The localization of ribulose 1,5-bisphosphate carboxylase/oxygenase(RuBisCO) in chloroplasts of the green alga Bryopsis maximawas examined by immunological techniques. Three strains of hybridomaswere established between myeloma cells and the spleen cellsfrom mouse immunized against B. maxima RuBisCO. The antibodiesreacted with the large subunit of B. maxima RuBisCO but notwith spinach RuBisCO. Immunofluorescence and immunoenzymaticstudies showed that the large subunit of B. maxima RuBisCO wasconcentrated in pyrenoids and on the surface of starch grainssurrounding the pyrenoids. Nitrogenous inhibitors of respiration, namely, KCN, NaN3 andNH2OH, which promote the germination of cocklebur (Xanthiumpennsylvanücum Wallr.) seeds, enhanced the accumulationof cyanogenic compounds. Urea and cyanamide, which were ineffective,did not. Part of the exogenously applied KCN was converted tocyanogenic glycosides and lipids, but these compounds were onlyproduced at low levels in NaN3-treated seeds. Exposure to NH2OHcaused a slight accumulation of both types of cyanogenic compounds.Thiourea, effective in stimulating germination, did not increasethe cyanogenic compounds, which suggests that the mechanismof its effect on germination is different from that of othernitrogenous compounds. Thiocyanate had a slightly promotiveeffect and caused minor increases in the levels of cyanogeniccompounds. Ethylene increased the metabolic utilization of the accumulatedcyanogens in response to KCN or NaN3. The response to ethyleneof seeds in secondary dormancy was restored only after pre-treatmentwith KCN or NaN3. The response occurred once the level of cyanogenshad increased. By contrast, enhancement of the effects of KCNand NaN3 on germination by propyl gallate or benzohydroxamate,inhibitors of CN-resistant respiration, was accompanied by theincreased accumulation of cyanogens within the seeds that hadbeen exposed to KCN or NaN3 in combination with the other typesof respiratory inhibitors. A polymerase chain reaction (PCR) method was performed for rapid and sensitive detection of pathogenic Vibrio trachuri isolated from cultured Japanese horse mackerel. A set of primers was selected from the base sequence of the Pst I fragment of T9210 chromosomal DNA and used for PCR detection of T9210. This PCR specifically amplified the DNAs from V. trachuri T9210, T9213, and T9216 but not of those other bacterial strains. PCR using a Pst I-1 primer set made it possible to detect 100 fg of T9210 DNA. The PCR method reported here may be useful for detection and identification of V. trachuri pathogenic to Japanese horse mackerel.  相似文献

The colH gene encoding 116-kDa collagenase of Clostridium histolyticum (cColH) was cloned into an Escherichia coli-Bacillus subtilis shuttle vector to develop a method for purification of recombinant collagenase (rColH). When plasmid pJCM310 containing the colH gene was introduced into B. subtilis DB104 and the transformant was grown in LB broth at 37 C, stability of the plasmid was not maintained. However, stability was partly improved by growing the transformant in a modified LB broth containing 0.5 M sodium succinate with gentle shaking at 35 C. When the transformant was grown to an optical density of 0.4 at 600 nm in this medium, pJCM310 was stable and rColH was produced in sufficient amounts. rColH was purified to homogeneity by ammonium sulfate precipitation, gel filtration and ion-exchange chromatography. The yield of rColH from an 800-ml culture was 0.53 mg and its specific activity was estimated to be 1,210 U per mg of protein. The purified rColH was capable of degrading native type-I collagen fibril from bovine achilles tendon, as was demonstrated by zymography. A comparison of the N-terminal amino acid sequence between cColH and rColH revealed that rColH has 10 extra N-terminal amino acid residues. However, the peptide mapping of rColH with V8 protease was virtually identical to that of cColH. Furthermore, the molecular mass of rColH was estimated to be 112,999 Da by mass spectrometry, coinciding with the value of 112,977 Da, which was predicted from the nucleotide sequence of the colH gene. Therefore, the recombinant B. subtilis culture is capable of serving as a useful source for enzyme purification.  相似文献

TRK-100 is a chemically stable analogue of prostacyclin and effective in inhibiting platelet aggregation when orally administered in experimental animals. In the present study we compared the potency of TRK-100 with those of PGI2 and PGE1 to cause an activation of adenylate cyclase activity in rat and human platelet membranes. TRK-100 was half as effective as PGI2, and 10 times more effective than PGE1 in both platelet membranes. TRK-100 also induced an activation of phosphodiesterase activity when directly added to intact platelets probably as a feedback mechanism of intracellular cAMP level like PGI2 did. TRK-100 would mimic PGI2 in the regulation of cAMP metabolism.  相似文献

Growth of the Bacillus subtilis wild type strain was severelyinhibited by mannosamine. This inhibitory effect was reversedby the addition of glucose or mannose. Growth in peptone mediumwas accompanied by the consumption of the added glucose andby the accumulation of acetoin. Glucose utilization and acetoinproduction were markedly repressed in the cells grown in thepresence of mannosamine. Growth in the presence of mannosaminewas accompanied by a relative decrease in a protein with themolecular weight of 70,000 as revealed by SDS polyacrylamidegel electrophoresis. Our results indicate that mannosamine'sinhibition of growth is due primarily to low glucose utilizationalthough other cellular metabolic activity also may be involved. A new unicellular red alga, Corynoplastis japonica gen. et sp. nov., is described from Tobishima, Japan. Cells are spherical, 18–33 µm in diameter, pale purple to brownish red and surrounded by a mucilaginous sheath. A single chloroplast with many lobes extends from the cell periphery to the cell center. A peripheral thylakoid is present. A pyrenoid occurs at each innermost chloroplast lobe end and one or two thylakoids are present in the pyrenoid matrix. The nucleus is eccentric to peripheral and Golgi bodies are scattered throughout the cell and associated with endoplasmic reticulum. Cells have a slow random gliding motility. The low molecular weight carbohydrate mannitol is present in the cells. Molecular phylogenetic analysis indicates that this alga is closely related to members of the genus Rhodella. A new order, Dixoniellales, is established for Dixoniella, Neorhodella and Glaucosphaera based on molecular and ultrastructural evidence (Golgi bodies associated only with the nucleus). The redefined order Rhodellales in which Rhodella and Corynoplastis are placed is characterized ultrastructurally by Golgi bodies scattered throughout the cytoplasm and associated with endoplasmic reticulum.  相似文献
