mAb induced rapid production of endogenous tumour necrosis factor in the spleens, and gamma interferon and interleukin-6 in the bloodstreams and spleens of mice. Administration of anti-CD4 mAb, anti-CD8 mAb, or anti-Thy1.2 mAb resulted in suppression of anti-TCR αβ mAb-induced endogenous cytokine production. Mice were protected against lethal L. monocytogenes infection when treated with anti-TCR αβ mAb. The protective effect was not demonstrated in CD4 + cell- or CD8 + cell-depleted mice. These results suggest that anti-TCR αβ mAb shows a protective effect on a lethal infection with L. monocytogenes in mice and that the mAb-induced endogenous cytokines might be involved in the effect of anti-TCR αβ mAb.  相似文献

The purpose of this study was to clarify the effect of grazing exclosures on the recovery and rehabilitation of overgrazed steppe vegetation on varying slope aspects in the Loess Plateau of northwest China. The annual precipitation in the area studied was 400–480 mm. Soil samples were taken on nine slopes in the five-year exclosure and on five slopes outside the exclosure after a vegetation survey; they were then analyzed chemically. Mean number of species recorded per 0.25 m2 was lower on the south-facing slope than all other slopes. The reverse trend was observed for aerial biomass. Species diversity estimated by information content was higher in the grazing zone than in a 3200-ha protected zone within an exclosure. From species ordination by principal component analysis, species with lower coverage in the grazing zone were Poa sphondylodes, Roegneria purpurascens, Hierochloe odorata, and Potentilla bifurca, which are all recognized as indicator species for rehabilitation efforts. In the soil surface layer, calcium contents were low, and the total contents of carbon and nitrogen were high on the north-facing slope in the exclosure. The protection by exclosure of overgrazed steppe was seen to be effective because the accumulation of soil organic matter increased and water balance improved.  相似文献

A murine monoclonal antibody recognizing (1→6)-β-d -glucopyranosyl laminaritriose (G4) was prepared by immunizing BALB/c mice with G4-bovine serum albumin conjugate and fusing the splenocytes with mouse myeloma cells. The monoclonal antibody (IgM) provoked by the cloned cells showed low reactivity with schizophyllan, an antitumor polysaccharide, but notable reactivity with some low-molecular-weight schizophyllans. This antibody was useful for determination of the epitope of several polysaccharides. The extent of reactivity of this monoclonal antibody was related only to the molecular weight of schizophyllan.  相似文献

We compared the vegetation structure, rodent density and seed loss rate between protected and disturbed sites affected from grazing by cattle, goats and sheep, in the Thar desert of India. A perennial tussocky grassLasiurus sindicus Hent. was largely dominant in the protected site, whileL. sindicus was rare and replaced by undershrub speciesAerva pseudotomentosa Blatt. & Halb. andCrotalaria burbira Buch.-Ham. in the overgrazed site. In the grazed site, plant coverage was low, but the density of rodent burrows and the frequency of rodent captures were significantly high as compared to the protected site. Corresponding with the density of desert rodents, seed predation was significantly higher in the grazed site than in the protected site. These results suggest that overgrazing by large mammals has strong effects on plant succession by altering not only the species composition and abundance of plant community, but also the habitat suitability for seed-eating rodents.  相似文献

ThePLC1 gene of the yeastSaccharomyces cerevisiae has been discovered to encode a homolog of mammalian phosphoinositide-specific phospholipase C (PLC). Five temperature-sensitiveplc1 mutants were isolated by in vitro mutagenesis with subsequent plasmid shuffling. All of the amino acid substitutions that caused a temperature-sensitive growth phenotype were located in the X or the Y region, both of which are conserved among PLC isoenzymes. The PLC activity of all products of mutantplc1 genes was dramatically lower than that of the wild-type product, indicating that PLC activity itself is important for cell growth. At the restrictive temperature,plc1 mutant cells ceased growth at random times during the cell cycle, a result that suggests thatPLC1 is required at several or all stages of the cell cycle.  相似文献
