湛江高桥红树林和盐沼湿地的大型底栖动物次级生产力   总被引:2,自引:0,他引:2  
为了比较湛江高桥潮间带不同植物生境的大型底栖动物次级生产力,根据2010年4个季度湛江高桥潮间带生境的大型底栖动物数据,运用Brey经验公式计算不同植物生境的大型底栖动物次级生产力.结果表明:湛江高桥红树林和盐沼湿地不同生境大型底栖动物平均次级生产力为11.77 g AFDM·m-2·a-1.其中,无瓣海桑生境次级生产力最高,为18.16 g AFDM·m-2·a-1,其次是桐花树、盐地鼠尾粟和木榄生境,分别为17.67、8.34和2.92 g AFDM·m-2·a-1.在4种生境中,木榄生境的年生产力/年均生物量(P/B)最高,为2.38,其次是无瓣海桑、盐地鼠尾粟和桐花树生境,分别为1.23、0.99和0.48.湛江高桥潮间带不同植物生境大型底栖动物次级生产力和P/B值的差异主要与总有机碳含量、食物类型和动物个体大小有关.  相似文献

利用扫描电镜(SEM)和光镜(LM)对臭椿花序及花器官的分化和发育进行了初步研究,表明:1)臭椿花器官分化于当年的4月初,为圆锥花序;2)分化顺序为花萼原基、花冠原基、雄蕊原基和雌蕊原基。5个萼片原基的发生不同步,并且呈螺旋状发生;5个花瓣原基几乎同步发生且其生长要比雄蕊原基缓慢;雄蕊10枚,两轮排列,每轮5个原基的分化基本是同步的;雌蕊5,其分化速度较快;3)在两性花植株中,5个心皮顶端粘合形成柱头和花柱,而在雄株中,5个心皮退化,只有雄蕊原基分化出花药和花丝。本研究着重观察了臭椿中雄花及两性花发育的过程中两性花向单性花的转变。结果表明,臭椿两性花及单性花的形成在花器官的各原基上是一致的(尽管时间上有差异),雌雄蕊原基同时出现在每一个花器官分化过程中,但是,可育性结构部分的形成取决于其原基是否分化成所应有的结构:雄蕊原基分化形成花药与花丝,雌蕊原基分化形成花柱、柱头和子房。臭椿单性花的形成是由于两性花中雌蕊原基的退化所造成,其机理有待于进一步研究。  相似文献

Bmi-1, the first functionally identified polycomb gene family member, plays critical roles in cell cycle regulation, cell immortalization, and cell senescence. Bmi-1 is involved in the development and progression of carcinomas and is a potent target for cancer therapy. One important pathway regulated by Bmi-1 is that involving two cyclin-dependent kinase inhibitors, p16Ink4a and p19Arf, as Bmi-1 represses the INK4a locus on which they are encoded. A close correlation between the up-regulation of Bmi-1 and down-regulation of p16 has been demonstrated in various tumors; however, how Bmi-1 regulates p16 expression is not clear. In this study, we revealed that Bmi-1 regulates the expression of p16 by binding directly to the Bmi-1-responding element (BRE) within the p16 promoter. The BRE resided at bp −821 to −732 upstream of the p16 ATG codon. BRE alone was sufficient to allow Bmi-1-mediated regulation of the CMV promoter. Bmi-1 typically functions by forming a complex with Ring2; however, regulation of p16 was independent of Ring2. Chromatin immunoprecipitation sequencing of Bmi-1-precipitated chromatin DNA revealed that 1536 genes were targeted by Bmi-1, including genes involved in tissue-specific differentiation, cell cycle, and apoptosis. By analyzing the binding sequences of these genes, we found two highly conserved Bmi-1-binding motifs, which were required for Bmi-1-mediated p16 promoter regulation. Taken together, our results revealed the molecular mechanism of Bmi-1-mediated regulation of the p16 gene, thus providing further insights into the functions of Bmi-1 as well as a sensitive high-throughput platform with which to screen Bmi-1-targeted small molecules for cancer therapy.  相似文献

对渔用抗菌剂复方五倍子有效成分进行薄层色谱与柱层析分离检测,以鉴定出复方中的各种有效成分;选用几种水产动物常见致病菌进行体外抑菌和联合用药试验,以确定各有效成分抑菌活性强弱和各成分间的药物关系,为分析复方五倍子抗菌剂的药物作用机理奠定理论基础,并为其质量控制提供参考依据。用薄层析硅胶GF254制板,以苯:乙醇:甲酸(7:3:1)为展开剂,用氨熏-三氯化铁显色,日光下观察层析结果;通过干法装硅柱,以苯:乙醇:甲酸(7:3:1)作为洗脱液进行洗脱,分离得到各单体,参照理化性质分析及薄层分析,鉴定出复方五倍子抗菌剂总有效成分。结果表明;复方五倍子抗菌剂有效成分为鞣质、没食子酸、槲皮素、黄芩苷、芦荟大黄素、大黄素和1,8-二羟基蒽醌,其中没食子酸含量最大,紫外分光光度法测定为10.47%。用复方的各有效成分别对温和气单胞菌、豚鼠气单胞菌、迟缓爱德华菌和柱状黄杆菌进行抑菌试验,以温和气单胞菌为菌种将复方主要有效成分两两配对进行体外联合抑菌试验,得出复方五倍子各有效成分抑菌活性顺序为:没食子酸>蒽醌(含1,8-二羟基蒽醌、大黄素、芦荟大黄素)>黄芩苷>槲皮素>鞣质。没食子酸和蒽醌具有协同作用,没食子酸和黄芩苷、没食子酸和槲皮素、蒽醌和黄芩苷三组同属于累加作用。    相似文献

This study aimed to increase the glycolytic flux of the multivitamin auxotrophic yeast Torulopsis glabrata by redirecting NADH oxidation from oxidative phosphorylation to membrane-bound ferric reductase. We added potassium ferricyanide as electron acceptor to T. glabrata culture broth at 20% dissolved oxygen (DO) concentration, which resulted in: (1) decreases in the NADH content, NADH/NAD(+) ratio, and ATP level of 45.3%, 60.3%, and 15.2%, respectively; (2) high activities of the key glycolytic enzymes hexokinase, phosphofructokinase, and pyruvate kinase, as well as high expression levels of the genes encoding these enzymes; and (3) increases in the specific glucose consumption rate and pyruvate yield of T. glabrata was by 45.5% and 23.1%, respectively. Our results showed that membrane-bound ferric reductase offers an alternative and efficient NADH oxidation pathway at lower DO concentration, which increases the glycolytic flux of T. glabrata.  相似文献

A continuous cell line, the flounder embryonic cell line (FEC), was established from gastrula-stage embryos of a marine cultured fish, the Japanese flounder Paralichthys olivaceus and cultured for more than 200 d with more than 60 passages. FEC cells were cultured in Dulbecco's Modified Eagle's Medium (DMEM) supplemented with antibiotics, fetal bovine serum (FBS), sea perch serum (SPS), and basic fibroblast growth factor (bFGF). The cells were small and round, and grew actively and stably in culture. The effect of temperature, FBS concentration and bFGF on FEC cell growth was examined. Cells grew well between 24 and 30 degrees C, but had a reduced growth rate below 18 degrees C. The growth rate of FEC cells in medium containing 15% FBS was higher than that in medium containing 7.5% FBS. Addition of bFGF to the medium also significantly increased the growth rate. Chromosome analysis revealed that FEC cells have a normal diploid karyotype with 2n = 48. High survival rate was obtained after cryopreservation of cell cultures. The susceptibility of the cell line to piscine viruses was examined. Two viruses tested were shown to induce CPE (cytopathic effect) on FEC cells. FEC cell culture infected with fish iridovirus was further elucidated by electron microscopy. Many virus particles were found in the cytoplasm of the virus-infected FEC cells. These results indicated that the FEC cell line could be potentially used to isolate and study fish viruses.  相似文献
