Flax-retting by polygalacturonase-containing enzyme mixtures and effects on fiber properties 总被引:5,自引:0,他引:5
Enzyme-retting of flax was accomplished via individual treatment with four polygalacturonase (PGase) containing solutions of various fungal sources and the resulting fibers were characterized. The retting solutions were equilibrated to contain 2.19 U of PGase activity as determined via a dinitrosalicylic acid (DNS) reducing sugar assay. As compared with the buffer control, treatment with the various enzyme solutions increased the yield of fine fibers. Treatment with Aspergillus niger PGase resulted in a 62% increase in fine fiber yield as compared with the buffer control and fiber strength did not statistically differ (P=0.05) between these treatments. Retting via PGases of Rhizopus origin produced the weakest fibers. These results illustrate that the crude PGases differ in their ability to ret flax and that under the defined experimental conditions the A. niger PGase is a better retting agent. Light microscopy demonstrated the ability of all enzymes to separate fiber from shive and epidermal tissues. Enzyme profiles of the solutions were determined via viscometric assays. Pectinolytic activity was the predominant activity of all enzymes tested. Activity against carboxymethyl cellulose (CMC) was a minor component of all solutions except A. niger PGase for which no activity was detected. 相似文献

Sixteen tooth characters were analyzed in 277 specimens of spinner dolphins ( Stenella longirostris ) from the eastern tropical Pacific to determine whether the characters were associated with size of the animal, species-stock membership or geographic location. The tooth characters included 5 continuous measurements and 11 qualitative variables. Statistical analyses of the characters included two-way ANOVA tests which assessed continuous measurement variables for differences based on stock membership, latitude and longitude of collection or whether the animal was collected north or south of the equator. Principal component analyses combined factors of continuous variables with those of qualitative variables into components which were used as variables in further analyses. Discriminant analyses were performed using continuous variables alone and using principal components as potentially discriminating variables. Results showed that tooth differences were not associated with size but significant differences were found in some tooth variables with stock membership, incremental changes in latitude and whether the animal was collected north or south of the equator. A discriminant function, containing only tooth length, correctly classified 67% of northern and 78% of southern animals according to location of collection north or south of the equator. 相似文献

Production of ligninolytic enzymes and degradation of 14C-ring labeled synthetic lignin by the white-rot fungus Cyathus stercoreus ATCC 36910 were determined under a variety of conditions. The highest mineralization rate for 14C dehydrogenative polymerizates (DHP; 38% 14CO2 after 30 days) occurred with 1 mM ammonium tartrate as nitrogen source and 1% glucose as additional carbon source, but levels
of extracellular laccase and manganese peroxidase (MnP) were low. In contrast, 10 mM ammonium tartrate with 1% glucose gave
low mineralization rates (10% 14CO2 after 30 days) but higher levels of laccase and manganese peroxidase. Lignin peroxidase was not produced by C. stercoreus under any of the studied conditions. Mn(II) at 11 ppm gave a higher rate of 14C DHP mineralization than 0.3 or 40 ppm, but the highest manganese peroxidase level was obtained with Mn(II) at 40 ppm. Cultivation
in aerated static flasks gave rise to higher levels of both laccase and manganese peroxidase compared to the levels in shake
cultures. 3,4-Dimethoxycinnamic acid at 500 μM concentration was the most effective inducer of laccase of those tested. The
purified laccase was a monomeric glycoprotein having an apparent molecular mass of 70 kDa, as determined by calibrated gel
filtration chromatography. The pH optimum and isoelectric point of the purified laccase were 4.8 and 3.5, respectively. The
N-terminal amino acid sequence of C. stercoreus laccase showed close homology to the N-terminal sequences determined from other basidiomycete laccases. Information on C. stercoreus, whose habitat and physiological requirements for lignin degradation differ from many other white-rot fungi, expands the
possibilities for industrial application of biological systems for lignin degradation and removal in biopulping and biobleaching
processes.

Bermudagrass (Cynodon dactylon) leaf blades and whole cordgrass (Spartina alterniflora) fiber were evaluated for degradation of cell walls by microbial groups in ruminal fluid. The groups were selected by the addition of antibiotics to the inoculum as follows: (i) whole ruminal fluid (WRF), no antibiotics; (ii) cycloheximide (C) to inhibit fungi, thus showing potential bacterial activity; (iii) streptomycin and penicillin (S,P) to inhibit fiber-degrading bacteria, showing potential fungal activity; (iv) streptomycin, penicillin, and chloramphenicol (S,P,CAM) to inhibit all bacteria including methanogens; (v) streptomycin, penicillin, and cycloheximide (S,P,C) to inhibit all microbial activity as a control; and (vi) autoclaved ruminal fluid (ARF) to inhibit all biological activity as a second control. Scanning electron microscopy of tissue degradation indicated that tissues not giving a positive histological reaction for lignin were more readily degraded. Cordgrass was more highly lignified, with more tissues resisting degradation than in bermudagrass. Patterns of degradation due to treatment resulted in three distinct groups of data based on the extent of fiber or component losses: WRF and C greater than S,P and S,P,CAM greater than S,P,C and ARF. Therefore, bacterial activity was responsible for most of the fiber loss. Fiber degradation by anaerobic fungi was significantly less (P = 0.05). Cupric oxide oxidation of undigested and digested bermudagrass fiber indicated that phenolic constituents differed in their order of resistance to removal or solubilization. Ghrelin (G-HH) synthesized in several tissues including salivary and stomach glands stimulates appetite in humans by modulating neuropeptide Y neurons in the hypothalamic arcuate nucleus. Loss of appetite is one of the most important symptoms of stomach cancer. We conducted a study using immunohistochemistry to determine whether salivary glands and stomach cancer tissues produce ghrelin. We determined that negative ghrelin immunohistochemistry discriminates tumors from normal tissues and may therefore further our understanding of the clinically important problem of reduced food intake and anorexia in cancer patients. Most of the biological, chemical or physical agents that cause cell death in certain doses and time of exposure may induce either apoptosis or necrosis. This study explores in what ways the genotoxic, cytotoxic and apoptotic effects of diethylstilbestrol (DES), a chemical agent currently used in the treatment of various types of cancer, on the human lymphocytes depend upon the dose and the exposure time. For this purpose, firstly it aims to determine in what dosages and durations of DES treatment, genotoxicity and cytotoxicity in human lymphocytes occur in vitro. Secondly, it explores the effects of DES on sister-chromatid exchanges (SCEs) and apoptosis and their relation with the nitric oxide (NO) levels. Finally, it investigates whether different dosages of DES and duration of treatment with it are correlated with each other. In so doing, we investigated the relationship among the viability, necrosis and apoptosis rates of human lymphocytes which were treated with five different DES concentrations (1, 5, 10, 15 and 20 μM) for 24, 48 and 72 h, DNA fragmentation analysis of these cells, their mean SCE values and NO levels. We concluded that 5 μM DES at 24 h is the most effective dosage that induces typical features of apoptosis in human lymphocytes. Despite the fact that there are many other studies on the effects of DES on the cancer cells, we thought it might be worth looking into the effects of DES on human lymphocytes in vitro.
