A disease in maize plants, not previously observed, appeared in 2003–2004 in Cosoleacaque, Tlalixcoyan and Paso de Ovejas counties, in the state of Veracruz, Mexico. Initial symptoms on leaves were small white‐yellow watery spots, which coalesced into dry necrotic stripes 0.3 wide and up to 8 cm long. Reddening sometimes developed on these leaves. Stems developed a rot in the crown. The flag leaf became rot and necrotic at the base, rolled inwards and dried out. Necrosis developed at the base of the corn ears and their growth was inhibited. These symptoms were initially observed in Asgrow‐7573 commercial maize plantings. A bacterium characterized by white colonies, gram negative, aerobic, rod‐shape, opaque, round colonies with entire margins on casaaminoacid peptone and glucose media was consistently isolated from diseased maize plants. On King’s Medium B, the isolates produced yellow, non‐mucoid colonies, with the majority of the strains secreting a diffusible yellow pigment into the media. The bacterium identity was confirmed by PCR amplification and sequencing of 16S and 23S genes rDNA fragments. The bacterium was identified as Burkholderia gladioli. Its pathogenicity on maize plants in Mexico is a new record.  相似文献

The mevalonate incorporation in vivo into total nonsaponifiable lipids by chick kidneys drastically increased after hatching, reaching similar levels to those previously observed in liver. Cholesterol was the major sterol formed from mevalonate from 11 days onward, while a fraction of polar nonsaponifiable lipid(s) was observed as the major compound(s) synthesized at 5-8 days. Relative percentages of squalene, squalene oxide(s) and lanosterol synthesized from mevalonate also increased between 11-18 days after hatching. Results in this paper demonstrate for the first time the accumulation of a fraction of nonsaponifiable lipid(s) identified as lanosterol derivatives and cholesterol precursors formed by kidneys from [5-14C]mevalonate in experiments carried out in vivo, as well as their evolution during postnatal period.  相似文献

Role of mitochondria in ethanol tolerance of Saccharomyces cerevisiae   总被引:7,自引:0,他引:7  
The presence of active mitochondria and oxidative metabolism is shown to be essential to maintain low inhibition levels by ethanol of the growth rate (), fermentation rate (v) or respiration rate () of Saccharomyces cerevisiae wild type strain S288C. Cells which have respiratory metabolism show K i (ethanol inhibition constant) values for , v and , higher (K i>1 M) than those of petite mutants or grande strains grown in anaerobiosis (K i=0.7 M). In addition, the relationship between or v and ethanol concentration is linear in cells with respiratory metabolism and exponential in cells lacking respiration. When functional mitochondria are transferred to petite mutants, the resulting strain shows K i values similar to those of the grande strain and the inhibition of and v by increasing ethanol concentrations becomes linear.  相似文献

Vitamin D deficiency is associated with wide range of pathologies. Some evidences have shown that low vitamin D circulating levels in children and adolescent are related to fat mass and obesity. The objectives of the present study were to characterize vitamin D status in children and adolescents and to determine if serum 25-hydroxyvitamin D (25(OH)D) concentration is related to adiposity assessed by body mass index (BMI). Serum 25(OH)D levels were measured by LIAISON method in 471 children and adolescents (2 to 18 years age) and analyzed according to gender, pubertal period, age, and BMI. An overall prevalence of 25(OH)D insufficiency and deficiency was present in the 67.1%. Lower 25(OH)D levels were found in females (25.56 ± 14.03 vs 29.71 ± 17.10 ng ml?1; P = 0.004) and pubertal children (25.52 ± 13.97 vs 29.21 ± 16.83 ng ml?1; P = 0.011). In addition, an inverse relation of BMI and age on 25(OH)D concentrations was observed in children. In conclusion, low vitamin D status was highly prevalent among children and adolescents. Of note, a non-lineal regression model showed that 39.6% of vitamin D levels variability was explained by BMI. These results indicate that adiposity assessed by BMI impacts vitamin D status.  相似文献

An understanding of biocontrol activities is important when developing microorganism‐based alternatives to conventional fungicides. From our bacterial collection, we selected two strains (BBC023 and BBC047) for their outstanding antagonistic capacity against fungal phytopathogens and growth‐promoting abilities towards Arabidopsis thaliana. According to physiological and molecular characterizations, both strains were classified as Bacillus amyloliquefaciens and were tested against Botrytis cinerea in vitro and in a tomato. Both strains secrete lipopeptide‐like compounds that contribute to their in vitro antagonism. SEM‐images showed altered B. cinerea mycelial structures that were consistent with previous reports of the direct action of lipopeptides against fungal hyphae. The strains were applied to the roots (R), leaves (foliar ‐ F) or root/leaves (R/F) on tomato plants. All treatments significantly reduced the severity of B. cinerea infection (measured as a control index). However, only root applications (R and R/F) led to growth promotion in the tomato plants. We detected the production of indole acetic acid (IAA) and 2,3‐butanediol as growth promotion traits in the two strains. For both strains, the R/F treatment showed the highest control index, suggesting a synergic effect of direct antagonism against B. cinerea and resistance induction in the plant. In addition, in vitro antagonism of BBC023 and BBC047 against B. cinerea was similar; whereas in the F application, strain BBC047 significantly improved plant resistance and maintained a higher population density over time on tomato leaves, compared to BBC023. BBC047 was also able to produce a complex and robust biofilm in Msgg medium compared with that of BBC023. We linked the reduced biocontrol of BBC023 on leaves with its limited ability to generate robust biofilms and colonize the phylloplane. At last, we highlight the potential of the native Bacillus strains as promising alternatives for the development of bioproducts for sustainable agriculture.  相似文献

Wildfires alter nitrogen (N) cycling in Mediterranean-type ecosystems, resetting plant and soil microbial growth, combusting plant biomass to ash, and enhancing N availability in the upper soil layer. This ash and soil N pool (that is, wildfire N) is susceptible to loss from watersheds via runoff and leaching during post-fire rains. Plant and soil microbial recovery may mitigate these losses by sequestering N compounds in new biomass, thereby promoting landscape N retention in N-limited chaparral ecosystems. We investigated the relative balance between wildfire N loss, and plant and soil microbial N uptake and stream N export for an upland chaparral watershed in southern California that burned (61%) in a high-intensity wildfire in 2009 by using a combination of stream, vegetation, soil microbial, and remote sensing analyses. Soil N in the burn scar was 440% higher than unburned soil N in the beginning of the first post-fire wet season and returned within 66 days to pre-fire levels. Stream N export was 1480% higher than pre-fire export during the first post-fire rain and returned within 106 days over the course of the following three rainstorms to pre-fire levels. A watershed-scale N mass balance revealed that 52% of wildfire N could be accounted for in plant and soil microbial growth, whereas 1% could be accounted for in stream export of dissolved nitrogen.  相似文献
