The aminoglycoside phosphotransferase(3′)-IIIa (APH) is a promiscuous enzyme and renders a large number of structurally diverse aminoglycoside antibiotics useless against infectious bacteria. A remarkable property of this ~31 kDa enzyme is in its unusual dynamic behavior in solution; the apo-form of the enzyme exchanges all of its backbone amide protons within 15 h of exposure to D 2 O while aminoglycoside-bound forms retain ~40% of the amide protons even after >90 h of exposure. Moreover, the number of observable peaks and their dispersion in HSQC spectra varies with each aminoglycoside, rendering the resonance assignments very challenging. Therefore, the binary APH–tobramycin complex, which shows the largest number of well-resolved peaks, was used for the backbone resonance assignments (Cα, C, N, H, and some Cβ) of this protein (BMRB-16337).  相似文献

Pachychilid gastropods are a conspicuous element of the freshwater macro‐invertebrate fauna of South‐East Asia. In this region, three spatially separated groups of pachychilids can be differentiated mostly by means of their brooding strategy. The largest group comprises about 100 described species and displays an extended Sundaland distribution that ranges from north‐east India in the west to southern China in the east and Borneo in the south‐east. The most conspicuous synapomorphy shared by females is a subhaemocoelic brood pouch in which developing embryos are retained. Mitochondrial phylogenies based on partial sequences of the genes cytochrome oxidase C subunit I (COI) and 16S rRNA (16S) support the monophyly of this group of subhaemocoelic brooders. Amongst them, the two genera Brotia and Paracrostoma form a sister pair of clades both together forming the sister taxon of a third clade. This third clade itself is split into three subclades, which occupy separate ranges except for the overlap of two of them in Borneo. The subclades comprise species from (A) Thailand, Cambodia, and northern Borneo; (B) Java and south‐western Borneo; and (C) Vietnam and southern China. In contrast with their spatial structuring, all species share a widely corresponding anatomy. Characteristic key features are a pallial oviduct with a seminal receptacle and an embryonic shell with a smooth, dome‐like inflated apex. Differences in the histology of the pallial oviduct are not congruent with phylogenetic patterns as suggested by the mitochondrial trees. Given the lack of obvious distinguishing morphological characteristics, all species of the subclades A to C are assigned to a single genus, Sulcospira, with Adamietta as a junior synonym.  相似文献

Chlamydia trachomatis ribonucleotide reductase (RNR) is a class Ic RNR. It has two homodimeric subunits: proteins R1 and R2. Class Ic protein R2 in its most active form has a manganese–iron metal cofactor, which functions in catalysis like the tyrosyl radical in classical class Ia and Ib RNRs. Oligopeptides with the same sequence as the C‐terminus of C. trachomatis protein R2 inhibit the catalytic activity of C. trachomatis RNR, showing that the class Ic enzyme shares a similar highly specific inhibition mechanism with the previously studied radical‐containing class Ia and Ib RNRs. The results indicate that the catalytic mechanism of this class of RNRs with a manganese–iron cofactor is similar to that of the tyrosyl‐radical‐containing RNRs, involving reversible long‐range radical transfer between proteins R1 and R2. The competitive binding of the inhibitory R2‐derived oligopeptide blocks the transfer pathway. We have constructed three‐dimensional structure models of C. trachomatis protein R1, based on homologous R1 crystal structures, and used them to discuss possible binding modes of the peptide to protein R1. Typical half maximal inhibitory concentration values for C. trachomatis RNR are about 200 µ m for a 20‐mer peptide, indicating a less efficient inhibition compared with those for an equally long peptide in the Escherichia coli class Ia RNR. A possible explanation is that the C. trachomatis R1/R2 complex has other important interactions, in addition to the binding mediated by the R1 interaction with the C‐terminus of protein R2. Introduction: Microwave and water bath postpolymerization have been suggested as methods to improve the mechanical properties of heat and autopolymerizing acrylic resins. However, the effects of autoclave heating on the fracture properties of autopolymerizing acrylic resins have not been investigated. Purpose: The aim of this study was to assess the effectiveness of various autoclave postpolymerization methods on the fracture properties of 3 different autopolymerizing acrylic resins. Methods: Forty-two specimens of 3 different autopolymerizing acrylic resins (Orthocryl, Paladent RR and Futurajet) were fabricated (40x8x4mm), and each group was further divided into 6 subgroups (n=7). Control group specimens remained as processed (Group 1). The first test group was postpolymerized in a cassette autoclave at 135°C for 6 minutes and the other groups were postpolymerized in a conventional autoclave at 130°C using different time settings (5, 10, 20 or 30 minutes). Fracture toughness was then measured with a three-point bending test. Data were analyzed by ANOVA followed by the Duncan test (α=0.05). Results: The fracture toughness of Orthocryl and Paladent-RR acrylic resins significantly increased following conventional autoclave postpolymerization at 130°C for 10 minutes (P<.05). However, the fracture toughness of autoclave postpolymerized Futurajet was not significantly different than its control specimens (P<.05). The fracture toughness of Futurajet was significantly less than Paladent RR and Orthocryl specimens when autoclaved at 130°C for 10 minutes. Conclusions: Within the limitations of this study, it can be suggested that autoclave postpolymerization is an effective method for increasing the fracture toughness of tested autoploymerized acrylic resins.  相似文献

IN most work on the problems of releasing radiostrontium which has been incorporated into the vertebrate skeleton the effects of various parenterally administered chemical agents on the excretion rate of radiostrontium have been studied. Someriecrease in skeletal retention was obtained when inactive salts of strontium1–4 or zirconium5 were given almost simultaneously with radiostrontium; so far all other efforts have been practically useless.  相似文献

Environmental fluctuations can select for generalism, which is also hypothesized to increase organisms’ ability to invade novel environments. Here, we show that across a range of temperatures, opportunistic bacterial pathogen Serratia marcescens that evolved in fluctuating temperature (daily variation between 24°C and 38°C, mean 31°C) outperforms the strains that evolved in constant temperature (31°C). The growth advantage was also evident in novel environments in the presence of parasitic viruses and predatory protozoans, but less clear in the presence of stressful chemicals. Adaptation to fluctuating temperature also led to reduced virulence in Drosophila melanogaster host, which suggests that generalism can still be costly in terms of reduced fitness in other ecological contexts. While supporting the hypothesis that evolution of generalism is coupled with tolerance to several novel environments, our results also suggest that thermal fluctuations driven by the climate change could affect both species’ invasiveness and virulence.  相似文献

Abstract Volcanic archipelagos represent excellent areas to study colonization and speciation processes. The grasshopper genus Arminda is one of many endemic taxa of the Canary Islands. It consists of seven wingless species, most of which are single‐island endemics. We sequenced two mitochondrial (12s rRNA, ND5) and two nuclear gene fragments (28s rRNA, ITS2) to reconstruct the colonization pattern of the genus. Our results are in accordance with a stepping‐stone colonization model from east to west, corresponding to the prevailing ocean currents, but alternative hypotheses cannot be fully rejected. The populations of A. brunneri from Tenerife belong to two different lineages (east and west) consistent with the geological history of the island. It remains to be tested whether these lineages represent different species and whether further lineages exist on this island. The five clades of the four western islands (A. brunneri group) have approximately similar branch lengths. The short internodes between these lineages resulted in a poorer phylogenetic resolution. Specimens from La Palma were genetically distinct and are subsequently described as a new species, Arminda palmae sp.n. Our results suggest in situ speciation on Gran Canaria, which was accompanied by a stronger degree of morphological diversification than the inter‐island speciation processes. The aberrant species A. canariensis has formerly been assigned to a monotypic subgenus Chopardminda, which is now synonymized with Arminda syn.n. based on its phylogenetic position. Gran Canaria seems to be the only island where Arminda species occur sympatrically, although allopatric speciation seems likely due to the long history of volcanism and erosion on the island.  相似文献

Four-week old plants of chamomile (Matricaria chamomilla) cultivated in nutrient solution were exposed to copper (3, 60 and 120 μM) for 10 days. At 120 μM, Cu decreased dry mass production, water, chlorophyll and nitrogen content in both the leaf rosettes and roots. Five phenolic acids were detected in methanol extracts of the leaf rosettes (protocatechuic, p-hydroxybenzoic, vanillic, chlorogenic and salicylic acid) and six additional compounds (gentisic, syringic, caffeic, sinapic and o-/p-coumaric acid) were released after acid hydrolysis. Most of the 11 phenolic acids detected increased in 60 μM Cu but in the 120 μM treatment their contents were lower or not significantly different from the control. Among coumarin-related compounds, (Z)- and (E)-2-ß-d-glucopyranosyloxy-4-methoxycinnamic acids increased in 60 and 120 μM Cu while herniarin rose in the 3 and 60 μM Cu by the end of the experiment. The amounts of umbelliferone were not affected by any of the doses tested. These facts in relation to antioxidative properties of phenolic metabolites are also discussed. The malondialdehyde content of the leaf rosettes was not affected by exposure of plants to 120 μM Cu in a time-course experiment but in the roots a sharp increase was observed after 24 and 48 h of treatment. At 120 μM, Cu stimulated a 9-fold higher K+ loss than the 60 μM treatment while at the lowest concentration it stimulated potassium uptake. Cu accumulation in the roots was 3-, 49- and 71-fold higher than that in the leaf rosettes in the 3, 60, and 120 μM Cu treatments, respectively. Results suggest that 120 μM Cu dose is limiting for chamomile growth under the conditions of present research.  相似文献

In this work, we present an addendum to the "Checklist of the helminth parasites of vertebrates in Costa Rica" with a parasite-host list as well as a host-parasite list. This addendum updates the available information on this group of parasites in Costa Rica, since very recently a new input has been made to describe the helminth fauna of vertebrates, particularly at the Area de Conservación de Guanacaste. In this paper, we add 33 records, representing 23 species. This raises the number of helminth species described in vertebrates from Costa Rica to 325, represented by 89 species of digeneans, 23 of monogeneans. 63 of cestodes, 13 of acanthocephalans, and 137 of nematodes. In total, 133 species of vertebrates have been studied for helminths in Costa Rica (31 species of fishes. 7 amphibians, 18 reptiles, 40 birds, and 37 mammals). Currently, 67 species (20.6 %) have been recorded as new species from Costa Rica and most of them are endemic to particular regions. The Colecci6n de Helmintos de Costa Rica, housed at the Universidad de Costa Rica (UCR), San José, is the depositary of only 23% of the species recorded so far; however this situation is changing and people recognizes.  相似文献
