Stroke is a disease that affects the blood vessels that supply blood to the brain. Although platelets are implicated in the pathophysiology of stroke the mechanism is still not clear and there antiplatelet agents available for the prevention and treatment of stroke. We herein examined the relationship between the potential cytokine, TNF-α platelet activation and apoptosis in acute ischemic stroke patients. We selected 60 patients (mean age 57.9 ± 10.2 years) who had not taken any antiplatelet drugs for 14 days. A group of 45 participants (mean age 51.05 ± 9.07 years) were selected as the control group. For both the patients and for the control group, P-selectin (CD62p) and Annexin-V binding, cytochrome-c levels, caspase-3 gene expression and caspase-3 releasing and plasma TNF-α levels were measured in platelets. The results showed significant increase in plasma TNF-α and platelet Annexin-V, CD62p, cytochrome-c and caspase-3 gene expression in stroke patients compared to the control group. The data of this work suggests that inflammation may have a role in platelet apoptosis in stroke which may suggest a new aspect of the role of inflammation in the development of acute ischemic stroke.  相似文献

Congenital adrenal hyperplasia (CAH) is a group of autosomal recessively inherited disorders characterized by impaired production of adrenal steroids. Approximately 95% of all CAH are caused by mutations of the CYP21A2 that encodes 21-hydroxylase. In this study, mutation analyses of CYP21A2 were performed in 48 CAH patients from 45 Turkish families with the clinical diagnosis of 21-hydroxylase deficiency (21OHD). While in 39 (86.7%) of 21OHD patients, disease causing CYP21A2 mutations were identified in both alleles, in two 21OHD patients CYP21A2 mutations were identified only in one allele. In four patients, mutation was not detected at all. In total, seventeen known and one novel, disease causing CYP21A2 mutations were observed. Among identified mutations, previously described c.293-13C/A>G, large rearrangements and p.Q319X mutations were the most common mutations accounting for 33.3%, 14.4% and 12.2% of all evaluated chromosomes, respectively. In six families (13.3%) a novel founder mutation, c.2T>C (p.M1?), inactivating the translation initiation codon was found. This mutation is not present in pseudogene CYP21A1P and causes the classical form of the disease in six patients. In addition, depending on the nature of the rearrangements CYP21A1P/CYP21A2 chimeras were further classified as CHc/d, and CH-1c was shown to be the most prominent chimera in our study group. In conclusion, with this study we identified a novel founder CYP21A2 mutation and suggest a further classification for CYP21A1P/CYP21A2 chimeras depending on the combination of junction site position and whether it is occurred as a result of deletion or conversion. Absence of disease causing mutation of CYP21A2 in ten of screened ninety chromosomes suggests the contribution of regulatory elements in occurrences of CAH due to the 21OHD.  相似文献

An aeropalynological study during the years 2014–2015 was performed in Hatay, which is a unique sociocultural and phytogeographical area located on the border of Turkey and Syria on the eastern coast of the Mediterranean. The sampling was performed by a Hirst-type volumetric sampler (Lanzoni VPPS 2000), and pollen grains of 54 taxa were identified, of which 83.21% of the annual sum belonged to woody taxa. The highest pollen concentration was recorded in February, of which a large amount came from the Cupressaceae/Taxaceae families. The diversity of the pollen reflected the vegetation of the area and plantations of the city center, but pollen grains from Euro-Siberian elements specific to Mount Amanos could not be recorded. Pollen types found at more than 3% of the annual pollen index and considered dominant pollen types were as follows: Cupressaceae/Taxaceae (50.86%), Olea europaea (12.67%), Moraceae (7.20%), Poaceae (5.99%), Quercus (5.35%), Urticaceae (3.79%) and Pinus (3.70%); almost all dominant pollen types in the city atmosphere were previously stated to be allergic. The main pollen season starting dates of common pollen types found were one or two weeks earlier than those of the surroundings. Many statistically significant correlations were found between daily pollen concentrations and daily meteorological parameters, e.g., Cupressaceae/Taxaceae Poaceae and Urticaceae pollen correlated negatively with mean temperature in both years, and in the hindermost two families daily pollen amounts significantly correlated with wind speed in the second year. Daily Olea europaea pollen concentration showed a significant negative correlation with the amount of total daily rainfall in the second year.  相似文献

Accumulation of radiolabelled naphthalene-1-acetic acid (1-NAA), 2,4-dichlorophenoxyacetic acid (2,4-D), and indole-3-acetic acid (IAA) has been measured in suspension-cultured tobacco (Nicotiana tabacum) cells. In this paper is presented a simple methodology allowing activities of the auxin influx and efflux carriers to be monitored independently by measuring the cellular accumulation of [3H]NAA and [14C]2,4-D. We have shown that 1-NAA enters cells by passive diffusion and has its accumulation level controlled by the efflux carrier. By contrast, 2,4-D uptake is mostly ensured by the influx carrier and this auxin is not secreted by the efflux carrier. Both auxin carriers contribute to IAA accumulation. The kinetic parameters and specificity of each carrier have been determined and new information concerning interactions with naphthylphthalamic acid, pyrenoylbenzoic acid, and naphthalene-2-acetic acid are provided. Many of the plants found in serpentine areas are endemics and they may accumulate Ni at high concentration. High accumulation of Cr is rare, or in some views, never properly demonstrated. Generally, a very small proportion of any serpentine flora shows high accumulation of Ni, in some serpentine areas Ni accumulators are completely absent. There are approximately 570 hyperaccumulator plant species found on earth, 450 of them are Ni hyperaccumulators. A few of the Ni accumulators have potential for phytoremediation or phytomining. In the present study, 19 different Brassicaceae members growing in serpentine habitats in the district of Yahyal?, Kayseri province, Turkey viz., members of genera Aethionema, Alyssum, Arabis, Heldreichia, Hesperis, Iberis, Isatis, Microthlaspi, Odontarrhena, Pseudosempervivum and Thlaspi were investigated. Nickel concentrations in the soil and underground and aboveground parts of plants were determined by using ICP-OES. It was observed that Ni concentrations of seven taxa (Odontarrhena muralis, O. oxycarpa, Isatis cappadocica subsp. cappadocica, Microthlaspi perfoliatum, Pseudosempervivum sempervivum, Thlaspi triangulare, Thlaspi rosulare) reach the threshold criterion of 1,000 mg kg?1 for Ni hyperaccumulation. In this study Ni concentrations in aboveground tissues of I. cappadocica subsp. cappadocica are determined as 5,587 mg kg?1 (in dry weight). Accordingly, it is suggested that this taxon be added to the list of Ni hyperaccumulator plants.  相似文献

The effect of defect locations on the mechanical properties of armchair graphene nanoribbons (AGNRs) and the various configurations of nitrogen (N) doping on the mechanical properties of AGNRs were examined using molecular dynamics (MD) simulations. The variation of the Young’s modulus (YM) and the ultimate tensile strength (UTS) of pyridinic-N, graphitic-N, and pyrrolic-N by increasing the concentration of N doping was investigated. The results of MD simulations show that the defect location has a significant effect on the UTS and failure strain (FS) of AGNRs in both vertical and horizontal directions. In the horizontal direction, variations of the UTS and FS are lower than in the vertical direction. On the other hand, the variations of the YM is almost similar in vertical and horizontal directions. The results of this work indicate that the UTS and FS of AGNRs are more sensitive than the YM of AGNRs for different defect directions. Pyridinic-N improves the mechanical properties of the defective AGNR and performs better YM and UTS values than the graphitic-N. Substitution N atoms, which are located at the defective sites and/or at the edges of AGNRs, are mechanically more favorable. Pyrrolic-N configuration has the lowest mechanical properties among the other configurations. Furthermore, pyrrolic-N with Stone-Wales-1 (SW-1) type of defect has higher mechanical properties than pyrrolic-N with Stone-Wales-2 (SW-2) type of defect.  相似文献

Computational studies of chondrocyte mechanics, and cell mechanics in general, have typically been performed using single cell models embedded in an extracellular matrix construct. The assumption of a single cell microstructural model may not capture intercellular interactions or accurately reflect the macroscale mechanics of cartilage when higher cell concentrations are considered, as may be the case in many instances. Hence, the goal of this study was to compare cell-level response of single and eleven cell biphasic finite element models, where the latter provided an anatomically based cellular distribution representative of the actual number of cells for a commonly used \(100 \, \upmu \hbox {m}\) edge cubic representative volume in the middle zone of cartilage. Single cell representations incorporated a centered single cell model and eleven location-corrected single cell models, the latter to delineate the role of cell placement in the representative volume element. A stress relaxation test at 10% compressive strain was adopted for all simulations. During transient response, volume- averaged chondrocyte mechanics demonstrated marked differences (up to 60% and typically greater than 10%) for the centered single versus the eleven cell models, yet steady-state loading was similar. Cell location played a marked role, due to inhomogeneity of the displacement and fluid pressure fields at the macroscopic scale. When the single cell representation was corrected for cell location, the transient response was consistent, while steady-state differences on the order of 1–4% were realized, which may be attributed to intercellular mechanical interactions. Anatomical representations of the superficial and deep zones, where cells reside in close proximity, may exhibit greater intercellular interactions, but these have yet to be explored.  相似文献

Several investigators have recently used fiberoptic cables to measure tendon forces in situ. The technique may be subject to significant error due to cable migration and differences in the loading rates used for calibration and those experienced during measurement. This in vitro study examined the impact of these potential sources of error on transducer accuracy. A fiberoptic cable was passed perpendicular to the fibers of four Achilles tendons in the mediolateral direction and each specimen was cyclically loaded to 1000 N. The influence of loading rate on transducer output was investigated by comparing results from tests conducted at 20, 200 and 1000 N/s. The effect of cable migration was examined by comparing the outputs obtained after displacing the cable one tendon width medially and laterally along its path in the tendon and then repeating the 200 N/s testing protocol. It was possible to obtain nonlinear specimen-specific relationships between the fiberoptic output and tendon force. Differences in loading rate resulted in root-mean-square (RMS) errors not larger than 17% maximum load. Hysteresis effects caused RMS errors smaller than 5% maximum load. Cable migration errors were less than 27%. The total RMS error due to the combined effects of loading rate difference and cable movement was less than 32%. Fiberoptic measurement of tendon force is attractive due to its low cost, easy implementation and comparable accuracy relative to other implantable force transducers. Although additional factors such as cable placement, edge artifacts due where the transducer exits the skin and non-uniform loading may also influence fiberoptic output, careful control of loading rate and transducer movement during calibration is imperative if maximum accuracy is to be achieved.  相似文献
