Background
Mechanical ventilation can promote lung injury by triggering a pro-inflammatory response. Macrolides may exert some immunomodulatory effects and have shown significant benefits over other antibiotics in ventilated patients. We hypothesized that macrolides could decrease ventilator-induced lung injury.Methods
Adult mice were treated with vehicle, clarithromycin or levofloxacin, and randomized to receive mechanical ventilation with low (12 cmH2O, PEEP 2 cmH2O) or high (20 cmH2O, ZEEP) inspiratory pressures for 150 minutes. Histological lung injury, neutrophil infiltration, inflammatory mediators (NFκB activation, Cxcl2, IL-10) and levels of adhesion molecules (E-selectin, ICAM) and proteases (MMP-9 and MMP-2) were analyzed.Results
There were no differences among groups after low-pressure ventilation. Clarithromycin significantly decreased lung injury score and neutrophil count, compared to vehicle or levofloxacin, after high-pressure ventilation. Cxcl2 expression and MMP-2 and MMP-9 levels increased and IL-10 decreased after injurious ventilation, with no significant differences among treatment groups. Both clarithromycin and levofloxacin dampened the increase in NFκB activation observed in non-treated animals submitted to injurious ventilation. E-selectin levels increased after high pressure ventilation in vehicle- and levofloxacin-treated mice, but not in those receiving clarithromycin.Conclusions
Clarithromycin ameliorates ventilator-induced lung injury and decreases neutrophil recruitment into the alveolar spaces. This could explain the advantages of macrolides in patients with acute lung injury and mechanical ventilation. 相似文献83.

Marigold (Tagetes erecta) flowers are primarily used in industry for their high pigment content. Flower color development implies that chloroplast–chromoplast
transition is associated with carotenoid biosynthesis. We report the recovery of undifferentiated pigmented marigold cells,
various callus tissues, and their analysis by transmission electron microscopy in order to observe accumulating pigment and
development of subcellular structures. Callus was generated from leaf explants and after several rounds of recurrent selection.
Green-, yellow-, and brown-colored callus were obtained that showed distinct carotenoid profiles. For green material, violaxanthin,
lutein, zeaxanthin, and β-carotene were produced, while yellow callus generated mainly lutein, as did the brown callus. Chloroplast–chromoplast
transition was followed by measuring plastid size and shape in undifferentiated marigold cells by digital image analysis.
Cellular alterations were evident in brown callus. Chloroplasts were the main structure in green callus, while yellow callus
clearly showed the formation of plastoglobules, structures that are correlated with chloroplast–chromoplast transition. The

The isotopic composition of the remains of cereals and pine has been studied, from three different chronological phases from 2140 to 1500 cal bc at the Terlinques site, southeast Iberian Peninsula. The δ13C values range between ?24.91 and ?21.19 ‰ (V-PDB), with an average of ?23.05 ‰ (STD = 0.69). The archaeological cereals show an average isotopic discrimination (Δ13C) with the past atmospheric CO2 of 16.96 ‰, which is much greater than the average Δ13C of 13.89 ‰ of the rainfed Triticeae (wheat and barley) in modern times. However, considering the effect of the atmospheric CO2 concentration, which is included in the WUEi (intrinsic water-use efficiency), this difference is even greater, 77 for archaeological samples versus 144 for present-day rainfed cereals. This could represent some of the earliest evidence of the use of irrigation techniques in Europe. Modern cereals which have been irrigated show a general Δ13C average of 17.17 ‰, very similar to those of the middle Holocene. However, when the WUEi is calculated, the value of 108 indicates that present-day irrigated cereals are more stressed than the archaeological samples. For comparison, we have included pine trees, since these have an extensive root development which is capable of reaching the water table. In the past, both cereals and pine present similar WUEi values (77 vs. 72), however at present only irrigated cereals show similar WUEi values to pine (108 vs. 107). This again suggests irrigation of cereals in the past. The processes of climatic degradation towards drier conditions which started in the middle Holocene could be responsible for the use of land near water sources, on riverbanks and near shallow lakes. According to the isotopic and plant macrofossil data, irrigation or water management techniques were used at the Terlinques site, located close to the Laguna de Villena, a lake which has now dried out. 相似文献

Chronic myelogenous leukemia (CML) is a myeloproliferative disorder characterized at the molecular level by the expression of Bcr-abl, a 210-kDa fusion protein with deregulated tyrosine kinase activity. Encouraged by the clinical validation of Bcr-abl as the target for the treatment of CML by imatinib, we sought to identify pharmacological agents that could target this kinase by a distinct mechanism. We report the discovery of a new class of Bcr-abl inhibitors using an unbiased differential cytotoxicity screen of a combinatorial kinase-directed heterocycle library. Compounds in this class (exemplified by GNF-2) show exclusive antiproliferative activity toward Bcr-abl-transformed cells, with potencies similar to imatinib, while showing no inhibition of the kinase activity of full-length or catalytic domain of c-abl. We propose that this new class of compounds inhibits Bcr-abl kinase activity through an allosteric non-ATP competitive mechanism. 相似文献

The use of correlative analyses might be insufficient to understand the processes that control biodiversity, because the variables accounting for different hypotheses (e.g. current climate, past climate change, post‐glacial dispersal limitation) are mutually correlated. We suggest here that, in order to gain insight, it could be useful to search for latitudinal thresholds that could provide information about qualitative changes in the way biodiversity varies in space. Such tipping points could inform about higher‐level processes that are not reflected in correlative analyses. We test whether similar breakpoints in latitudinal beta‐diversity patterns exist for different vertebrate groups with diverse life histories and dispersal abilities. In birds, bats and non‐volant mammals we find breakpoints similar to those of amphibians. Differences in species composition are mainly due to species replacement from the equator to the breakpoint, but are dominated by nested species losses from the breakpoint to higher latitudes. Thus, marked thresholds discriminate two world regions where different processes appear to drive biodiversity. 相似文献

Our aim was to measure the correlation between fetal electrocardiographic (FECG) recordings of low-risk pregnancies and polysomnographic (PSG) study parameters in low-risk infants born at term as a measurement of perinatal sleep-development continuity.We designed a short, prospective, observational follow-up of physiologic parameters between fetuses and newborns. We studied 10 fetuses from low-risk pregnant female out-patients and the same subjects as low-risk newborns delivered at term. Fetal state (FS) was defined in FECG recordings reassembling the following: fetal state I (quiet sleep or QS); fetal state II (active sleep or AS); fetal state III (quiet waking), and fetal state IV (active waking). Percentages of AS, QS, and wakefulness in PSG studies of newborns were also determined.Comparisons of FS I with QS showed a significant reduction in QS, while comparison of FS II with AS showed significant reduction in AS. Negative correlations were found between FS I with QS, and FS II with AS. Number of cycles in FECG recordings and PSG sleep cycles also demonstrated significant correlation.In conclusion our data showed partial but significant sleep function continuity from fetal to neonatal period. 相似文献

The aim of this review is to survey the various naturally occurring phenanthrene compounds that have been isolated from different plants. Only one review has previously been published on this topic. Gorham (1989) reviewed the structures, biosynthesis, separations and spectroscopy of stilbenes and phenanthrenes. The present study furnishes an overview of the hydroxy or/and methoxy-substituted 9,10-dihydro/phenanthrenes, methylated, prenylated and other monomeric derivatives, dimeric and trimeric phenanthrenes and their biological activities. A fairly large number of phenanthrenes have been reported from higher plants, mainly in the Orchidaceae family, in the species Dendrobium, Bulbophyllum, Eria, Maxillaria, Bletilla, Coelogyna, Cymbidium, Ephemerantha and Epidendrum. A few phenanthrenes have been found in the Hepaticae class and Dioscoreaceae, Combretaceae and Betulaceae families. Their distribution correlates strongly with the taxonomic divisions. These plants have often been used in traditional medicine, and phenanthrenes have therefore been studied for their cytotoxicity, antimicrobial, spasmolytic, anti-inflammatory, antiplatelet aggregation, antiallergic activities and phytotoxicity. On the basis of 120 references, this review covers the phytochemistry and pharmacology of phenanthrenes, describing 252 compounds.
