A recent study has linked the butyrylcholinesterase (BChE) K-variant and the apolipoprotein epsilon4 isoform to late-onset Alzheimer's disease. These findings have been controversial and have led us to examine the differences between wild-type and K-variant BChE in enzyme activity, protein stability, and quaternary structure. J-variant BChE (E497V/A539T) was also studied because it is associated with the K-variant mutation. The K-variant mutation (A539T) is located in the C-terminal tetramerization domain. Wild-type, K-variant, and J-variant BChE were expressed in Chinese hamster ovary cells and purified. The purified enzymes had similar binding affinity (Km) values and catalytic rates for butyrylthiocholine and benzoylcholine. In pulse-chase studies the K-variant, J-variant, and wildtype BChE were degraded rapidly within the cell, with a half-time of approximately 1.5 h. Less than 5% of the intracellular BChE was exported. The C-terminal peptide containing the K-variant mutation interacted with itself as strongly as did the wild-type peptide in the yeast two-hybrid system. Both K-variant and wild-type BChE assembled into tetramers in the presence of poly-L-proline or the proline-rich attachment domain of the collagen tail. The native K-variant BChE in serum showed the same proportion of tetramers as the native serum wild-type BChE. We conclude that the K-variant BChE is similar to wild-type BChE in enzyme activity, protein turnover, and tetramer formation.  相似文献

The chick stomach is composed of two parts: glandular or proventriculus and muscular or gizzard which are morphological and functionally different. Since characterization of the mucosubstances in the adult stomach and its comparison with the embryonary stomach have not been made, we performed the study of the cytochemical characteristics of mucosubstances in the chick glandular stomach during the embryonic and post-natal periods, to obtain information on changes produced in these components during functional differentiation. In this work we established that during development, the epithelial cells of the superficial layer content predominantly glycoproteins and the glycosaminoglycans of the glands decrease when they begin to secrete other compounds, such as proteolytic enzymes necessary for digestion. This sequence of mucosubstances appearance is concordant with the increase of carbonic anhydrase, which reaches its highest specific activity from 15 to 20 days of incubation. In this period hydrochloric acid secretion increases and therefore, glycoprotein secretion becomes necessary to protect the mucous membranes.  相似文献

Methylation of glucosamine-6-phosphate isomerase deaminase (2-amino-2-deoxy-D-glucose-6-phosphate ketol-isomerase, deaminating, or glucosamine-6-phosphate deaminase, EC 5.3.1.10), from Escherichia coli produces a modified protein having two alkylated sulfhydryls per each polypeptide chain. The enzyme is still active and allosteric, but exhibits a lower homotropic cooperativity and its Vmax/Etotal is almost exactly half that of the native enzyme. Arsenite produces comparable kinetic changes that can be reversed with ethanedithiol but not with 2-thioethanol or dialysis. Thiols can be oxidized by molecular oxygen using the (1,10-phenanthroline)3-Cu(II) complex as catalyst; the enzyme obtained no longer has titrable SH groups with 5,5'-dithiobis(2-nitrobenzoic acid) and displays kinetic behavior similar to that of the other chemically modified forms of the deaminase using monofunctional or bifunctional reagents. The results reported indicate that the involved sulfhydryls are vicinal groups, and are located in a region of the molecule that moves as a whole in the allosteric transition.  相似文献

Mutagenic treatment with N-methyl-N′-nitro-N-nitrosoguanidine (MNNG) inPhaffia rhodozyma generated 15 mutants with a wide diversity of color variants ranging from white to dark red. Characterization of the mutants by absorption spectra, TLC and HPLC was performed. Two categories could be distinguished: astaxanthin hyperproducing and astaxanthin hypoproducing mutants. Hyperproducing mutants exhibited considerable increases in astaxanthin content whereas hypoproducing mutants showed higher β-carotene contents than the wild-type strain. The characterization of carotenoid mutants inP. rhodozyma could contribute to the knowledge of the biosynthetic pathway of astaxanthin production of this microorganism.  相似文献

Glucosamine-6-phosphate deaminase (EC 5.3.1.10) from dog kidney cortex was purified to homogeneity, as judged by several criteria of purity. The purification procedure was based on two biospecific affinity chromatography steps, one of them using N-epsilon-amino-n-hexanoyl-D-glucosamine-6-phosphate agarose, an immobilized analog of the allosteric ligand, and the other by binding the enzyme to phosphocellulose followed by substrate elution, which behaved as an active-site affinity chromatography. The enzyme is an hexameric protein of about 180 kDa composed of subunits of 30.4 kDa; its isoelectric point was 5.7. The sedimentation coefficient was 8.3S, and its frictional ratio was 1.28, indicating that dog deaminase is a globular protein. The enzyme displays positive homotropic cooperativity toward D-glucosamine-6-phosphate (Hill coefficient = 2.1, pH 8.8). Cooperativity was completely abolished by saturating concentrations of GlcNAc6P; this allosteric modulator activated the reaction with a typical K-effect. Under hyperbolic kinetics, a Km value of 0.25 +/- 0.02 mM for D-glucosamine-6-phosphate was obtained. Assuming six catalytic sites per molecule, kcat is 42 s-1. Substrate-velocity data were fitted to the Monod's allosteric model for the exclusive-binding case for both substrate and activator, with two interacting substrate sites. The Kdis for N-acetyl-D-glucosamine-6-phosphate was estimated at 14 microM.  相似文献

  1. Semi-arid and arid landscapes (dry-lands) cover 41% of the Earth’s land surface over five continents. These areas are home to 55% of mammal species. Bats have the second highest species richness among mammals, and, although many species are adapted to arid conditions, they are particularly sensitive in these habitats and require conservation priority.
  2. Information on bats in arid and semi-arid landscapes is scattered, patchy and focused on small-scale studies; therefore, we undertook a systematic review using the PRISMA protocol to identify the current knowledge status, detect knowledge gaps and propose future research priorities.
  3. We analysed 346 published articles and evaluated 40 topics within five topic categories (taxa studied, methodologies used, biology, ecology and conservation). The most commonly studied topic categories were ecology and biology. However, we found a gap in the topic category conservation (including topics such as conservation status and roost conservation). Our network analysis of topics within the categories showed that most ecology papers were focused on distribution, species richness and habitat use.
  4. When we analysed keywords, we found that phylogeny, taxonomy and distribution demonstrated relatively high presence. Moreover, comparison of the percentage of studies conducted in dry-lands and the percentage of land surface area covered by dry-lands in the continents revealed that dry-lands in Africa and Australia were especially under-represented. Our review shows that knowledge of bats in semi-arid and arid landscapes is biased towards new records of the distribution of species, as well as covering systematic/taxonomic and morphological aspects of bat biology.
  5. We suggest that research on conservation measures and guidelines to protect the bat species found in semi-arid and arid landscapes should be prioritised, together with the sharing of knowledge with local practitioners and the development of citizen science programmes.
