Summary Many conclusions concerning cell culture mycoplasmas are based on data from studies in fibroblast cultures. Some conclusions may not be valid in other types of differentiated cell cultures.M. salivarium was isolated from 35 human lymphocyte cultures (HLC), 34 from the same laboratory. The organism grew to more than 108 colony forming units (CFU) per ml of lymphocyte suspensions and was readily detectable by microbiological culture, uridine phosphorylase, and uridine/uracil assays. Direct mycoplasmal assays on HLC by DNA fluorochrome staining and scanning electron microscopy (SEM) yielded artifacts that interfered with diagnosis. For DNA and SEM of HLC, inoculation into indicator cell cultures is recommended.M. salivarium infection of HLC did not produce any immediate difference in growth rates; however, infected cultures eventually died 14 to 29 passages after infection in contrast to uninfected controls. The same organism in 3T6 fibroblasts effected a 60% decrease in growth rate. AlthoughM. salivarium is a frequent isolate from the oral cavity, it is a rare cell culture isolate.M. salivarium was able to initiate growth over a wide pH range, grew as well in cell cultures as in cell-free media, and was resistant to 50 μg per ml of gentamycin, tylocine, kanamycin, and erythromycin. By C0t1/2 analysis,M. salivarium had a genomic molecular weight of 4.2×108 daltons.M. salivarium did not increase chromosome aberrations in one HLC. Some of these results have application to infection of HLC by other mycoplasmal species. Retroviral RNA genome (gRNA) harbors cis-acting sequences that facilitate its specific packaging from a pool of other viral and cellular RNAs by binding with high-affinity to the viral Gag protein during virus assembly. However, the molecular intricacies involved during selective gRNA packaging are poorly understood. Binding and footprinting assays on mouse mammary tumor virus (MMTV) gRNA with purified Pr77Gag along with in cell gRNA packaging study identified two Pr77Gag binding sites constituting critical, non-redundant packaging signals. These included: a purine loop in a bifurcated stem-loop containing the gRNA dimerization initiation site, and the primer binding site (PBS). Despite these sites being present on both unspliced and spliced RNAs, Pr77Gag specifically bound to unspliced RNA, since only that could adopt the native bifurcated stem–loop structure containing looped purines. These results map minimum structural elements required to initiate MMTV gRNA packaging, distinguishing features that are conserved amongst divergent retroviruses from those perhaps unique to MMTV. Unlike purine-rich motifs frequently associated with packaging signals, direct involvement of PBS in gRNA packaging has not been documented in retroviruses. These results enhance our understanding of retroviral gRNA packaging/assembly, making it not only a target for novel therapeutic interventions, but also development of safer gene therapy vectors.  相似文献

Ternary copper(II) complex [Cu(TAN)(O2CMe)] (1), where H-TAN is 1-(2-thiazolylazo)-2-naphthol, is prepared and structurally characterized by X-ray crystallography. The complex has a distorted square pyramidal (4+1) CuN2O3 coordination geometry with the acetate showing chelating axial-equatorial binding mode and TAN as a tridentate ligand bonded to the metal in the basal plane. Complex 1 is one-electron paramagnetic and displays ligand-to-metal charge transfer bands at 575 and 398 nm in dimethylformamide. The reactions of 1 with bases (B) like 1,10-phenanthroline (phen) and kanamycin-A (kan-A) afford ternary complexes of formulation [Cu(TAN)B]+ (B=phen, 2; kan-A, 3) under in situ reaction conditions. Complexes 2 and 3, prepared to explore their DNA binding and photo-induced DNA cleavage activity, display good binding propensity to calf thymus (CT) DNA giving a relative order: 2-3>1. The apparent binding constant (Kapp) for 1 is determined as 9.8 x 10(5)M(-1) from fluorescence quenching experiments using ethidium bromide. The quenching constants (K) values of 1-3, obtained from the Stern-Volmer plots, are 0.28, 0.52 and 0.49, respectively. All the complexes show photo-induced DNA cleavage activity when irradiated with a monochromatic UV light of 365 nm wavelength. A 200 microM complex 1 cleaves approximately 75% supercoiled (SC) DNA on 2h exposure time at 365 nm. A 50 microM solution of 1 in presence of 100 microM phen and kanamycin-A cleaves approximately 99% and approximately 60% SC DNA to its nicked circular form, respectively, for an exposure of 30 min. The complexes also exhibit significant cleavage of SC DNA on irradiation with visible light of wavelengths 532, 575 and 632.8 nm. Control experiments reveal the minor groove binding nature of the complexes. The cleavage reactions involve the formation reactive hydroxyl species as significant inhibition in the presence of dimethyl sulfoxide (DMSO) and catalase is observed. There is no apparent inhibition in cleavage in the presence of singlet oxygen quenchers like sodium azide. The cleavage activity has been found to be higher at the CT band position of 575 nm in comparison to those at 532 and 632.8 nm. The results indicate the involvement of the CT band in the photo-excitation process.  相似文献

Ternary cobalt(III) complexes [CoL(B)] (1-3) of a trianionic tetradentate phenolate-based ligand (L) and phenanthroline bases (B), viz. 1,10-phenanthroline (phen in 1), dipyridoquinoxaline (dpq in 2) and dipyridophenazine (dppz in 3) are synthesized, characterized from X-ray crystallographic, analytical and spectral techniques, and their utility in photodynamic therapy (PDT) of thyroid diseases caused by TSH receptor dysfunction is probed. The complexes display a visible spectral band within the PDT spectral window at ~690 nm. Photodynamic potential was estimated through DNA cleavage activity of the dpq and dppz complexes in UV-A light of 365 nm and red light of 676 nm. The reactions proceed via the hydroxyl radical pathway. The complexes retain their DNA photocleavage activity in red light under anaerobic conditions, a situation normally prevails in hypoxic tumor core. Investigation into the photocytotoxic potential of these complexes showed that the dppz complex 3 is approximately 4-fold more active in the HEK293 cells expressing human thyrotropin receptor (HEK293-hTSHR) than in the parental cell line and has an insignificant effect on an unrelated human cervical carcinoma cell line (HeLa). Photoexcitation of complex 3 in HEK293-hTSHR cells leads to damage hTSHR as evidenced from the decrease in cAMP formation both in absence and presence of hTSH and decrease in the TSHR immunofluorescence with a concomitant cytoplasmic translocation of the membrane protein, cadherin. The involvement of hTSHR is evidenced from the ability of complex 3 to bind to the extracellular domain of hTSHR (hTSHR-ECD) with a K(d) value of 81 nM and from the photocleavage of hTSHR-ECD.  相似文献

We studied thisphenomenon and its physiological basis in eight New Zealand Whiterabbits that were anesthetized, paralyzed, and mechanically ventilatedat a fixed minute ventilation. Isovolemic anemia was induced bysimultaneous blood withdrawal and infusion of 6% hetastarch insequential stages; exchange transfusions ranged from 15-30 ml involume. Variables measured after each hemodilution included hematocrit(Hct), arterial and venous blood gases, mixed expiredPCO2 andPO2, and blood pressure; also, O2 consumption,CO2 production, cardiac output(), and physiological dead space were calculated.Data were analyzed by comparison of changes in variables with changesin Hct and by using the model of capillary gas exchange described byBidani (J. Appl. Physiol. 70:1686-1699, 1991). There was complete compensation for anemia withstability of venous and arterial PCO2between Hct values of 36 ± 3 and 12 ± 1%, which was predictedby the mathematical model. Over this range of hemodilution, rose 50%, and theO2 extraction ratio increased 61%without a decline in CO2production or a rise in alveolar ventilation. The dominantcompensations maintaining CO2transport in normovolemic anemia include an increased and an augmented Haldane effect arising from theaccompanying greater O2extraction.

Docking and fusion of single proteoliposomes reconstituted with full-length v-SNAREs (synaptobrevin) into planar lipid bilayers containing binary t-SNAREs (anchored syntaxin associated with SNAP25) was observed in real time by wide-field fluorescence microscopy. This enabled separate measurement of the docking rate k(dock) and the unimolecular fusion rate k(fus). On low t-SNARE-density bilayers at 37 degrees C, docking is efficient: k(dock) = 2.2 x 10(7) M(-1) s(-1), approximately 40% of the estimated diffusion limited rate. Full vesicle fusion is observed as a prompt increase in fluorescence intensity from labeled lipids, immediately followed by outward radial diffusion (D(lipid) = 0.6 microm2 s(-1)); approximately 80% of the docked vesicles fuse promptly as a homogeneous subpopulation with k(fus) = 40 +/- 15 s(-1) (tau(fus) = 25 ms). This is 10(3)-10(4) times faster than previous in vitro fusion assays. Complete lipid mixing occurs in <15 ms. Both the v-SNARE and the t-SNARE are necessary for efficient docking and fast fusion, but Ca2+ is not. Docking and fusion were quantitatively similar on syntaxin-only bilayers lacking SNAP25. At present, in vitro fusion driven by SNARE complexes alone remains approximately 40 times slower than the fastest, submillisecond presynaptic vesicle population response.  相似文献

The adsorption of tri‐ and hexavalent chromium by the husk of Lathyrus sativus (HLS), which is an agro‐waste has been investigated to find a potential solution to environmental pollution. The pH‐dependent adsorption process finds the optimum values for trivalent and hexavalent chromium ions at about pH 5.0 and pH 2.0, respectively. The process is very fast initially and attains an equilibrium within 90 min following pseudo second‐order rate kinetics. Equilibrium adsorption data can best elucidated by the Langmuir–Freundlich dual model (r2 = 0.998) in comparison with other isotherm models examined indicating that both physi‐ and chemisorption are components of the binding mechanism of chromium ions on HLS. The results show that one gram of HLS can adsorb 24.6 mg Cr3+ and 44.5 mg Cr6+. Fourier transform infrared data and functional group modification experiments indicate that –NH2, ‐COOH, ‐OH, ‐PO43? groups of the biomass interact chemically with the chromium ions. SEM‐energy dispersive X‐ray analysis and X‐ray diffraction spectrum analysis were used to further assess the morphological changes and the mechanisms of chromium ion interaction with HLS. The analysis signified that the biosorption process involved surface morphological changes, complexation and an ion exchange mechanism. The amorphous nature of HLS facilitating metal biosorption was indicated by the X‐ray diffraction analysis.  相似文献

Recent genome-wide association scans (GWAS) and meta-analysis studies on European populations have identified many genes previously implicated in lipid regulation. Validation of these loci on different global populations is important in determining their clinical relevance, particularly for development of novel drug targets for treating and preventing diabetic dyslipidemia and coronary artery disease (CAD). In an attempt to replicate GWAS findings on a non-European sample, we examined the role of six of these loci (CELSR2-PSRC1-SORT1 rs599839; CDKN2A-2B rs1333049; BUD13-ZNF259 rs964184; ZNF259 rs12286037; CETP rs3764261; APOE-C1-C4-C2 rs4420638) in our Asian Indian cohort from the Sikh Diabetes Study (SDS) comprising 3,781 individuals (2,902 from Punjab and 879 from the US). Two of the six SNPs examined showed convincing replication in these populations of Asian Indian origin. Our study confirmed a strong association of CETP rs3764261 with high-density lipoprotein cholesterol (HDL-C) (p?=?2.03×10(-26)). Our results also showed significant associations of two GWAS SNPs (rs964184 and rs12286037) from BUD13-ZNF259 near the APOA5-A4-C3-A1 genes with triglyceride (TG) levels in this Asian Indian cohort (rs964184: p?=?1.74×10(-17); rs12286037: p?=?1.58×10(-2)). We further explored 45 SNPs in a ~195 kb region within the chromosomal region 11q23.3 (encompassing the BUD13-ZNF259, APOA5-A4-C3-A1, and SIK3 genes) in 8,530 Asian Indians from the London Life Sciences Population (LOLIPOP) (UK) and SDS cohorts. Five more SNPs revealed significant associations with TG in both cohorts individually as well as in a joint meta-analysis. However, the strongest signal for TG remained with BUD13-ZNF259 (rs964184: p?=?1.06×10(-39)). Future targeted deep sequencing and functional studies should enhance our understanding of the clinical relevance of these genes in dyslipidemia and hypertriglyceridemia (HTG) and, consequently, diabetes and CAD.  相似文献
