2018年中国植物科学继续呈现快速发展的态势, 我国科学家在国际植物科学主流学术刊物发表论文数量大幅增加, 取得了多项具有重要影响的成果。调控植物生长-代谢平衡实现可持续农业发展入选2018年度中国科学十大进展; 中国被子植物区系进化历史研究入选2018年度中国生命科学十大进展。以水稻为代表的农作物和果蔬等经济作物研究在国际上已呈现出明显的优势, 若干领域已从“追赶”状态跨越到“领跑”地位。该文对2018年中国科学家在植物科学若干领域取得的重要研究成果进行了概括性评述, 旨在全面追踪和报道当前中国植物科学领域的发展前沿和热点, 展示中国科学家所取得的杰出成就。  相似文献

Homologous recombination (HR) of nuclear DNA occurs within the context of a highly complex chromatin structure. Despite extensive studies of HR in diverse organisms, mechanisms regulating HR within the chromatin context remain poorly elucidated. Here we investigate the role and interplay of the histone chaperones NUCLEOSOME ASSEMBLY PROTEIN1 (NAP1) and NAP1‐RELATED PROTEIN (NRP) and the ATP‐dependent chromatin‐remodeling factor INOSITOL AUXOTROPHY80 (INO80) in regulating somatic HR in Arabidopsis thaliana. We show that simultaneous knockout of the four AtNAP1 genes and the two NRP genes in the sextuple mutant m123456‐1 barely affects normal plant growth and development. Interestingly, compared with the respective AtNAP1 (m123‐1 and m1234‐1) or NRP (m56‐1) loss‐of‐function mutants, the sextuple mutant m123456‐1 displays an enhanced plant hypersensitivity to UV or bleomycin treatments. Using HR reporter constructs, we show that AtNAP1 and NRP act in parallel to synergistically promote somatic HR. Distinctively, the AtINO80 loss‐of‐function mutation (atino80‐5) is epistatic to m56‐1 in plant phenotype and telomere length but hypostatic to m56‐1 in HR determinacy. Further analyses show that expression of HR machinery genes and phosphorylation of H2A.X (γ‐H2A.X) are not impaired in the mutants. Collectively, our study indicates that NRP and AtNAP1 synergistically promote HR upstream of AtINO80‐mediated chromatin remodeling after the formation of γ‐H2A.X foci during DNA damage repair.  相似文献

HIRA是组蛋白H3.3的特异分子伴侣, 在组蛋白H3.3掺入染色质的过程中发挥重要作用。研究表明, HIRA在哺乳动物胚胎发育和DNA损伤修复过程中不可或缺。而目前人们对于植物中HIRA同源基因功能的研究相对较少。该研究主要关注拟南芥(Arabidopsis thaliana) AtHIRA基因在植物体细胞同源重组以及减数分裂同源重组过程中的功能。将体细胞同源重组和减数分裂同源重组报告系统分别导入野生型和hira-1突变体后统计同源重组频率, 结果表明在正常生长条件下及在伯莱霉素(bleomycin)或UV-C处理条件下, hira-1突变体体细胞的分子内和分子间同源重组频率均低于野生型。而在正常生长条件下, 野生型与hira-1突变体花粉母细胞间的减数分裂同源重组频率没有明显差异, hira-1突变体的DNA损伤水平与野生型接近。qRT-PCR结果表明, DNA损伤修复相关基因RAD51RAD54hira-1突变体中的表达水平均高于野生型。此外, 盐胁迫处理实验表明, hira-1突变体对于高盐胁迫更加敏感。综上, AtHIRA在拟南芥体细胞同源重组及盐胁迫响应过程中发挥了一定作用。  相似文献

作者对22例溶脲脲原体培养阴性的精液进行细菌及L型培养,其中6例分离出金黄色葡萄球菌及其L型,占27.27%。精液常规分析显示,分离出金葡萄菌及其L型的精液,白细胞计数5例超过正常达10~15个/高倍视野;2例精子的活动率与活动力分别是50%以下和Ⅱ级(低于正常)。金葡菌及L型分离阳性的精子电镜扫描,可见有些精子的头尾部有巨形体、园球体或短杆状菌体吸附,并经免疫酶PAP染色证实为金葡菌L型。实验结果表明:细菌L型感染吸附可能是部分男性不育的原因。  相似文献

Functional structure of the somatomedin B domain of vitronectin   总被引:1,自引:0,他引:1  
The N-terminal somatomedin B domain (SMB) of vitronectin binds PAI-1 and the urokinase receptor with high affinity and regulates tumor cell adhesion and migration. We have shown previously in the crystal structure of the PAI-1/SMB complex that SMB, a peptide of 51 residues, is folded as a compact cysteine knot of four pairs of crossed disulfide bonds. However, the physiological significance of this structure was questioned by other groups, who disputed the disulfide bonding shown in the crystal structure (Cys5-Cys21, Cys9-Cys39, Cys19-Cys32, Cys25-Cys31), notably claiming that the first disulfide is Cys5-Cys9 rather than the Cys5-Cys21 bonding shown in the structure. To test if the claimed Cys5-Cys9 bond does exist in the SMB domain of plasma vitronectin, we purified mouse and rat plasma vitronectin that have a Met (hence cleavable by cyanogen bromide) at residue 14, and also prepared recombinant human SMB variants from insect cells with residues Asn14 or Leu24 mutated to Met. HPLC and mass spectrometry analysis showed that, after cyanogen bromide digestion, all the fragments of the SMB derived from mouse or rat vitronectin or the recombinant SMB mutants are still linked together by disulfides, and the N-terminal peptide (residue 1-14 or 1-24) can only be released when the disulfide bonds are broken. This clearly demonstrates that Cys5 and Cys9 of SMB do not form a disulfide bond in vivo, and together with other structural evidence confirms that the only functional structure of the SMB domain of plasma vitronectin is that seen in its crystallographic complex with PAI-1.  相似文献

Recent evidence have suggested that neuroinflammation and ischemia induce the activation of two different types of reactive astrocytes, termed A1 and A2. Additionally, A1 astrocytes contribute to the death of neurons and oligodendrocytes in neurodegenerative diseases, such as Alzheimer’s disease (AD). In the current study, we constructed an Aβ42-activated microglia-conditioned medium to induce A1 astrocytic activation via secretion of interleukin 1α, tumor necrosis factor, and complement component 1q in vitro, and indicated the regulatory role of milk fat globule epidermal growth factor 8 (MFG-E8) on A1/A2 astrocytic alteration through the downregulation of nuclear factor-κB and the upregulation of PI3K-Akt. This study showed that MFG-E8 suppressed A1 astrocytes and holds great potential for the treatment of AD.  相似文献
