Mosquitoes, especially Aedes aegypti, are becoming important models for studying invasion biology. We characterized genetic variation at 12 microsatellite loci in 79 populations of Ae. aegypti from 30 countries in six continents, and used them to infer historical and modern patterns of invasion. Our results support the two subspecies Ae. aegypti formosus and Ae. aegypti aegypti as genetically distinct units. Ae. aegypti aegypti populations outside Africa are derived from ancestral African populations and are monophyletic. The two subspecies co‐occur in both East Africa (Kenya) and West Africa (Senegal). In rural/forest settings (Rabai District of Kenya), the two subspecies remain genetically distinct, whereas in urban settings, they introgress freely. Populations outside Africa are highly genetically structured likely due to a combination of recent founder effects, discrete discontinuous habitats and low migration rates. Ancestral populations in sub‐Saharan Africa are less genetically structured, as are the populations in Asia. Introduction of Ae. aegypti to the New World coinciding with trans‐Atlantic shipping in the 16th to 18th centuries was followed by its introduction to Asia in the late 19th century from the New World or from now extinct populations in the Mediterranean Basin. Aedes mascarensis is a genetically distinct sister species to Ae. aegypti s.l. This study provides a reference database of genetic diversity that can be used to determine the likely origin of new introductions that occur regularly for this invasive species. The genetic uniqueness of many populations and regions has important implications for attempts to control Ae. aegypti, especially for the methods using genetic modification of populations.  相似文献

Higher plant hydraulic conductivity (K plant) is vital for plant growth, especially under PEG-induced water deficit stress (PEG-IWDS). Leaf venation architecture is a key determinant of leaf hydraulic conductivity (K leaf) and K leaf is a major component of K plant across different plant species. However, there is little information about (1) varietal difference in leaf vein development in cereal crops, such as rice plants; (2) the effects of PEG-IWDS on leaf vein development; (3) the coordination between leaf venation architecture and K plant as well as K leaf under PEG-IWDS. In the present study, widely cultivated eight rice cultivars were grown hydroponically under well-watered condition (WWC) and PEG-IWDS, simulated by adding 15 % (w/v) PEG6000. Leaf venation architecture, including total longitudinal leaf vein number, leaf vein numbers per unit width (LVNW), vein thickness and leaf mass per area, as well as K plant and K leaf were measured to address above-mentioned questions. The results showed that leaf venation architecture exhibited significant varietal differences and PEG-IWDS significantly increased LVNW while decreased vein thickness. PEG-IWDS suppressed both K plant and K leaf but the decrease was much higher in K plant than K leaf. There was a significant and positive correlation observed between LVNW and K leaf under both WWC and PEG-IWDS but the correlation between LVNW and K plant was only significant under WWC. K leaf was significantly and positively correlated with K plant under WWC but not under PEG-IWDS. It is concluded that K leaf is a major determinant for K plant under WWC but not under PEG-IWDS; therefore, breeding or selecting rice cultivars with high LVNW can improve shoot water supplement under WWC but not under PEG-IWDS condition.  相似文献

A rapid, sensitive, and simple method was developed to detect the sapstain fungi Ophiostoma piceae and O. quercus in stained wood. By using microwave heating for DNA extraction and PCR with internal transcribed spacer-derived-specific primers, detection was feasible within 4 h, even with DNA obtained from a single synnema. Environmental stresses from climate change can alter source–sink relations during plant maturation, leading to premature senescence and decreased yields. Elucidating the genetic control of natural variations for senescence in wheat (Triticum aestivum) can be accelerated using recent developments in unmanned aerial vehicle (UAV)-based imaging techniques. Here, we describe the use of UAVs to quantify senescence in wheat using vegetative indices (VIs) derived from multispectral images. We detected senescence with high heritability, as well as its impact on grain yield (GY), in a doubled-haploid population and parent cultivars at various growth time points (TPs) after anthesis in the field. Selecting for slow senescence using a combination of different UAV-based VIs was more effective than using a single ground-based vegetation index. We identified 28 quantitative trait loci (QTL) for vegetative growth, senescence, and GY using a 660K single-nucleotide polymorphism array. Seventeen of these new QTL for VIs from UAV-based multispectral imaging were mapped on chromosomes 2B, 3A, 3D, 5A, 5D, 5B, and 6D; these QTL have not been reported previously using conventional phenotyping methods. This integrated approach allowed us to identify an important, previously unreported, senescence-related locus on chromosome 5D that showed high phenotypic variation (up to 18.1%) for all UAV-based VIs at all TPs during grain filling. This QTL was validated for slow senescence by developing kompetitive allele-specific PCR markers in a natural population. Our results suggest that UAV-based high-throughput phenotyping is advantageous for temporal assessment of the genetics underlying for senescence in wheat.

The study was conducted to assess whether selenium (Se) application modulates fertility to alter grain yield in bread wheat grown under different moisture regimes. Seeds of wheat cultivar Millat-2011 were sown in the plots using a randomized complete block design with three replicates per treatment. After germination, the plants were exposed to six moisture regimes, viz. no irrigation after germination, irrigation at boot stage, irrigations at boot and grain-filling stages, irrigations at crown root, boot and grain-filling stages, irrigations at crown root, boot, heading and grain-filling stages and irrigations at crown root, stem elongation, boot, heading and grain-filling stages. At the heading stage, foliar spray of sodium selenate (0, 2 and 4 mg Se L?1) was done. Withholding water at early growth stages significantly increased oxidative stress and decreased growth and grain yield. Irrespective of moisture regimes, foliar application of Se (2 mg L?1) decreased oxidative stress, modulated photosynthetic pigments and fertility and increased grain yield in wheat. The Se-mediated increase in grain yield was attributed to the increase in chlorophyll and ascorbic acid contents and fertility coupled with decrease of oxidative stress under different moisture regimes. The results could be helpful to manage wheat production in the semi-arid environments.  相似文献

Analyzing chromosomal traits is one of the pragmatic ways to establish evolutionary and genetic database of plants that has complicated phylogenetic system. There are some conflicts on the exact phylogeny and evolutionary pathway of Lilium, and section martagon is the most complicated part among them. In this study, chromosomal traits of martagon lily species are described. All martagon lilies were analyzed with FISH (Fluorescence in situ hybridization) technique, followed by detailed karyotyping. Each species showed 2n = 2x = 24 of chromosome complement. Size of chromosomes ranged from 451.04 to 680.06 µm. 5S and 45S ribosomal DNA, general molecular markers in modern evolutionary research were used as probe in this study. Variation in rDNA loci and chromosome translocation were observed in Lilium hansonii; the highest number of 45S rDNA loci was detected in Lilium hansonii, followed by other martagon lilies, in similar locations but with differences, and chromosome translocation was observed from one individual of Lilium hansonii. Additionally, Lilium tsingtauense from Jeju-do Island, Korea was detected with two extra chromosomes. These kind of genetic variations through karyotyping indicate ongoing genetic variations in martagon lilies. In this study, precise analysis of chromosome traits in Lilium species belonging to section martagonperformed to contribute to better comprehension of the evolutionary pathway and establishment of cytogenetic database for further plant breeding research.  相似文献

Selective alkylation of dyphylline (1) with (2-acetoxyethoxy)methyl bromide (2a) or 4-acetoxybutyl bromide (2b) afforded 3'-O-[(acetoxyethoxy)methyl]dyphylline (3a) and 3'-O-(4-acetoxybutyl)-dyphylline (3b), respectively. A trans esterification process rather than alkylation of the dihydroxy-propyl side chain in 1 had taken place during the reaction with 2-p-toluoyloxy)ethyl chloride (5) to afford the respective 3'-toluoyloxy derivative 7 and not the anticipated 3'-O-[(p-toluoyloxy)ethyl]-dyphylline (6). Deacylation of 3a,b and 7 afforded 4a,b and 1, respectively. Viral screening of selected compounds against HBV has been investigated.  相似文献

Rapid colour change is a remarkable natural phenomenon that has evolved in several vertebrate and invertebrate lineages. The two principal explanations for the evolution of this adaptive strategy are (1) natural selection for crypsis (camouflage) against a range of different backgrounds and (2) selection for conspicuous social signals that maximise detectability to conspecifics, yet minimise exposure to predators because they are only briefly displayed. Here we show that evolutionary shifts in capacity for colour change in southern African dwarf chameleons (Bradypodion spp.) are associated with increasingly conspicuous signals used in male contests and courtship. To the chameleon visual system, species showing the most dramatic colour change display social signals that contrast most against the environmental background and amongst adjacent body regions. We found no evidence for the crypsis hypothesis, a finding reinforced by visual models of how both chameleons and their avian predators perceive chameleon colour variation. Instead, our results suggest that selection for conspicuous social signals drives the evolution of colour change in this system, supporting the view that transitory display traits should be under strong selection for signal detectability.  相似文献
