Here, we report a novel role for hGas7b (human growth arrest specific protein 7b) in the regulation of microtubules. Using a bioinformatic approach, we studied the actin-binding protein hGas7b with a structural similarity to the WW domain of a peptidyl prolyl cis/trans isomerase, Pin1, that facilitates microtubule assembly. Thus, we have demonstrated that hGas7b binds Tau at the WW motif and that the hGas7b/Tau protein complex interacts with the microtubules, promoting tubulin polymerization. Tau, in turn, contributes to protein stability of hGas7b. Furthermore, we observed decreased levels of hGas7b in the brains from patients with Alzheimer disease. These results suggest an important role for hGas7b in microtubular maintenance and possible implication in Alzheimer disease.  相似文献

The human ATP-binding cassette (ABC) transporters ABCB1, ABCC4 and ABCC5 are involved in resistance to chemotherapeutic agents. Here we present molecular models of ABCB1, ABCC4 and ABCC5 by homology based on a wide open inward-facing conformation of Escherichia coli MsbA, which were constructed in order to elucidate differences in the electrostatic and molecular features of their drug recognition conformations. As a quality assurance of the methodology, the ABCB1 model was compared to an ABCB1 X-ray crystal structure, and with published cross-linking and site directed mutagenesis data of ABCB1. Amino acids Ile306 (TMH5), Ile340 (TMH6), Phe343 (TMH6), Phe728 (TMH7), and Val982 (TMH12), form a putative substrate recognition site in the ABCB1 model, which is confirmed by both the ABCB1 X-ray crystal structure and the site-directed mutagenesis studies. The ABCB1, ABCC4 and ABCC5 models display distinct differences in the electrostatic properties of their drug recognition sites.  相似文献

The implementation of genetic groups in BLUP evaluations accounts for different expectations of breeding values in base animals. Notwithstanding, many feasible structures of genetic groups exist and there are no analytical tools described to compare them easily. In this sense, the recent development of a simple and stable procedure to calculate the Bayes factor between nested competing models allowed us to develop a new approach of that method focused on compared models with different structures of random genetic groups. The procedure is based on a reparameterization of the model in terms of intraclass correlation of genetic groups. The Bayes factor can be easily calculated from the output of a Markov chain Monte Carlo sampling by averaging conditional densities at the null intraclass correlation. It compares two nested models, a model with a given structure of genetic groups against a model without genetic groups. The calculation of the Bayes factor between different structures of genetic groups can be quickly and easily obtained from the Bayes factor between the nested models. We applied this approach to a weaning weight data set of the Bruna dels Pirineus beef cattle, comparing several structures of genetic groups, and the final results showed that the preferable structure was an only group for unknown dams and different groups for unknown sires for each year of calving.  相似文献

In this report, a novel D-shaped long-range surface plasmon resonance (LRSPR) fiber base sensor has been introduced. The demonstration of proposed sensor involves two D-shaped silver-coated models to study the sensitivity responses. The entire study with the constructed models is based on a single-mode fiber. The models are multilayered consisting of metal, dielectric, and analyte as separate layers. Silver (Ag) and magnesium fluoride (MgF2) strips are used as metal and dielectric layers respectively. The constituency of analyte as an interface excellently standardized the models for sensitivity detection. In this report, a large range of analyte refractive indices (RI) which varies from 1.33 to 1.38 is appraised for the proposed models to characterize the sensitivity. The entire context is encompassed by the wavelength region from 450 to 850 nm with an interval of 20 nm. Sensitivities in this report are measured based on the analyte position from the core and metal for both models. For each of the two models, the analyte is placed as the top layer. RIs of the applied metal (Ag) are measured using the Drude-Lorentz formula. The simulated sensitivities for model-1 and model-2 vary from 6.3?×?103 nm/RIU to 8.7?×?103 nm/RIU.

Vancomycin-resistant enterococcci (VRE) in Europe are thought to have emerged partly due to the use of the glycopeptide avoparcin in animal husbandry. We compared the occurrence of VRE in geographical regions of Europe in which until 1997 large amounts of avoparcin were used (Spain, United Kingdom, and Denmark) with the occurrence of VRE in Sweden, where avoparcin was banned in 1986. We also studied the relatedness between VRE strains from different regions and habitats. In total, 2,580 samples were collected from humans, animals, and the environment (soil, sewage, recipient water). VRE resistant to 20 μg/ml vancomycin were identified in 8.2% of the samples and were found most frequently in raw and treated urban sewage samples (means, 71% and 36% of the samples, respectively), pig manure (17%), and hospital sewage (16%). The proportions of VRE-positive sewage samples were similar in Sweden, Spain, and the United Kingdom, whereas pig feces and manure were more often positive in Spain than in Sweden (30% versus 1%). Most VRE were Enterococcus faecium carrying vanA, and computerized biochemical phenotyping of the isolates of different ecological origins showed a high degree of polyclonality. In conclusion, it seems that animal-associated VRE probably reflect the former use of avoparcin in animal production, whereas VRE in human-associated samples may be a result of antibiotic use in hospitals. Since there seems to be a reservoir of the resistance genes in all countries studied, precautions must be taken to limit the use of antibiotics and antibiotic-like feed additives.  相似文献

An Arthrobacter sp. strain, F101, able to use fluorene as the sole source of carbon and energy, was isolated from sludge from an oil refinery wastewater treatment plant. During growth in the presence of fluorene, four major metabolites were detected and isolated by thin-layer chromatography and high-performance liquid chromatography. 9-Fluorenol, 9H-fluoren-9-one, and 3,4-dihydrocoumarin were identified by UV spectra, mass spectrometry, and 300-MHz proton nuclear magnetic resonance. The fourth metabolite has been characterized, but precise identification was not possible. Since strain F101 is not able to grow with fluorenone, two different pathways of fluorene biodegradation are suggested: one supports cell growth and produces 3,4-dihydrocoumarin as an intermediate and probably the unidentified metabolite, and the other produces 9-fluorenol and 9H-fluoren-9-one and appears to be a dead-end route.  相似文献

A survey for West Nile Complement Fixing (CF) antibody was carried out in humans and domestic animals in Nigeria. Human sera were obtained from two communities namely Ibadan and Ogbomoso but animal sera were collected from Ibadan and Maiduguri. The overall CF antibody to West Nile virus in the two localities surveyed was 65%. Of 170 persons tested, 53% and 75% were positive in Ibadan and Ogbomoso respectively. Antibody prevalence increased with age in both communities. Tests for antibody against other flaviviruses revealed that monotypic complement fixation reactions were found frequently in young people, but broadly reacting sera were common among the older age groups. Sex distribution of West Nile CF antibody showed that 49/82 (60%) of females and 62/88 (75%) of males had West Nile CF antibody. Tests on animal sera showed that 33% contained CF antibody to West Nile virus. Prevalence of CF antibody in different animal species was 62% in camels, 4% in cattle and 0% in goats.  相似文献

Nitrate (NO3) and nitrite (NO2) are the physiological sources of nitric oxide (NO), a key biological messenger molecule. NO3/NO2 exerts a beneficial impact on NO homeostasis and its related cardiovascular functions. To visualize the physiological dynamics of NO3/NO2 for assessing the precise roles of these anions, we developed a genetically encoded intermolecular fluorescence resonance energy transfer (FRET)-based indicator, named sNOOOpy (sensor for NO3/NO2 in physiology), by employing NO3/NO2-induced dissociation of NasST involved in the denitrification system of rhizobia. The in vitro use of sNOOOpy shows high specificity for NO3 and NO2, and its FRET signal is changed in response to NO3/NO2 in the micromolar range. Furthermore, both an increase and decrease in cellular NO3 concentration can be detected. sNOOOpy is very simple and potentially applicable to a wide variety of living cells and is expected to provide insights into NO3/NO2 dynamics in various organisms, including plants and animals.  相似文献
