Analogues of the dsRNA poly(I).poly(C), namely double-stranded like complexes of poly(I) or poly(C), respectively, with corresponding vinyl nucleobases copolymerized with various vinyl compounds are highly effective interferon inducers in human lymphocyte cultures. Like poly(I).poly(C) they stimulate the liberation of an inhibitor(s) of interferon activity and, moreover, display mitogenic effect. The isolation and regeneration of tobacco mesophyll protoplasts from fully developed leaves, an important methodological step in plant genetic engineering as well as in plant cell biology and physiology, has been proven unreliable to the extent that it has become a significant setback to basic research. This unfortunate situation is primarily due to the suboptimal physiological state of greenhouse-grown protoplast donor plants. A technically simple and inexpensive method, based on the utilization of commercial Phototron units, is described for the production of suitable tobacco donor plants. Furthermore, a modified version of such a culture unit can be used to regenerate plants from protoplast-derived calli.  相似文献

The present study aimed to determine the chemical composition and biological activity of the essential oil obtained from Phyllogonium viride Brid. (Phyllogoniaceae, Bryophyta), whose samples were collected in southern Brazil. For the first time, the cytotoxic activity of the essential oil of P. viride in breast and colorectal tumor cells (MCF-7 and HCT-116) was evaluated, as well as the cytotoxic potential of this oil in non-tumoral cells of human immortalized keratinocytes (HaCaT) via MTT assay. The compounds majorly found in P. viride essential oil were β-bazzanene (20.30 %), β-caryophyllene (17.06 %), β-chamigrene (14.02), and germacrene B (11.72 %). Treatment with P. viride essential oil in the different tested cell lines did not induce any toxicity in most of the tested concentrations. These data contribute to generating new scientific information about this understudied plant species. Furthermore, the chemical characterization of the compounds present in the essential oil of P. viride can lead to greater elucidation of its biotechnological potential.  相似文献

The anticonvulsant activity of U-54494A was studied in a 4-aminopyridine (4-AP) epilepsy model using extracellular recordings in in vitro hippocampal slices. Field potentials were evoked by stimulation of Schaffer collaterals, and recorded from the CA1 region of the hippocampus after infusion of 4-AP in the absence and presence of U-54494A. The number and the total area of after discharges (AD) in the presence of 4-AP were significantly decreased by increasing concentrations of U-54494A. In contrast, U-54494A did not significantly change the latency, duration, or area of the evoked PS in this paradigm. Phenytoin, a standard anticonvulsant, decreased the PS area without affecting either the PS latency or duration, or the AD number or area in the same paradigm. These present results provide more evidence that U-54494A is a novel and effective anticonvulsant that may be useful in the treatment of paroxysmal activity, without having generalized depressive effects.  相似文献

We present the ontogeny of the integrated musculoskeletal complex that comprises the pelvic girdle and hind limbs of anurans. Our histological data show that the pelvic girdle originates from a single mesenchymatic condensation. The tissue differentiation sequence is cartilage, muscle and tendon. The intrusion of the ischiadic nerve into the limb bud is produced very early in ontogeny. The pre‐cartilage appears in the pre‐motile stage. Therefore, the nerve produces a movement analogous to the ‘embryonic motility’ that would induce the emergence of the pre‐cartilage. The acetabulum is the first of all cavitation processes to form, the second one being the knee. The acetabulum appears before the muscles are mature, although it has been stressed that the muscle contraction maintains joint progenitors committed to their fate. Our data indicate an explosive differentiation of all 11 muscular masses together. We provide three new characters that support the monophyly of Hyloides, Acosmanura and Neobatrachia.  相似文献

The behavior of endogenous polyamines was studied in somatic embryos and zygotic embryos of Habanero pepper (Capsicum chinense). In the first part of the work, the polyamine content was evaluated in both types of embryos (somatic and zygotic). As a result, in addition to the common polyamines (putrescine, spermidine and spermine), it was also possible to detect cadaverine, a polyamine rarely found in plants. In general, all the polyamines were found to be more abundant in somatic embryos than in zygotic embryos, with significantly higher contents of putrescine and cadaverine. Subsequently, the content of putrescine, spermidine, spermine and cadaverine, in their different forms (free, bound and conjugated) was determined in somatic embryos which were cultured in non-ventilated and ventilated containers. Detection of polyamines was carried out at 28 and 42 days of culture by the HPLC method. The ethylene content was monitored during the process in both culture conditions (ventilated and non-ventilated). As a result of the analysis, cadaverine was always found present, indicating that it is a common polyamine in the species. Ethylene was detected in containers without ventilation throughout the culture, except during replenishment of the culture medium (R1, R2 and R3). The behavior pattern of each polyamine, analyzed under different culture conditions (ventilated and non-ventilated) and at different moments of culture (28 and 42 days of culture), show that the polyamines are not only involved in morphogenic processes in plants; polyamines are also significantly affected by the surrounding environment. However, the most novel result, presented for the first time in this paper, is that cadaverine is found to be a common polyamine in C. chinense since it is present in both zygotic embryos and somatic embryos.

A total of 243 samples from Hemlock-Hardwood, Boreal Forest, and Alpine Tundra associations of New York, Vermont, New Hampshire, and Maine were analyzed for species of Bakernema, Criconema, and Criconemoides and for selected edaphic factors. The Hemlock-Hardwood formation contained 13 species of these genera, but the Boreal Forest and Alpine Tundra contained only Criconema menzeli and Criconemoides sphagni. Criconemoides axeste, C. rusticum, and C. xenoplax were associated primarily with mineral soils that have high pH, low moisture after drainage, and organic matter content of less than 15%. Criconemoides sphagni was associated with organic soils that had low pH, high moisture after drainage, and organic matter content greater than 15%.  相似文献
