Ichnological analysis of the upper Ypresian–lower Lutetian interval at the Gorrondatxe section (W Pyrenees, N Spain), reveals a relationship between sea-level dynamics and the eco-sedimentary factors influencing trace fossil assemblages. The 600 m thick section of deep-sea turbiditic deposits contains 41 ichnospecies belonging to 28 ichnogenera, which are typical of the Nereites ichnofacies, and mostly of the Paleodictyon ichnosubfacies, suggesting deposition in a basin plain to fan-fringe setting. The trace fossil diversity and abundance fluctuate, irrespective of turbidite frequency. These ichnological features are strongly affected by trophic level changes related partly to sea-level dynamics according to the sequence stratigraphic interpretations for the studied section. Temperature, oxygenation and substrate changes are also considered as relevant factors. Increased ichnodiversity, particularly among graphoglyptids, coincides with moderate oligotrophy and stable ecological conditions. Eutrophisation, lowered oxygenation and drop of temperature, typical of low sea level, can reduce ichnodiversity.  相似文献

Growth and concomitant production of an extracellular pectin lyase (PL) [poly(methoxylgalactosiduronate) endolyase; EC 4.2.2.10] were investigated in a group of 16 fungi grown in liquid medium containing pectin as a supplementary carbon source. Culture filtrates of both Penicillium italicum (CECT 2294) and P. expansum (CECT 2275) showed the highest PL activity and contained polygalacturonase but not pectinesterase activity. The effect of the inoculum size, the carbon source (sucrose and glucose syrup), and the presence of pectin on the production of PL by P. italicum was studied. The presence of 2.6 mM glycerophosphate in the culture medium enhanced the appearance of PL but was not inhibitory for the in vitro activity. However, glycerol inhibited the enzyme nearly 50% at such a concentration.  相似文献

Eutrophication is a major threat to coastal ecosystems. Within Europe, the Water Framework Directive (WFD) has established the need of developing methods of assessment. Bulk chlorophyll “a” is used world-wide as an indicator of eutrophication. However, the size structure of the phytoplankton communities has not been investigated, in detail, in relation to eutrophication pressures. This study investigates the suitability of fractionated chlorophyll “a” (<10 and >10 μm), as an indicator of eutrophication. Along the Basque coast (northern Spain) several water bodies, ranging from offshore waters to the heads of the estuaries, were surveyed during 2008. Physico-chemical conditions and chlorophyll “a” showed a distinct spatial gradient. Trophic richness increased towards the middle and inner parts of the estuaries, where nutrient inputs, from natural or anthropogenic sources, together with the residence time of the water are usually higher. In summer, phytoplankton biomass and abundance decreased, generally, in offshore and coastal waters; in estuaries, they tended to increase. The chlorophyll distribution within the two size fractions was coherent with the phytoplankton taxonomic composition. In summer, the relative abundance of small-sized cells (diatoms and non-siliceous taxa) increased. The relationships between size-fractionated chlorophyll and physico-chemical variables differed, when comparing the offshore and coastal waters, with the estuaries. In the offshore and coastal waters, a strong seasonality was observed; in estuaries, an important spatial component was found. Fractionated chlorophyll provided complementary useful information on anthropogenic pressures. However, more research is necessary to utilise this variable as a tool for ecological status assessment, within the WFD.  相似文献

The translocation of apoptosis-inducing factor (AIF) from mitochondria to the nucleus has been implicated in the mechanism of glutamate excitotoxicity in cortical neurons and has been observed in vivo following acute rodent brain injuries. However, the mechanism and time course of AIF redistribution to the nucleus is highly controversial. Because elevated intracellular calcium is one of the most ubiquitous features of neuronal cell death, this study tested the hypothesis that cleavage of AIF by the calcium-activated protease calpain mediates its release from mitochondria. Both precursor and mature forms of recombinant AIF were cleaved near the amino terminus by calpain I in vitro. Mitochondrial outer membrane permeabilization by truncated Bid induced cytochrome c release from isolated liver or brain mitochondria but only induced AIF release in the presence of active calpain. Enzymatic inhibition of calpain by calpeptin precluded AIF release, demonstrating that proteolytic activity was required for release. Calpeptin and the mitochondrial permeability transition pore antagonist cyclosporin A also inhibited calcium-induced AIF release from mouse liver mitochondria, implicating the involvement of an endogenous mitochondrial calpain in release of AIF during permeability transition. Cleavage of AIF directly decreased its association with pure lipid vesicles of mitochondrial inner membrane composition. Taken together, these results define a novel mechanism of AIF release involving calpain processing and identify a potential molecular checkpoint for cytoprotective interventions.  相似文献

Astydamia latifolia is the only species of the genus Astydamia, which forms an early-diverging lineage of Apiaceae, subfamily Apioideae. This species is subendemic to the Canary Islands and one of the most representative species of the coastal environments of this archipelago. Astydamia displays diplochory, that is, diaspores with two long-distance dispersal (LDD) syndromes. In particular, A. latifolia has both anemochorous and thalassochorous fruit traits (corky and winged mericarps). Although we expect this species to have a high dispersal capacity, there is no genetic study addressing it. The divergence time of this species from its sister taxon is also unknown. In this study, we aimed (i) to unveil the phylogenetic relationships and divergence times of A. latifolia; (ii) to reconstruct its phylogeographic structure across the Canary Islands; and (iii) to estimate the number of inter-island colonization events. To these ends, we first sequenced the internal transcribed spacer (ITS) region for A. latifolia, reconstructed the phylogenetic relationships of Astydamia and Apiaceae relatives and estimated divergence times. Then, two plastid DNA regions (psaI-aacD and psbK-trnS) were sequenced for 158 individuals (from 36 Canarian population and one NW African population) to reconstruct a haplotype network. The recently developed method Phylogeographic Analysis of Island Colonization Events (PAICE) was used to estimate the number of inter-island colonization events. Results show that A. latifolia is a phylogenetically isolated lineage that diverged from the most closely related genus (Molopospermum) in the Eocene–Miocene. It displays a low plastid DNA diversity (only four haplotypes detected), which is accompanied by a high degree of haplotype sharing between islands and highly linear rarefaction curves of colonization obtained in PAICE. These findings suggest a high colonization ability for this species, most likely related to the availability of two LDD syndromes.  相似文献

Novel biotechnologies to valorize waste emissions are based on the use of specialized microbial groups that produce different compounds of industrial interest. On this scenario, the retention of such specific microorganisms in the system is of critical interest; however, the potential limitations of working with simplified cultures in a competitive open environment are neither fully explored nor well understood. In this work, a series of biofilters treating methanol vapors coupled with heterologous endochitinase production were used to evaluate the performance of a specialized microbial population during a typical open-to-environment operation. The biofilters were inoculated with a transformed strain of Pichia pastoris and were operated identically for about 90 days. The results showed that the biofiltration performance became diverse with time in terms of the elimination capacity (EC) shifting from a variation coefficient of 1.5% (EC = 274 ± 24, 279 ± 5, and 281.9 ± 25 g/[m3 h]) at the beginning of the operation to 33% (EC = 297 ± 9, 338 ± 7, and 341 ± 2 g/[m3 h]) at the end of operation. Epifluorescence analysis and cloning-sequencing suggested that P. pastoris remained as the dominant microorganism of methanol degradation, whereas diverse airborne bacteria, including Ochrobactrum spp. and Klebsiella oxytoca, played a secondary role possibly associated with the consumption of intermediates. Overall, this study found that low diversity systems operated under non-sterile conditions could be susceptible to contamination with external microorganisms causing a diversifying behavior at the performance and microbial community levels. © 2018 American Institute of Chemical Engineers Biotechnol.
