Despite increased social awareness, marketing restraints, tobacco taxation, and available smoking cessation rehab programs, active and passive smoking remain a worldwide challenging epidemic and a key risk factor for cardiovascular diseases development. Although cardiovascular (CV) protection is more pronounced in women than in men due to estrogenic effects, tobacco cigarette smoking exposure seems to alter this protection by modulating estrogen actions via undefined mechanisms. Premenopausal cigarette smoking women are at higher risk of adverse CV effects than non-smokers. In this study, we investigated the impact of cigarette smoking on early CV injury after myocardial infarction (MI) in non-menopausal female mice. Aortic arch calcification, fibrosis, reactive oxygen species, and gene expression of inflammatory and calcification genes were exaggerated in mice exposed to cigarette smoke (CS). These findings suggest that aortic injury following MI, characterized by vascular smooth muscle cells transdifferentiation, calcification, inflammation, and collagen deposition but not cardiac dysfunction is exacerbated with CS exposure. The novel findings of this study highlight the importance of aortic injury on short and long-term prognosis in CS-exposed MI females. Linking those findings to estrogen alteration is probable and entails investigation. 相似文献

Clustered regularly interspaced short palindromic repeats (CRISPR)-based genome editing, derived from prokaryotic immunity system, is rapidly emerging as an alternative platform for introducing targeted alterations in genomes. The CRISPR-based tools have been deployed for several other applications including gene expression studies, detection of mutation patterns in genomes, epigenetic regulation, chromatin imaging, etc. Unlike the traditional genetic engineering approaches, it is simple, cost-effective, and highly specific in inducing genetic variations. Despite its popularity, the technology has limitations such as off-targets, low mutagenesis efficiency, and its dependency on in-vitro regeneration protocols for the recovery of stable plant lines. Several other issues such as persisted CRISPR activity in subsequent generations, the potential for transferring to its wild type population, the risk of reversion of edited version to its original phenotype particularly in cross-pollinated plant species when released into the environment and the scarcity of validated targets have been overlooked. This article briefly highlights these undermined aspects, which may challenge the wider applications of this platform for improving crop genetics. 相似文献

The significant reduction in the plant growth was observed at an initial inoculum level of 2 immature females of Rotylenchulus reniformis/g soil and hence this inoculum level was considered as the minimum damaging threshold level for broccoli. The population of reniform nematode increased with the increasing levels of inoculum. Though, the maximum nematode population was recorded at the highest level of inoculum, the rate of reproduction of nematode was found to be highest at the lowest level of initial inoculum. Symptoms like chlorosis, stunted growth, shedding of leaves, early sprouting of inflorescence and sparsely developed roots were found during the experimental studies. Glasshouse experiment was conducted to assess the impact of green chopped leaves of four test plants (Lantana camara, Ficus virens, Kigelia pinnata and Ficus bengalensis) and two nematicides (Phorate and Carbofuran) on the plant growth parameters of tomato cv. K25 and on the root-knot development. Results revealed that all the tested treatments significantly (p = 0.05) improved plant growth parameters and reduced root-knot development compared to control. Among the tested organic additives, chopped green leaves of Lantana camara added to soil gave the highest enhancement in plant growth parameters, including plant height, fresh and dry weight, number of fruits and fruit weight with the values of 94.2 cm, 106.8 g, 31.6 g, 7.2 and 153.3 g respectively, as well as a greater reduction of Meloidogyne javanica reproduction and development but exhibiting a lower response compared to nematicides. Abstract A total of 106 Fusarium spp. were isolated from infected roots and soil samples of wheat and rice. Of the 106 isolates, 32 from wheat, and 74 from rice, were isolated. Six Fusarium spp. (F. oxysporum, F. moniliforme, F. poae, F. graminearum, F. tricinctum and F. equiseti) were identified at specie level. In aggressiveness tests Fusarium spp. root rot causing fungi were screened out into different aggressiveness classes according to disease severity scales. The aggressiveness of Fusarium spp. was studied on wheat varieties (Inqalab-91 and chakwal-86) and on rice varieties (Basmati-385 and IRRI-6) under controlled conditions. The overall total number of aggressive isolates was higher in rice than in wheat. However, the percentage of severely aggressive isolates was high in wheat, whereas the percentage of moderately and slightly aggressiveness isolates was high in rice. In rice, five isolates were non-aggressive and on wheat 17 were non-aggressive. Random Amplified Polymorphism DNAs (RAPDs) were used to study the polymorphism and genetic variations within the population of Fusarium spp. that established to study correlation between taxonomical and genetical characters of fungi. Five random primers were used P1 (5′-AGGAGGACCC-3′), P2 (5′-ACGAGGGACT-3′), PE7 (5′-AGATGCAGCC-3′), P14 (5′-CCACAGCACG-3′) and PE20 (5′-AACGGTGACC-3′). Each of the 10-mer primers produced results based on the respective banding patterns they generated in present investigations. Primers distinguished the F. oxysporum, F. moniliforme, F. graminearum, F. tricinctum, F. poa and F. equiseti. All the tested primers yielded amplification products, and that were reproducible. Although there was some intraspecific variation with primers, some strains were similar and some were different in banding pattern. In F. oxysporum, F. moniliforme, F. graminearum, F. tricinctum, F. poa and F. equiseti were seen clustered close to one another but each primer separated them unambiguously. All primer (P1, P2, P14, PE7 and PE20) combination produced 62 bands. All primers have shown interspecific and intraspecific variations in banding patterns. 相似文献

The effect of four opportunistic fungi viz., Paecilomyces lilacinus, Cladosporium oxysporum, Gliocladium virens and Talaromyces flavus on the life cycle of the root-knot nematode, Meloidogyne javanica, on brinjal was evaluated under glasshouse conditions. The results revealed that these fungi affected the penetration and development of M. javanica. The life cycle of M. javanica was delayed by 10, 7, 4 and 2 days in the presence of P. lilacinus, C. oxysporum, G. virens and T. flavus respectively. Fecundity, number of eggs per eggmass and number of larvae was also reduced in the presence of these opportunistic fungi. Pathogenic effect of root-knot nematode Meloidogyne arenaria was studied on balsam (Impatiens balsamina) by inoculating the different inoculum levels of root-knot nematode. It was observed that the inoculum levels up to 2000 J2 of root-knot nematode did not show significant reduction in plant growth characters as compared to control. Although the significant reduction in plant growth characters was recorded at and above 3000 J2 of root-knot nematode, progressive increase in the host infestation as indicated by the number of galls as well as the population of root-knot nematode was recorded with an increase in the level of inoculum. However, the rate of nematode multiplication was reduced with the increase in the inoculum density of M. arenaria. It can be concluded from these results that the damaging threshold level of M. arenaria on balsam was found to be as 3000 J2/plant. 相似文献

Abstract Phytophthora infestans is one of the most destructive pathogens of potato and causal agents of notorious disease late blight. Different chemicals are used to control the pathogen of late blight but the most commonly used is metalaxyl; its extensive use of has caused decreased sensitivity in the P. infestans population. The metalaxyl sensitivity of the Pakistani population of P. infestans is investigated in the present study. For this purpose, 178 isolates of P. infestans were obtained from the lesions of diseased potato leaves and stems, and samples were collected from the different potato-growing areas of Pakistan, where late blight is a problem. Sensitivity of the isolates of P. infestans was investigated by metalaxyl sensitivity test and with the help of test isolates were divided into three categories, i.e. sensitive, intermediate and resistant, based on their Co-efficient of mycelial growth inhibition (CMGI) values. During the study, highest percentage (50.17%) of resistant isolates was observed in the population of Punjab (zone 2), whereas the lower percentage (33.33%) was observed in the population of Swat valley (zone 6b). In the present study, it was discovered that P. infestans late blight-causing fungus has adopted more resistance against metalaxyl because of its wide use. 相似文献
