For landfarming of oil refinery waste to be sustainable, it is crucial to know the toxicity and chemical composition of the waste. We determined the chemical composition of waste known as American Petroleum Institute sludge and tested its toxicity in two different soils using the earthworm Eisenia andrei (Oligochaeta) in standardized bioassays. We compared the chemical composition of the sludge with available chemical profiles of the landfarming site where sludge of the same origin had been landfarmed for several years. The concentrations of Al, Mn, Pb, S and Zn and diesel range organics were higher in the landfarmed soil than in the sludge, indicating possible accumulation in the soil over time. Although no traces of volatile organic compounds were previously found in the landfarming site soil, we found high levels in the American Petroleum Institute sludge, indicating that these volatiles may not pose a long-term problem in the soil. Polycyclic aromatic hydrocarbons and gasoline range organics were present in the sludge at higher concentrations than in the site soil, indicating that their remediation through landfarming may have had some degree of success.

Earthworm biomass and reproduction were detrimentally affected at low sludge concentrations, which could have population level effects over time in a landfarming site.  相似文献

Summary Early development ofPlatynereis dumerilii is characterized by an extremely constant cleavage pattern in which the volumes and cytoplasmic contents of the blastomeres show remarkably little variability (Dorresteijn 1990). In order to test the necessity of a precise partitioning of the cytoplasm, we have stratified the ooplasm by mild centrifugation (10 min at 300 g) after completion of meiosis but before first cleavage. The cytoplasm of the zygote stratifies randomly with respect to the pre-existing animal-vegetal axis, but first cleavage follows the animal-vegetal axis dividing the plasm before it has rearranged to its normal distribution. As usual, first cleavage is unequal in the majority of centrifuged eggs. Different sorts of cytoplasm are always distributed abnormally in comparison to normal two-cell stages. Under two circumstances this leads to the formation of double trunk structures in the young worm. Such double monsters either originate from zygotes whose clear cytoplasm has been distributed equally to the two daughter blastomeres at first cleavage, or from unequal two-cell embryos whose larger blastomere cleaved equally at second cleavage forming blastomeres with equal lots of clear cytoplasm. Cell-lineage could be followed in an individual embryo of the latter category and showed the existence of two adjacent D-quadrants, giving rise to a double monster with a forked trunk. Embryos of the former category give rise to two opponent D-quadrants and double monsters with a four-sided trunk. As in the normal embryo, the amount of clear cytoplasm in a blastomere is positively correlated with the speed of its cell cycle, and endows the cell with D-quadrant developmental capacities as can be judged by the cleavage pattern.  相似文献

Molecular pathogen detection from blood is still expensive and the exact clinical value remains to be determined. The use of biomarkers may assist in preselecting patients for immediate molecular testing besides blood culture. In this study, 140 patients with ≥ 2 SIRS criteria and clinical signs of infection presenting at the emergency department of our hospital were included. C-reactive protein (CRP), neutrophil-lymphocyte count ratio (NLCR), procalcitonin (PCT) and soluble urokinase plasminogen activator receptor (suPAR) levels were determined. One ml EDTA blood was obtained and selective pathogen DNA isolation was performed with MolYsis (Molzym). DNA samples were analysed for the presence of pathogens, using both the MagicPlex Sepsis Test (Seegene) and SepsiTest (Molzym), and results were compared to blood cultures. Fifteen patients had to be excluded from the study, leaving 125 patients for further analysis. Of the 125 patient samples analysed, 27 presented with positive blood cultures of which 7 were considered to be contaminants. suPAR, PCT, and NLCR values were significantly higher in patients with positive blood cultures compared to patients without (p < 0.001). Receiver operating characteristic curves of the 4 biomarkers for differentiating bacteremia from non-bacteremia showed the highest area under the curve (AUC) for PCT (0.806 (95% confidence interval 0.699–0.913)). NLCR, suPAR and CRP resulted in an AUC of 0.770, 0.793, and 0.485, respectively. When compared to blood cultures, the sensitivity, specificity, positive predictive value (PPV), and negative predictive value (NPV) for SepsiTest and MagicPlex Sepsis Test were 11%, 96%, 43%, 80%, and 37%, 77%, 30%, 82%, respectively. In conclusion, both molecular assays perform poorly when one ml whole blood is used from emergency care unit patients. NLCR is a cheap, fast, easy to determine, and rapidly available biomarker, and therefore seems most promising in differentiating BSI from non-BSI patients for subsequent pathogen identification using molecular diagnostics.  相似文献

Cytokines play an important role in ischemic injury and repair. However, little is known about their prognostic value in cardiovascular disease. The aim of this study was to investigate the prognostic importance of chemokines CCL3/MIP-1α, CCL5/RANTES and CCL18/PARC for the risk of future cardiovascular events in patients with acute coronary syndromes (ACS). Baseline levels of CCL3/MIP-1α, CCL5/RANTES and CCL18/PARC were determined in ACS patients from the Bad Nauheim ACS II registry (n = 609). During the following 200 days, patients were monitored for the occurrence of fatal and non-fatal cardiovascular events. Patients with CCL3/MIP1α, CCL5/RANTES and CCL18/PARC concentrations in the highest tertile were associated with an increased risk of a fatal event during follow-up (HR: 2.19, 95%CI: 1.04–4.61 for CCL3/MIP1α, HR: 3.45, 95%CI: 1.54–7.72 for CCL5/RANTES and HR: 3.14, 95%CI: 1.33–7.46 for CCL18/PARC). This risk was highest for patients with all three biomarkers concentrations in the upper tertile (HR: 2.52, 95%CI: 1.11–5.65). Together with known risk predictors of cardiovascular events, CCL3/MIP-1α, CCL5/RANTES and CCL18/PARC combined improved the c-statistics from 0.74 to 0.81 (p = 0.007). In conclusion, CCL3/MIP-1α, CCL5/RANTES and CCL18/PARC are independently associated with the risk of short-term mortality in ACS patients. Combining all three biomarkers further increased their prognostic value.  相似文献

Abstract The in situ method for determination of reduction levels of cytochromes b and c pools during steady-state growth (Pronk et al., Anal. Biochem. 214, 149–155, 1993) was applied to chemostat cultures of the wild-type, a cytochrome aa3 single mutant and a cytochrome aa3/d double mutant of Azorhizobium caulinodans . For growth with NH4+ as the N source, the results indicate that (i) the aa3 mutant strains growing at a dissolved O2 tension of 0.5% possess an active alternative cytochrome c oxidase, which is hardly present during fully aerobic growth, and assuming that (i) also pertains to the wild-type, (ii) the wild-type uses cytochrome aa3 under fully aerobic conditions. For growth with N2 as the N source, it was found that the aa3 mutant strains growing at dissolved O2 tensions ranging from 0.5 to 3.0% also contain an active alternative cytochrome c oxidase.  相似文献

A selection scheme was devised to isolate Paracoccus denitrificans mutants with increased recipient qualities in transfer experiments, using broad host range plasmids. In some of the mutants obtained, a DNA modifying activity that prevents the activity of the restriction endonucleases BamHI and BglII on isolated P. denitrificans DNA had simultaneously been lost. From a detailed analysis of the restriction properties of the enzymes SAU3 AI, MboI and DpnI, it was concluded that a subset of GATC sequences in P. denitrificans DNA may be methylated at an unusual position. It was concluded that P. denitrificans possesses at least one potent host-dependent restriction/modification system which affects conjugation. In addition to the class of restriction-defective mutants, at least one other class of enhanced transfer mutants with unknown defect(s) was isolated. Strains, in which the two mutant classes were combined, exhibited transfer frequencies which were significantly higher than strains containing either mutation alone. Such double mutant strains appeared to be well suited for future experiments like complementation analysis, transposon mutagenesis and gene replacement by homologous recombination.  相似文献

Weather surveillance radars are increasingly used for monitoring the movements and abundances of animals in the airspace. However, analysis of weather radar data remains a specialised task that can be technically challenging. Major hurdles are the difficulty of accessing and visualising radar data on a software platform familiar to ecologists and biologists, processing the low‐level data into products that are biologically meaningful, and summarizing these results in standardized measures. To overcome these hurdles, we developed the open source R package bioRad, which provides a toolbox for accessing, visualizing and analyzing weather radar data for biological studies. It provides functionality to access low‐level radar data, process these data into meaningful biological information on animal speeds and directions at different altitudes in the atmosphere, visualize these biological extractions, and calculate further summary statistics. The package aims to standardize methods for extracting and reporting biological signals from weather radars. Here we describe a roadmap for analyzing weather radar data using bioRad. We also define weather radar equivalents for familiar measures used in the field of migration ecology, such as migration traffic rates, and recommend several good practices for reporting these measures. The bioRad package integrates with low‐level data from both the European radar network (OPERA) and the radar network of the United States (NEXRAD). bioRad aims to make weather radar studies in ecology easier and more reproducible, allowing for better inter‐comparability of studies.  相似文献

The development of gut microbiota during ontogeny is emerging as an important process influencing physiology, immunity and fitness in vertebrates. However, knowledge of how bacteria colonize the juvenile gut, how this is influenced by changes in the diversity of gut bacteria and to what extent this influences host fitness, particularly in nonmodel organisms, is lacking. Here we used 16S rRNA gene sequencing to describe the successional development of the faecal microbiome in ostriches (Struthio camelus, n = 66, repeatedly sampled) over the first 3 months of life and its relationship to growth. We found a gradual increase in microbial diversity with age that involved multiple colonization and extinction events and a major taxonomic shift in bacteria that coincided with the cessation of yolk absorption. Comparisons with the microbiota of adults (n = 5) revealed that the chicks became more similar in their microbial diversity and composition to adults as they aged. There was a five‐fold difference in juvenile growth during development, and growth during the first week of age was strongly positively correlated with the abundance of the genus Bacteroides and negatively correlated with Akkermansia. After the first week, the abundances of six phylogenetically diverse families (Peptococcaceae, S24‐7, Verrucomicrobiae, Anaeroplasmataceae, Streptococcaceae, Methanobacteriaceae) were associated with subsequent reductions in chick growth in an age‐specific and transient manner. These results have broad implications for our understanding of the development of gut microbiota and its associations with animal growth.  相似文献

Acontias plumbeus has traditionally been considered a monotypic, invariable species, a fact that highly contrasts with documented examples of high phylogenetic complexity and phenotypic diversity in other members of the Acontinae. We employed mitochondrial and nuclear DNA markers to investigate genetic structuring among A. plumbeus populations and the relationship between A. plumbeus and the closely related A. poecilus. Molecular genetic analyses revealed three clades with non‐overlapping distributions: an Eastern clade, a widely distributed Northern clade, and a Southern clade that includes topotypical A. poecilus. Morphometric analyses of preserved specimens showed that Southern clade populations are comprised of individuals with absolute and proportionally smaller body sizes than their Northern and Eastern relatives. Phylogenetic affinities within A. plumbeus indicate a complex biogeographic scenario within South Africa and suggest that A. poecilus should be considered a junior synonym of A. plumbeus instead of a truly valid species.  相似文献

The structure-specific endonuclease XPG is an indispensable core protein of the nucleotide excision repair (NER) machinery. XPG cleaves the DNA strand at the 3' side of the DNA damage. XPG binding stabilizes the NER preincision complex and is essential for the 5' incision by the ERCC1/XPF endonuclease. We have studied the dynamic role of XPG in its different cellular functions in living cells. We have created mammalian cell lines that lack functional endogenous XPG and stably express enhanced green fluorescent protein (eGFP)-tagged XPG. Life cell imaging shows that in undamaged cells XPG-eGFP is uniformly distributed throughout the cell nucleus, diffuses freely, and is not stably associated with other nuclear proteins. XPG is recruited to UV-damaged DNA with a half-life of 200 s and is bound for 4 min in NER complexes. Recruitment requires functional TFIIH, although some TFIIH mutants allow slow XPG recruitment. Remarkably, binding of XPG to damaged DNA does not require the DDB2 protein, which is thought to enhance damage recognition by NER factor XPC. Together, our data present a comprehensive view of the in vivo behavior of a protein that is involved in a complex chromatin-associated process.  相似文献
