Wild and managed pollinators are the key component of biodiversity, contributing to important ecosystem services such as pollination and supporting human food security. Pollination by insects is a crucial component of the food chain that ensures the production of fruits and strongly affects the fruit quality, but the effect of insect pollination on fruit physiological and chemical parameters is largely unknown. The current study was conducted to determine the insect pollinators diversity and their relative abundance in the loquat (Eriobotrya japonica) orchard during 2017–2019. Further, the effect of insect pollinators pollination on the physiological and chemical parameters of fruit quality as compared to control pollinated flowers was investigated. The results revealed that a total of 22 species from 3 families (Apidae, Halictidae, and Syrphidae) were identified during the flowering season. The Apidae and Syrphidae were the most frequently observed families with major groups honey bees (67.89%) and hoverflies (21.57%), respectively. Moreover, results indicated that the fruit yield by the open-pollinated flowers (22.31 ± 0.34 kg/tree) was significantly higher than the control pollinated flowers (14.80 ± 0.25 kg/tree). Physiological and chemical parameters of loquat fruit differed significantly when fruits obtained from open-pollinated flowers as compared to control pollinated flowers. These results suggested that native insect pollinators play important role in the fruit quality of loquat. Hence, maintenance of appropriate habitat of native pollinators near loquat orchards is necessary to ensure good productivity and fruit quality.  相似文献

Eight newly synthesized carbacylamidophosphates with the general formula RC(O)NHP(O)Cl2 with R = pCl–C6H4 1a, pBr–C6H4 2a, C6H5 3a, and pMe–C6H4 4a and RC(O)NHP(O)(NC4H8O)2 R = pCl–C6H4 1b, pBr–C6H4 2b, C6H5 3b, pMe–C6H4 4b, were selected to compare the inhibition kinetic parameters, IC50, Ki, kp and KD, on human erythrocyte acetylcholinesterase (hAChE) and bovine serum butyrylcholinesterase (BuChE), Also, the in vivo inhibition potency of compound 2a, 2b and 3a, were studied. The data demonstrates that compound 2a and compound 2b are the potent sensitive as AChE and BuChE inhibitors respectively, and the inhibition of hAChE is about 10-fold greater than that of BuChE.  相似文献

Carbacylamidophosphates with the general formula RC(O)NHP(O)R1R2 constitute organophosphorus compounds that are used as insecticides, pesticides and ureas inhibitors. In this work, we studied the inhibition potency of CCl3C(O)NHP(O)Cl21, CHCl2C(O)NHP(O)Cl22, CH2ClC(O)NHP(O)Cl23 and CF3C(O)NHP(O)Cl24, which are the major intermediates for carbacylamidophosphates synthesis towards human erythrocyte acetylcholinesterase (hAChe) activity using Ellman's modified kinetic method. Unexpectedly, it was observed that they were not only hydrolytically unstable but also inhibited hAChE in a similar manner to that produced by organophosphorus insecticides. Enzymatic data, bimolecular inhibition rate constants (ki) and IC50 values for inhibition of hAChE demonstrated that they are irreversible inhibitors and the inhibition potency of compound 2 (IC50 = 88 μM) was the greatest in comparison with compounds 1, 3 and 4. Also the electropositivity of the phosphorus atom and the hydrophobicity of the compounds demonstrated that these two factors play an additional effect and different role in the inhibitory activity of these compounds. Hydrolytic stability of the compounds was determined by 31P NMR monitoring of the loss of the parent molecules with D2O as a function of time. This study considers antiacetylcholinesterase activity according to the structural and the electronic aspects of compounds 14, according to IR, 1H, 13C and 31P NMR spectral data.  相似文献

Hydrogen sulfide (H2S) and nitric oxide (NO) are major gasotransmitters produced in endothelial cells (ECs), contributing to the regulation of vascular contractility and structural integrity. Their interaction at different levels would have a profound impact on angiogenesis. Here, we showed that H2S and NO stimulated the formation of new microvessels. Incubation of human umbilical vein endothelial cells (HUVECs‐926) with NaHS (a H2S donor) stimulated the phosphorylation of endothelial NO synthase (eNOS) and enhanced NO production. H2S had little effect on eNOS protein expression in ECs. L‐cysteine, a precursor of H2S, stimulated NO production whereas blockage of the activity of H2S‐generating enzyme, cystathionine gamma‐lyase (CSE), inhibited this action. CSE knockdown inhibited, but CSE overexpression increased, NO production as well as EC proliferation. LY294002 (Akt/PI3‐K inhibitor) or SB203580 (p38 MAPK inhibitor) abolished the effects of H2S on eNOS phosphorylation, NO production, cell proliferation and tube formation. Blockade of NO production by eNOS‐specific siRNA or nitro‐L‐arginine methyl ester (L‐NAME) reversed, but eNOS overexpression potentiated, the proliferative effect of H2S on ECs. Our results suggest that H2S stimulates the phosphorylation of eNOS through a p38 MAPK and Akt‐dependent pathway, thus increasing NO production in ECs and vascular tissues and contributing to H2S‐induced angiogenesis.  相似文献

Background

Somatic mutations of the epidermal growth factor receptor (EGFR) are reportedly associated with various responses in non-small cell lung cancer (NSCLC) patients receiving the anti-EGFR agents. Detection of the mutation therefore plays an important role in therapeutic decision making. The aim of this study was to detect EGFR mutations in formalin fixed paraffin embedded (FFPE) samples using both Scorpion ARMS and high resolution melt (HRM) assay, and to compare the sensitivity of these methods.

Results

All of the mutations were found in adenocarcinoma, except one that was in squamous cell carcinoma. The mutation rate was 45.7% (221/484). Complex mutations were also observed, wherein 8 tumours carried 2 mutations and 1 tumour carried 3 mutations.

Conclusions

Both methods detected EGFR mutations in FFPE samples. HRM assays gave more EGFR positive results compared to Scorpion ARMS.  相似文献

Halyomorpha halys (Stål, 1855) is an invasive pest causing serious damage to agricultural crops in Europe and the USA. Very little is known about H. halys predators in Europe. This survey evaluated the potential of generalist predators/omnivorous species by means of predation bioassays in tri-dimensional arenas, where the predator had to locate H. halys prey items on the leaves of a bean plant. Eleven species of different taxa were tested and the prey items consisted in fresh eggmasses, 1st and 2nd instar nymphs. One species was also tested against adults. Some predators were species commercially available as biocontrol agents against plant pests, other predators were wild, captured in habitats shared with H. halys. All tested specimens were starved 24 h before starting the experiment. The survivorship of control prey items in predator-excluding cages was compared to that of predator treatment groups to determine the effect of predator presence. According to the results, the generalist species showed a quite low acceptance of H. halys prey items, since only two species caused 80% mortality on at least one item (Eupholidoptera chabrieri and Rhynocoris iracundus) and mortality due the other species never exceed 60%. Among commercially available species only Adalia bipunctata adults and Chrysoperla carnea larvae were effective, predating the eggs and 1st instar nymphs, respectively. Among the field collected specimens, the orthopteran E. chabrieri and the predatory hemipterans R. iracundus, Nagusta goedelii and Himacerus mirmicoides showed efficacy against 1st instar nymphs, E. chabrieri and R. iracundus showed efficacy against 2nd instar nymphs, whereas only E. chabrieri and N. goedelii predated the eggs. R. iracundus was also tested on the adults and successfully predated them. By identifying some of the species that can exploit H. halys as a suitable prey in southern Europe, the present investigation provides an important contribution for conservation biological control of this pest.

BackgroundBreast cancer is an extensively identified malignant tumor and is a prime cause of cancer mortalities in females. It has been shown that alteration of miRNAs expression (up or down regulation) can affect the initiation and progression of many malignancies. We aimed to evaluate the role of circulating miRNA-148a and miRNA-30c in female patients with breast cancer and estimate their usage as potential biomarkers in the diagnosis, prognosis and survival of breast cancer.MethodsThis study included 75 breast cancer female patients.They were compared with 55 apparently healthy female subjects. miRNAs expression analysis was assessed via real-time PCR.ResultsTo discriminate breast cancer patients from controls, miR-30c showed the best performance at a cut off value of ≤20.6 (AUC = 0.998, 97.33% sensitivity, 96.36% specificity, p < 0.001), followed by miR-148a (AUC = 0.995, 94.67% sensitivity, 90.91% specificity, p < 0.001 at a cut off value of ≤0.1), CA 15-3 (AUC = 0.930, 88.0% sensitivity, 81.82% specificity, p < 0.001 at a cut off value of >21.3), and finally CEA (AUC = 0.751, 70.67% sensitivity, 63.64% specificity, p < 0.001 at a cut off value of >2.5).ConclusionmiRNA-148a and miRNA-30c expressions were down regulated in female patients with breast cancer and might be considered as potential blood biomarkers. Both also might have rule in disease treatment and selection of therapeutic targets. Future studies are needed to improve their role in predicting response to treatment and prognosis.  相似文献

Recent investigations have demonstrated that human milk contains a variety of bacterial genera; however, as of yet very little work has been done to characterize the full diversity of these milk bacterial communities and their relative stability over time. To more thoroughly investigate the human milk microbiome, we utilized microbial identification techniques based on pyrosequencing of the 16S ribosomal RNA gene. Specifically, we characterized the bacterial communities present in milk samples collected from 16 women at three time-points over four weeks. Results indicated that milk bacterial communities were generally complex; several genera represented greater than 5% of the relative community abundance, and the community was often, yet not always, stable over time within an individual. These results support the conclusion that human milk, which is recommended as the optimal nutrition source for almost all healthy infants, contains a collection of bacteria more diverse than previously reported. This finding begs the question as to what role this community plays in colonization of the infant gastrointestinal tract and maintaining mammary health.  相似文献
