Oxidative phosphorylation in a yeast mutant lacking α-ketoglutarate dehydrogenase activity

The effect of α-ketoglutarate deficiency on the oxidative phosphorylation in yeast mitochondria was studied. By determining the properties of electron transport and energy transduction systems of mutant mitochondria it was found that the lack of α-ketoglutarate dehydrogenase activity in mitochondria does not result in any functional defect in the oxidative phosphorylation system.     

Distribution of sulphate and iduronic acid residues in heparin and heparan sulphate

1. A method was developed for determination of the uronic acid composition of heparin-like glycosaminoglycans. Polymers or oligosaccharides are degraded to monosaccharides by a combination of acid hydrolysis and deamination with HNO₂. The resulting uronic acid monosaccharides (accounting for about 70% of the uronic acid contents of the starting materials) are isolated and converted into the corresponding aldono-1,4-lactones, which are separated by g.l.c. The calculated ratios of glucuronic acid/iduronic acid are reproducible within 5%. 2. Samples of heparin from pig intestinal mucosa (molar ratio of sulphate/disaccharide unit, 2.40) and heparan sulphate from human aorta (sulphate/disaccharide ratio, 0.46) were subjected to uronic acid analysis. l-Iduronic acid constituted 77% and 19% respectively of the total uronic acid contents. 3. The correlation between the contents of sulphate and iduronic acid indicated by this finding also applied to the fractionated deamination products of the two polymers. The sulphated fragments varied in size from disaccharide to octasaccharide (or larger) and showed sulphate/disaccharide molar ratios in the range of 0.05–2.0. The proportion of iduronic acid increased with increasing ester sulphate contents of the oligosaccharides. 4. Previous studies on the biosynthesis of heparin in a cell-free system have shown that l-iduronic acid residues are formed by C-5 epimerization of d-glucuronic acid units at the polymer level; the process requires concomitant sulphation of the polymer. The results obtained in the present structural study conform to these findings, and suggest further that similar mechanisms may operate in the biosynthesis of heparan sulphate. The epimerization reaction appears to be linked to the sulphation of hydroxyl groups but does not seem to require sulphation of the target uronic acid residues. The significance of sulphamino groups in relation to the formation of iduronic acid is unknown.     

Intracytoplasmatic inclusions in cells of lettuce leaves with mosaic-like symptoms

Ultrathin sections were prepared from the tissues of lettuce leaves with mosaic-like symptoms and thickened nervature which were studied by means of electron microscopy. Intracellular inclusions surrounded by a membrane were found in the cytoplasm of parenchym cells of the investigated lettuce leaves(Lactuca saliva L. provar.capitata L. nid.jaggeri Helm., cv. Pra?an). Crystals with a distinctly apparent hexagonal lattice could be observed in the inclusions. No crystal containing inclusions were found in the tissues from the leaves without mosaic-like symptoms and in those from thickened nervature.     

The production of ethylene by plants determined by means of paper chromatography

Ethylene was collected in methanol solution of mercuric acetate and the addition compound formed was then separated by means of paper chromatography. The spot area and colour intensity after detection were determined using a densitometer. The amount of collected ethylene was calculated from a calibration curve. The ethylene liberated from plant samples was collected during one or two days. During this period the amputation of the whole plant organs did not influence ethylene production. Changes in ethylene production were found after segmentation of the tissue or after the treatment with auxin and Co²⁺ ions. The above-ground parts of investigated herbs released 0.3 to 3.5 μl of ethylene per kg fresh weight per hour. The leaves of investigated trees released 1 to 20 μl of ethylene per kg f.w. per hour. The rate of the production of ethylene seems to be specific for a given species.     

Comparison of the mutagenic activity of N-ethyl- N-nitrosourea and N-methyl- N’- nitro- N- nitrosoguanidine inLens esculenta

Mutagenic activity of N-ethyl-N-nitrosourea (ENU) and of N-methyl-N’-nitro-N--nitrosoguanidine (MNG) in lentil was studied. The highest proportion of segregating progenies with chlorophyll mutants and chimeric plants was 34.8% from the total number of analysed offsprings, ENU being applied in this case in the concentration of 0.005% for 20 h at 18 to 19 °C. When MNG was applied in the concentration of 0.001 % for 10h at 22 to 23 °C the proportion was 5.1%. Progenies segregating two or more chlorophyll mutants originated with ENU only; their relative frequencies varied from 1.4% to 7.1%. The number of different types of mutants or of their combinations segregating at the same time in the same progeny was shown to be dependent with the two agent tested on the mutagenic activity of the concentration used. The most efficient concentration of ENU induced the total of 8 different mutants at the same time, together with a combination of two or three mutant types in the same progeny. With MNG no combination of chlorophyll mutants in the same progeny was ever found simultaneously. The greatest number of mutants corresponding to 1 progeny M₁ was 0.53 when ENU was applied; with MNG the maximum values were approximately ten times lower. The maximum number M₂ of chlorophyll mutants and chimeric plants was 3.58% with ENU and 0.23 with MNG.     

On the interaction of growth retardants with IAA and kinetin

In the pea test a highly positive response to the treatment with IAA reversed to a negative one or became 5 to 6 times weaker when CCC was applied together with IAA. In cultivating pea seedlings, following their decapitation, for two days in a 0.25 per cent CCC solution and then in water, growth of their cotyledonous axillaries (cotylaries) were inhibited. This inhibitive action of CCC could be made ineffective when the seedlings, following two-days’ cultivation in the CCC solution, were grown further in kinetin solutions (0.37–3 mg per 1). Cotylaries of decapitated pea seedlings, when grown in kinetin solutions were inhibited. With kinetin solutions of 6–12 mg/l a strong inhibition also occured in the growth of roots at the apical parts of which spherical swellings were developing. The CCC supplied to the roots of intact etiolated pea seedlings is translocated acropetally into the stem at a rate of about 5 cm per hour. Decapitation of the plant causes retardation of this transport, yet a coat of 0.00001–1% IAA or kinetin paste produces acceleration of the stream. Existence of an antagonism between CCC and IAA, demonstrated earlier, was found holding true also for B-9 (N, N-dimethyl-aminesuccinamic acid) and IAA, as the inhibitive action of B-9, 0.06% solution on the growth of lettuce hypocotyls was reduced to a highly significant degree when the plants were supplied with B-9 together with IAA at a concentration of 10 mg/l.     

Conductivity and photoconductivity of NaDNA in the solid state

The d. c. conductivity of five samples of NaDNA in dry state was investigated. All specimens show similar behavior, with specific resistivity at 40°C of the order of 10¹³–10¹⁴ ohm-cm and an energy gap of E_c = 2.04–2.25 eV. Based on the Eley's model for conductivity mechanism, the possibility of conservation of an ordered structure in the dry state (temperature range 20–80°C) is discussed. This is ascribed to the interaction of electron systems of individual bases. From the activation energy values it may be estimated that 17–19 π-electrons take part in the interaction. Photoconductivity measurements indicated activation energies E_f of 0.88–0.96 eV in the visible region.
