Under magnetic resonance (MR) guidance, high intensity focused ultrasound (HIFU) is capable of precise and accurate delivery of thermal dose to tissues. Given the excellent soft tissue imaging capabilities of MRI, but the lack of data on the correlation of MRI findings to histology following HIFU, we sought to examine tumor response to HIFU ablation to determine whether there was a correlation between histological findings and common MR imaging protocols in the assessment of the extent of thermal damage. Female FVB mice (n = 34), bearing bilateral neu deletion tumors, were unilaterally insonated under MR guidance, with the contralateral tumor as a control. Between one and five spots (focal size 0.5 × 0.5 × 2.5 mm3) were insonated per tumor with each spot receiving approximately 74.2 J of acoustic energy over a period of 7 seconds. Animals were then imaged on a 7T MR scanner with several protocols. T1 weighted images (with and without gadolinium contrast) were collected in addition to a series of T2 weighted and diffusion weighted images (for later reconstruction into T2 and apparent diffusion coefficient maps), immediately following ablation and at 6, 24, and 48 hours post treatment. Animals were sacrificed at each time point and both insonated/treated and contralateral tumors removed and stained for NADH-diaphorase, caspase 3, or with hematoxylin and eosin (H&E). We found the area of non-enhancement on contrast enhanced T1 weighted imaging immediately post ablation correlated with the region of tissue receiving a thermal dose CEM43 ≥ 240 min. Moreover, while both tumor T2 and apparent diffusion coefficient values changed from pre-ablation values, contrast enhanced T1 weighted images appeared to be more senstive to changes in tissue viability following HIFU ablation. 相似文献

A survey for West Nile Complement Fixing (CF) antibody was carried out in humans and domestic animals in Nigeria. Human sera were obtained from two communities namely Ibadan and Ogbomoso but animal sera were collected from Ibadan and Maiduguri. The overall CF antibody to West Nile virus in the two localities surveyed was 65%. Of 170 persons tested, 53% and 75% were positive in Ibadan and Ogbomoso respectively. Antibody prevalence increased with age in both communities. Tests for antibody against other flaviviruses revealed that monotypic complement fixation reactions were found frequently in young people, but broadly reacting sera were common among the older age groups. Sex distribution of West Nile CF antibody showed that 49/82 (60%) of females and 62/88 (75%) of males had West Nile CF antibody. Tests on animal sera showed that 33% contained CF antibody to West Nile virus. Prevalence of CF antibody in different animal species was 62% in camels, 4% in cattle and 0% in goats. 相似文献

In the present paper we report on experiments conducted to find out if there is a positive correlation between the destruction of meningeal cells over the newborn rat cerebellum by 6-hydroxydopamine (6-OHDA) and the subsequent development of abnormalities in cerebellar fissuration, lamination, and granule cell number. Both destruction of meningeal cells and quality and magnitude of 6-OHDA-induced cerebellar defects show the same threshold sensitivity without further dose responsiveness. Blockade of neuronal uptake 1 for catecholamines with nomifensine prevents neither destruction of meningeal cells nor the development of abnormalities in cerebellar structure after 6-OHDA treatment. Blockade of extraneuronal uptake 2 for catecholamines with normetanephrine prevents both destruction of meningeal cells and the development of typical cerebellar abnormalities after 6-OHDA treatment. All three parallel experiments suggest that there is a positive correlation between the destruction of meningeal cells and the development of abnormal cerebellar structure, indicating that meningeal cells are involved in these defective morphogenetic processes, i.e., fissuration, lamination, and cell proliferation in the external granular layer. The preferential localization of defects in cerebellar fissures indicates that, in analogy to the mesenchyme surrounding other epithelia with a branching morphogenesis, the role of meningeal cells could be the production of interstitial collagen which is necessary to stabilize the epithelial basal lamina in the fissures. 相似文献

The effects of 6-hydroxydopamine (6-OHDA) on meningeal cells were studied at different ages in conjunction with blockade of the neuronal uptake 1 for catecholamines using nomifensine (NMF) and of the extraneuronal uptake 2 using normetanephrine (NMN). Our results show that maximal numbers of meningeal cells over the cerebellum of the newborn rat are destroyed by a threshold dose of 6-OHDA of 25 micrograms. The morphological characteristics which mark the time course of degeneration of meningeal cells were used to assess the effects of 6-OHDA given in conjunction with either NMF or NMN to differentiate between neuronal (uptake 1) and extraneuronal (uptake 2) effects of 6-OHDA. Uptake of 6-OHDA into meningeal cells and their subsequent degeneration was prevented by pretreatment with NMN but not with NMF. This finding indicates that meningeal cells have uptake 2 capacity but no uptake 1 membrane pump and explains both their uptake of 6-OHDA and their extreme sensitivity to the toxic effects of this drug. Application of this pharmacological regimen using NMF and NMN in conjunction with 6-OHDA thus allows selective destruction of meningeal cells which may be used experimentally to study the contribution of meningeal cells to brain development. 相似文献

Membrane potentials were measured in lateral statocytes of vertically and nonvertically growing roots of Lepidium sativum L. using conventional glass-microelectrode techniques. Statocytes in vertically growing roots showed a stable resting potential of-118±5.9 mV without spontaneous fluctuations. Upon tilting the root 45° from the vertical, an electrical asymmetry was observed. Statocytes on the physically lower side of the root depolarized by approx. 25 mV. This depolarization occurred following a latent period of 8 s reaching a minimum (approx.-93 mV) after 170 s. This depolarization is the earliest event in graviperception ever recorded. After this depolarization, the cell repolarized within 60 s to a potential approx. 10 mV more positive than the original resting potential. Statocytes on the upper flank showed a slow hyperpolarization (t
1/2h=half time for hyperpolarization=168 s) reaching a final, stable potential at a level 10 mV more negative. These effects of gravistimulation were statenchyma-specific, since cells in the cortex and rhizodermis showed no similar effects. The gravi-electrical responses were observed in 25% of all roots tested. Roots which showed no gravi-electrical response had a reduced elongation growth, lacked gravity-induced bending and lacked the typical structural polarity in punctured statocytes. This observed transition from a symmetrical pattern of resting potential in the statenchyma to an asymmetrical pattern following gravistimulation supports the results observed with external current measurements (Behrens et al., Plant Physiol. 70, 1079–1083, 1982) and extends these results to the cellular level and to considerably improved temporal resolution. The asymmetry in the gravi-electrical response extends the graviperception model of Sievers and Volkmann (Planta 102, 160–172, 1972) which comprises an asymmetrical sedimentation of the amyloplasts on the distal endoplasmic reticulum of statocytes. Pair bonds are considered important for successful breeding in monogamous birds but their maintenance may be challenging for migratory species, as mates can be separated for months during the non‐breeding period. To investigate whether mates of monogamous migratory seabirds stay together throughout the non‐breeding period and how and when they start synchronizing their activity before breeding, we tracked seven pairs and 22 individuals of Rhinoceros Auklets Cerorhinca monocerata with geolocators and saltwater immersion loggers. Mates migrated across similar areas during the non‐breeding period but with a sustained temporal shift, putting them an average of 377 km apart and resulting in an average difference of return date at the colony of 5.6 days, with no sex biases. These values did not differ significantly from those between ‘pairs’ of randomly selected, non‐mated birds. Mates showed synchronized on‐water/in‐air at‐sea activities once both birds returned and spent the first night together at the colony. The synchronization of activities was highest on the day following the nights when both mates visited the colony, and decreased with elapsed time. Mates then left the colony together for a pre‐laying exodus of 8–9 days and males returned 2–4 days earlier than females before incubation started. Mates kept synchronizing at‐sea activity during the early part of the exodus. We interpret this as the mates staying together at sea during the pre‐laying period, increasing the males’ chances of copulation at sea. The patterns of mate association observed in Rhinoceros Auklets contrast with those of the Procellariiformes, presumably reflecting differences in the place and timing of copulation. 相似文献

In various studies, auxin (IAA)-induced coleoptile growth has been reported to be closely correlated with an increased occurrence of osmiophilic particles (OPs) at the inner surface of the outer, growth-limiting epidermal cell wall, indicating a possible function related to the mechanism of IAA-induced wall loosening. In order to test whether changes in cell elongation rates of upper and lower flanks (UFs, LFs, respectively) during graviresponsive growth are reflected in appropriate changes in the occurrence of OPs, rye (Secale cereale L.) coleoptiles either as segments or as part of intact seedlings, were gravitropically stimulated by positioning them horizontally for 2 h. Ultrastructural analyses within the UFs and LFs of the upward-bending coleoptiles revealed a distinct imbalance in the occurrence of OPs. The number of OPs per transverse epidermal cell section of the elongation-inhibited UF on average amounted to twice the number of OPs counted in epidermal cell sections of the faster-growing LF. As a hypothesis, the results lead us to suggest that OPs are involved in the mechanism of wall loosening and that temporary growth inhibition of epidermal cells of the UF during upward bending is mediated by inhibition of OP entry into the cell walls. In addition to the statocytes of roots and shoots, a number of tip-growing cells also sense gravity, which influences the cells' growth and development. Since these tip-growing cells are highly suitable for observations in vivo, the movement and sedimentation of their statoliths can be studied in detail. Experimental manipulation by centrifugation, drug application, optical tweezers or microgravity can be monitored by light microscopy. The statoliths are localized in distinct cytoplasmic areas by interactions with actin filaments or microtubules, and their sedimentation seems to be narrowly confined. Since gravisensing and the graviresponse take place within the same cell, the gravitropic signal transduction chain is not complicated by signal transmission between sensing and responding cells. Studies on tip-growing cells have now enabled the formulation of models explaining positive and negative gravitropism. 相似文献
